Due to limited information available, the objective of this study was to develop a quantification method for the three impurities of finasteride using high performance liquid chromatography (HPLC) with an ultraviolet (UV) detector. The compounds (impurities) of finasteride that are registered with the European Pharmacopeia, which we sought to validate are: -N-(1,1-dimethylethyl)-3-oxo-4-aza-5 alpha-androstane-17 beta-carboxa mide (impurity A), methyl 3-oxo-4-aza-5 alpha-androst-1-ene-17 beta-carboxylate (impurity B), and -N-(1,1-dimehylethyl)-3-oxo-4-azaandrosta-1,5-diene-17 beta-carboxamide (impurity C). Analyses were performed using a Nova Pac C-18 column for HPLC with isocratic elution. Detection was carried out at 210 nm, the concentration of the three impurities was in the range was 1.5-4.5 mu g mL(-1) at ambient temperature with a mobile phase of water + acetonitrile + tetrahydrofuran (80:10:10, v/v/v) and the flow rate was 2.0 mL min(-1). The recoveries were: 101.35 +/- 0.62% (impurity A), 101.60 +/- 2.66% (impurity B) and 101.97 +/- 2.05% (impurity C). Validation of the method yielded fairly good results as it relates to the precision and accuracy. It is, therefore, concluded that the method would be suitable for not only the separation and determination of processed impurities to monitor the reactions, but also for the quality assurance of finasteride and its related substances. Whole blood samples were extracted with a WAX cartridge on auto-SPE. Stability test of the drug substances performed at the same time in working solution and matrix at +4 degrees C, -20 degrees C, +20 degrees C in dark and +20 degrees C in light conditions for 1, 2, 3 and 4 weeks. For short-time stability, the effects of 6 h storage at +55 degrees C was also investigated. Repeated data were statistically analyzed with ANOVA. All substances in working solution were stable at -20 degrees C and +4 degrees C. In the blood matrix, higher temperatures (+20 degrees C) in light and dark caused degradation of substances at the end of four weeks and short-term study (P<0.05). Anksiyete bozuklukları toplumda en yaygın görülen ruhsal bozukluklardır. 1990’lardan bu yana anksiyete bozuklukları üzerinde daha fazla durulmaktadır. Dünyada çeşitli ülkelerde yapılan çalışmalarda yaşam boyu yaygınlığa ilişkin benzer oranlar saptanmıştır. Tedavi genellikle farmakoterapi ve / veya psikoterapiden oluşur. Antidepresan ajanlar, seçici serotonin geri alım inhibitörleri (SSRI'lar) gibi eski trisiklik antidepresanlardan (TCA'lar) daha güvenli bir yan etki profiline ve daha yüksek kullanım kolaylığına sahip daha yeni ajanlar olup anksiyete bozukluklarının tedavisinde tercih edilen ilaçlardır. Çalışmanın amacı; anksiyete tedavisinde en çok kullanılan SSRI ve SNRI grubu etken maddelerin tayini için son yıllarda yapılmış analitik çalışmaları araştırmak ve sunmaktır. SSRI ve SNRI grubu etken maddelerin analizleri HPLC-ESI-MS, MAE/UPLC-TOF-MS, FPSE/HPLC-DAD, UPLC-MS-MS, HPTLC, SPE/HPLC, LC-MS-MS gibi yöntemlerle incelenmiştir. Tocopherols and tocotrienols are collectively named 'tocols'. Tocols are well-known for their potent biological activities, such as antioxidant, anticancer, anti-aging, anti-inflammatory activities, and they prevent vascular occlusion by providing fluidity in the veins. They are one of the main sources of human nutrition. Therefore, it is vital to analyze various forms of tocols in plants, foods, and other commercial products in addition to monitoring it in body fluids. Capparis Ovata is a plant from the family Capparaceae grown widely in Turkey and has a commercial value. We investigated eight tocols (alpha, beta, delta, gamma tocopherols and alpha, beta, delta, gamma tocotrienols) in C. Ovata seed; optimized a pressurized liquid extraction (PLE) method for isolation, and developed fully validated RP-HPLC-DAD method. The delta-tocopherol, gamma-tocopherol and alpha-tocopherol quantities were 1505.61 ppm, 930.24 ppm and 47.10 ppm with methanolic PLE method, respectively. All quantities detected with Soxhlet extraction were less than those obtained with PLE. In the present study, delta-tocotrienol was 32 times, gamma-tocopherol was 20 times more than alpha-tocopherol. In consideration of recently reported pharmacological activities of delta- and gamma-tocopherols, and the higher effectiveness of delta-tocopherol, our results are of particular importance. We detected and separated all tocols in about 10 min, and beta- and gamma-isomers of tocopherols had the comparable resolution value of 1.52. The limits of quantification values were low with the range of 0.29 x 10(-4) to 9.60 x 10(-4) mg mL(-1); the recoveries were between 95.84 and 113.83% for all eight tocols, and due to the wide linear quantification ranges, the developed method was stated to be suitable for various amounts of tocol analysis.Publication Open Access Quantitative Determination of The Heavy Metals (Lead, Zinc And Manganese) in White Cheese Produced in Ergene Basin by Using Atomic Absorption Spectrophotometry(MARMARA UNIV, FAC PHARMACY, 2015-04-09) CÜCÜ, AYŞEN; Cucu, Aysen Kurt; Yavuz, Mustafa; Demir, Hulya DemircanIt is well known that heavy metals are harmful for human health appearing only after exceeding the specific concentrations of each metal. This study was carried out to determine the concentrations of heavy metals in cheese that was produced in diaries of Ergene Basin. The dairies near the Ergene River were investigated and suitable ones were selected. Since the sampling process directly affects the sensitivity of the results, particular attention was given to this process. A total of twelve samples of cheese were collected from different diaries and each samples were digested three times by dry ashing and wet ashing methods and totally 108 cheese samples were obtained for metal analyses. The lead, manganese content of the cheese samples were measured by graphite furnace atomic absorption spectrophotometer and zinc content of the cheese samples were measured by flame atomic absorption spectrophotometer. Contents of heavy metals in the examined samples were compared with the excepted limit values in the literature and the results were evaluated.
