Abstract
Amaç: Sambucus nigra L. meyvesinin antioksidan bileşenlerinin tayin etmek ve Sambucus nigra L. meyvesinin bazı hücre hatları üzerindeki sitotoksik etkisini belirlemektir.Gereç ve Yöntem : Sambucus nigra L. meyveleri taze olarak temin edildi. Ekstraksiyon aşamasında etanol (E1), %75 etanol (E2) ve su (E3) ekstreleri elde edildi. Ekstrelerin toplam fenolik bileşen içeriği Folin-Ciocalteu'nun fenol reaktifi kullanılarak belirlendi. Antosiyanin profili HPLC yöntemi ile belirlendi. Ekstrelerin antioksidan aktivitesini belirlemek için DPPH yöntemi ile serbest radikal giderme kapasitesi ölçümü, Demir (III) iyonu indirgeme antioksidan gücü tayini (FRAP), Cu (II) iyonu indirgeme antioksidan gücü tayini (CUPRAC) yöntemleri kullanıldı. Ekstrelerin HCT116 insan kolon kanseri hücre hattı, PC-3 prostat kanseri hücre hattı ve HacaT insan keratinosit hücre hattı üzerindeki sitotoksik etkisinin belirlenmesi için MTT (3- (4,5-Dimetiltiazol-2-il)-2,5-difeniltetrazolyum bromür) analizi gerçekleştirildi. Sonuçlar SPSS- 22 istatistik programı ile değerlendirildi.Bulgular Sambucus nigra L. meyvelerinin yüksek antioksidan içeriğe sahip olduğu saptandı. Ekstre eldesinde kullanılan çözücünün elde edilen ekstrenin antioksidan özelliklerini etkilediği ve E2 ekstresinin daha yüksek antioksidan özelliğe sahip olduğu tespit edildi (p<0,05). Sambucus nigra L. meyve ekstrelerinin HCT-116 ve HacaT hücre hatları üzerinde sitotoksik etkisi bulunmakla birlikte hücre canlılığında %50 azalmaya neden olacak doz tespit edilmemiştir.Sonuçlar Sambucus nigra L. meyveleri yüksek antioksidan özelliğe sahiptir. Ekstre hazırlanırken kullanılan çözücü ekstrenin biyoaktif bileşen miktarını ve hücre hatları üzerindeki sitotoksik etkisini etkileyebilmektedir.
Objective: To determine the antioxidant components of Sambucus nigra L. fruit and the cytotoxic effect of this fruit on some cell lines.Material and Methods: Sambucus nigra L. fruits were obtained fresh. In the extraction process, ethanol (E1), 75% ethanol (E2) and water (E3) extracts were obtained. The total phenolic component content of the extracts was determined using Folin-Ciocalteu's phenol reagent. Anthocyanin profile was determined by HPLC method. In order to determine the antioxidant activity of the extracts, DPPH (1,1-diphenyl-2-picryl hydrazyl) Radical Scavenging Activity, Ferric Reducing Antioxidant Power Assay (FRAP), and Cupric Ion Reducing Antioxidant Capacity Assay (CUPRAC) methods were used. MTT (3-(4,5-Dimethylthiazol-2- yl)-2,5-diphenyltetrazolium bromide) analysis was performed to determine the cytotoxic effect of the extracts on HCT116 human colon cancer cell line, PC-3 prostate cancer cell line, and HacaT human keratinocyte cell line. All results were evaluated with SPSS-22 statistical program.Results: It was determined that Sambucus nigra L. fruits have high antioxidant content. It was determined that the solvent used in the extraction affected the antioxidant properties of the obtained extract and the E2 extract had higher antioxidant properties (p<0.05). Although Sambucus nigra L. fruit extracts have a cytotoxic effect on HCT-116 and HacaT cell lines, the dose that will cause a 50% decrease in cell viability has not been determined.Conclusion: Sambucus nigra L. fruits have high antioxidant properties. The solvent used in the preparation of the extract may affect the amount of bioactive component and its cytotoxic effect on cell lines.
