    The pathways connecting the hippocampal formation, the thalamic reuniens nucleus and the thalamic reticular nucleus in the rat

    Most dorsal thalamic nuclei send axons to specific areas of the neocortex and to specific sectors of the thalamic reticular nucleus; the neocortex then sends reciprocal connections back to the same thalamic nucleus, directly as well indirectly through a relay in the thalamic reticular nucleus. This can be regarded as a 'canonical' circuit of the sensory thalamus. For the pathways that link the thalamus and the hippocampal formation, only a few comparable connections have been described. The reuniens nucleus of the thalamus sends some of its major cortical efferents to the hippocampal formation. The present study shows that cells of the hippocampal formation as well as cells in the reuniens nucleus are retrogradely labelled following injections of horseradish peroxidase or fluoro-gold into the rostral part of the thalamic reticular nucleus in the rat. Within the hippocampal formation, labelled neurons were localized in the subiculum, predominantly on the ipsilateral side, with fewer neurons labelled contralaterally. Labelled neurons were seen in the hippocampal formation and nucleus reuniens only after injections made in the rostral thalamic reticular nucleus (1.6-1.8 mm caudal to bregma). In addition, the present study confirmed the presence of afferent connections to the rostral thalamic reticular nucleus from cortical (cingulate, orbital and infralimbic, retrosplenial and frontal), midline thalamic (paraventricular, anteromedial, centromedial and mediodorsal thalamic nuclei) and brainstem structures (substantia nigra pars reticularis, ventral tegmental area, periaqueductal grey, superior vestibular and pontine reticular nuclei). These results demonstrate a potential for the thalamo-hippocampal circuitry to influence the functional roles of the thalamic reticular nucleus, and show that thalamo-hippocampal connections resemble the circuitry that links the sensory thalamus and neocortex.

    6-Hidroksidopamin Uygulanan Wistar Ve Genetik Absans Epilepsili SıçanlarınRotasyonlarının Karşılaştırılması

    GİRİŞ VE AMAÇ: 6-Hidroksidopamin (6-OHDA); nigro-striatal dopaminerjik nöronlar için toksik bir ajandır ve Parkinson hastalığının modellenmesinde kullanılır (1). Çalışmamızda, Strasbourg orijinli genetik absans epilepsili sıçanlarda (GAERS) ve Wistar sıçanlarda 6-OHDA toksisitesinin dopaminerjik hasar oluşturmasının göstergelerinden biri olan ve subkutan apomorfinle indüklenen rotasyon davranışını, 2 grup arasında kıyaslamak hedeflendi. YÖNTEM:Bu çalışmada, 30 günlük Wistar ve GAERS hayvanlar kendi içlerinde iki gruba ayrıldı. 6-OHDA (8 µg doz ve 4 µL/4dk hızında); GAERS-MFB (n=8) grubuna medial ön beyin demetine (MFB) (AP:-1,4; ML:1,6; V:7,1 mm) tek enjeksiyon, GAERS-Striatum (n=5) grubuna striatuma iki enjeksiyon (AP:-0,5/+0,5; ML:3,0; V:5,0 mm), Wistar-MFB (n=4) grubuna (AP:-1,4; ML:1,6; V:7,1 mm) tek enjeksiyon, Wistar-Striatum (n=2) grubuna da striatuma iki enjeksiyon (AP:-0,5/+0,5; ML:3,0; V:5,0 mm) olacak şekilde stereotaksik cerrahi yöntemle uygulandı.6-OHDA uygulamasından 21 gün sonra tüm hayvanlara apomorfin enjeksiyonu (0,05 mg/kg, subkutan) uygulandı. Apomorfin uygulanmasından sonra 30 dakika boyunca hayvanların 3600 sağa ve sola rotasyonları kaydedildi. Veriler ortalama±standart hata olarak ifade edildi. Tek yönlü ANOVA ve Tukey’in post-hoc testi kullanıldı (p<0,05 anlamlı olarak kabul edildi). BULGULAR:GAERS-MFB rotasyon sayısı dakikada ortalama 6,56±0,05 iken, GAERS-Striatum dakikada ortalama 6,19±1,72 oldu. Wistar-MFB rotasyon sayısı dakikada ortalama 4,74±2,33 iken, Wistar-Striatum rotasyon sayısı dakikada ortalama 3,27±1,40 olarak bulundu. Gruplar arasında istatistiksel olarak anlamlı bir fark bulunmadı. TARTIŞMA VE SONUÇ:Bulgular; gruplar arasında bir fark olmadığını göstermekle beraber, çalışmamızda grup sayılarının sınırlı olması nedeniyle, sayı arttırılarak çalışmanın devamı hedeflenmektedir.     The relationship between UGT1A4 polymorphism and serum concentration of lamotrigine in patients with epilepsy

    Lamotrigine (LTG) which has a widespread use in epilepsy treatment as an antiepileptic agent is metabolized by UDP-glucuronosyl transferase (UGT) enzymes. In this study, single nucleotide polymorphisms, P24T and L48V, of the UGT1A4 enzyme have been investigated in a Turkish population of patients with epilepsy (n=131) by comparing serum levels of LTG of wild type and polymorphic subjects. High performance liquid chromatography (HPLC) was used to measure serum concentrations of LTG. The P24T and L48V polymorphisms of the UGT1A4 enzyme were analyzed with a matrix assisted laser desorption-time of flight (MALDI-TOF) mass spectrometry method. The frequencies of the heterozygous alleles for L48V or P24T polymorphisms were 22.4% and 3.8%, respectively. L48V polymorphism was found to decrease the serum concentration of LTG in patients on monotherapy or polytherapy. The LTG levels of non smoking monotherapy patients were 52% lower for the L48V polymorphism than for wild type alleles. Also the LTG levels were significantly lower for non smoking or smoking polymorphic alleles than for normal. The high frequency of the L48V polymorphism detected in the Turkish population indicates that LTG dose adjustments in patients with the UGT1A4 L48V polymorphic enzyme should be taken into account.     Lakozamid ile birlikte karbamazepin, fenobarbital veya valproik asit kullanımının hepatik metabolizma üzerindeki in vitro etkileri

    Amaç: Karbamazepin ve fenobarbital hepatik sitokromP450 (CYP) metabolizma enzimlerini indükleyerek, valproik asit ise inhibe ederek ilaçilaç etkileşimlerine yol açabilmektedir. Yeni jenerasyon anti-nöbet ilaçlardan (ASD) biri olan lakozamid (LCM), CYP2C19, CYP2C9 ve CYP3A4 enzimleri ile metabolizma olmaktadır. Çalışmamızda lakozamid ile birlikte kullanılan ASD’lerin CYP enzimleri üzerindeki etkilerini in vitro yöntemlerle mRNA düzeyinde araştırmayı amaçladık. Gereç ve Yöntem: İnsan hepatoselüler karsinom hücrelerine (HepaRG veya HepG2) terapötik konsantrasyonlarda karbamazepin, fenobarbital veya valproik asit uygulanmasının ardından, kuyucuklara kanda minimum veya maksimum terapötik konsantrasyonlara (Cmin veya Cmaks) karşılık gelecek şekilde LCM eklendi. Hücre örneklerinde CYP2C19, CYP2C9 ve CYP3A4 genlerinin mRNA ekspresyon düzeyleri gerçek zamanlı polimeraz zincir reaksiyonu ile ölçüldü ve değişiklikler kat değişimi (FC) değerleri olarak hesaplandı. FC değerleri 0,5’in altında kalan gen ekspresyonları inhibisyon, 2’nin üzerinde olanlar ise indüksiyon olarak değerlendirildi ve GraphPad Prism 8 ile analiz edildi. Bulgular: HepaRG hücrelerinde en yüksek mRNA ekspresyonu CYP2C19 ve CYP3A4 genlerinde karbamazepin ve LCMCmin'nin birlikte uygulandığı gruplarda (sırasıyla, FC:5,2; FC:1009,9) saptanırken, CYP2C9 gen ekspresyonu, fenobarbital ile LCMCmin uygulanan grupta anlamlı derecede yüksek bulundu (FC:795,16). HepaRG hücrelerinde valproik asit ile LCMCmaks uygulanan grupta en düşük CYP gen ekspresyonu CYP3A4 olarak tespit edildi (FC:2,3). HepG2 hücrelerinde ise CYP genlerinin ekspresyonları anlamlı değişiklik göstermedi. Sonuç: CYP enzim ekspresyonunun daha yüksek oranda gerçekleştiği HepaRG hücre hattı deneylerinden elde ettiğimiz sonuçlar özellikle karbamazepin ve fenobarbitalin CYP genlerinin ekspresyonlarını değiştirerek (sırasıyla CYP2C19-CYP3A4 ve CYP2C9) LCM ile etkileşmeye girebileceğine işaret etmektedir. Sonuçlarımızın doğrulanması için LCM konsantrasyonunun eş zamanlı olarak ölçülmesi ve deney sayılarının arttırılması planlanmaktadır.     Electron microscopic GABA evaluation in hippocampal mossy terminals of genetic absence epilepsy rats receiving kindling stimulations

    Objective: The hypotheses related to the fact of epileptic mechanisms are mainly based on excitation-inhibition imbalance in central nervous system. GAERS (Genetic Absence Epilepsy Rats from Strasbourg) is a well-known animal model of absence epilepsy, and frequently used in experimental studies. In the present study, we aimed to examine possible morphological and gamma-aminobutyric acid (GABA) density changes in GAERS hippocampus after electrical kindling stimulations. Methods: All control and test group rats received 6 kindling stimulations. Rats were decapitated 1 h after the last stimulation. Ultrastructural GABA immunocytochemistry was used to evaluate GABA density quantitatively in mossy terminals of hippocampal CA3 region. Results: GABA levels were less in kindling groups compared to their controls, and in GAERS groups compared to Wistar groups; mitochondrial and dendritic spine area ratios were greater in GAERS groups compared to Wistar groups, although all these evaluations were statistically nonsignificant. Depletion of synaptic vesicles was evident in the mossy terminals of kindling groups. Conclusion: The reason of decreased levels of GABA found in the present study might be that GABA has been released from the synaptic pool rapidly at an early time period after the last stimulation, for compansation mechanisms. Depletion of synaptic vesicles observed in kindling groups shows that even 6 kindling stimulations have an impact of changing hippocampal morphology in trisynaptic cycle. The increased mitochondrial area in GAERS might be related to the increased mitochondrial activity. The increased dendritic spine area might be related to the increased performance of learning in GAERS. Our findings indicating that absence epilepsy and temporal lobe epilepsy have different mechanisms of epileptogenesis might be a basis for further experimental studies.
