    Clinical evaluation of dental enamel defects and oral findings in coeliac children

    Purpose To examine dental hard and soft tissue changes of coeliac children in order to increase the awareness of the pediatric dentists in prediagnosis of especially undiagnosed coeliac disease. Materials and methods Sixty children, 28 (46.7%) boys and 32 (53.3%) girls whose ages were between 6 to 16 years were included in the present study. Thirty children who had undergone endoscopy and diagnosed with the coeliac disease in the Sisli Hamidiye Etfal Hospital, Istanbul, Turkey, formed the study group. Also, thirty children clinically suspected of having the coeliac disease with the same gastrointestinal complaints had undergone endoscopy and proven not coeliac were chosen as the control group. Oral examination involved assessment of dentition and specific and unspecific dental enamel defects. Also, soft tissue lesions, clinical delay of the dental eruption, salivary flow rate, pH, and buffering capacity were examined. Results Twenty coeliac patients had enamel defects, however none in the control subjects. In the coeliac group, all enamel defects were diagnosed in permanent teeth and as specific in all children. Grade I dental enamel defects found mainly in the incisors. The clinical delayed eruption was observed in 10 (33.3%) of 30 coeliac children and none of the children in the control group. While the level of DMFT/S numbers and stimulated salivary flow rate were found significantly lower in the coeliac group, pH was found significantly higher. Conclusion Oral cavity may be involved in coeliac disease and pediatric dentists can play an important role in the early diagnosis of the coeliac disease.

    Effects of Exercise and Calorie Restriction on Brain_x000D_ and Testis in Natural Aging Model

    The aim of our study was to examine the effects of exercise and calorie_x000D_ restriction on various tissue damage and antioxidant parameters_x000D_ in the brain and testis of rats in a natural aging model. For this_x000D_ purpose, male Sprague-Dawley rats were the control group (C),_x000D_ the elderly (A), the elderly with calorie restriction (ACR), the elderly_x000D_ who were exercised (AE) and the elderly who were exercised with_x000D_ calorie restriction (ACRE), they were divided into 5 groups. The control_x000D_ group was composed of three-month-old animals. The other_x000D_ group consisted of 15-month-old rats. Exercise and calorie restriction_x000D_ were applied for 6 weeks. At the end of the experiment, lipid_x000D_ peroxidation (LPO), nitric oxide (NO), glutathione (GSH) levels and_x000D_ superoxide dismutase (SOD), catalase (CAT), glutathione-S-transferase_x000D_ (GST) and tissue factor (TF) were determined in brain and_x000D_ testicular tissues homogenates. As a result of the study, the A_x000D_ group’s brain and testis LPO, NO levels and TF activity increased,_x000D_ GSH levels and SOD, CAT and GST activities decreased, when compared_x000D_ to the C group. As a result of our study, an increase in oxidant_x000D_ damage was observed with TF activity in the brain and testis in the_x000D_ natural aging model, and positive effects of exercise and calorie_x000D_ restriction on the antioxidant levels in the brain were determined,_x000D_ especially in aging.

    Effect of Sample Storage on Stability of Salivary Glutathione, Lipid Peroxidation Levels, and Tissue Factor Activity

    Saliva samples are often required to be stored for longer periods of time either because of the project protocol or because of lack of funding for analysis. The effects of 6 months storage (fresh, 30, 60, 90 120, 150, and 180d) on the stability of salivary reduced glutathione (GSH), lipid peroxidation (LPO) and 90days of storage (fresh, 15, 30, 60, and 90d) on the stability of salivary tissue factor (TF) activity and the stability of saliva imprint samples at 20 C were evaluated in this study. Salivary GSH, malondialdehyde (MDA) levels as an index of LPO, and TF activities were determined using the methods of Beutler, Yagi, and Quick, respectively. Saliva imprint samples were stained with Giemsa and microscopically examined. Salivary GSH levels and TF MDA levels increased significantly after 6 months of storage at -20 C. Leucocyte, epithelium and bacterium cell counts did not significantly change at the end of 90 d of storage. Saliva samples may be stored up to 1 month at -20 C for LPO assay. For cytological examinations, saliva samples may be stored for 90 d at -20 C. Further studies are needed to determine the stability of salivary GSH, and salivary TF activity stored less than 30 days at -20 C. On the other hand, if saliva samples are required to be stored, to avoid the changes because of different storage periods, we recommend that they must be stored under the same circumstances and in the same time period.     Tooth replantation with adipose tissue stem cells and fibrin sealant: microscopic analysis of rat's teeth

    Treatment for dental avulsion cases is early or late replantation of the traumatized teeth. Prognosis of the replanted tooth depends on the level of periodontal injury. Adipose tissue stem cells (ATSCs) were reported to improve periodontal ligament tissue (PDL) regeneration. Fibrin sealant (FS) contains thrombin and fibrinogen to form an adhesive fibrin clot routinely used in surgical procedures. Here, we aimed to investigate the effects of ATSCs + FS treatment on healing of PDL after tooth replantation in a rat model. After 60 min of extraction, maxillary central incisor teeth were replanted with ATSCs + FS. Two months later, the rats were sacrificed and hemimaxilla blocks were dissected out for histological analysis. The results showed that there was a significant improvement in histological findings of ATSCs + FS treated group compared to only FS treated and non-treated groups corresponding to reduced inflammatory resorption and increased new PDL formation. Furthermore, the ankylosis levels were lowered after ATSCs + FS treatment. Singular use of FS improved PDL healing moderately. Our results indicated that ATSCs + FS treatment improves PDL healing after tooth replantation suggesting a new therapeutic potential in the treatment of dental avulsion cases.

    Investigation of the Effects of Edaravone on Valproic Acid Induced Tissue Damage in Pancreas

    Valproic acid (VPA), an effective antiepileptic and anticonvulsant drug, has some toxic side effects due to causing elevated oxidant production. The aim of this study is to investigate the effects of edaravone, a potent free radical scavenger on VPA induced toxicity and tissue damage by biochemical and histological examinations on pancreas. Female Sprague Dawley rats were divided into four groups as follows; control, edaravone, VPA, VPA+edaravon. VPA and edaravone were injected intraperitonally for seven days. Total protein, lipid peroxidation (LPO), sialic acid (SA) and glutathione (GSH) levels and alkaline phosphatase (ALP), tissue factor (TF), superoxide dismutase (SOD), glutathione-S-transferase GST), catalase (CAT), glutathione peroxidase (GPx) and myeloperoxidase (MPO) activities were determined in pancreas homogenates. In VPA given group, LPO and SA levels, and ALP, TF, MPO activities significantly increased and GST, CAT, GPx activities significantly decreased compared to control group. A marked morphological damage was detected in the VPA group. Ameliorative effects of edaravone were observed in SA, TF, CAT, GPx parameters and histological examination in the VPA group. Therefore, edaravone may be effective in moderation and/or reduction of toxic effects of VPA on pancreas.

    Effect of non-surgical periodontal therapy on salivary melatonin levels

    Objective: Melatonin, a hormone secreted predominantly by pineal gland in a circadian manner, has antioxidant and anti-inflammatory effects. The current research is conducted to explore the influence of non-surgical periodontal therapy (NSPT) on levels of salivary melatonin in subjects with gingivitis and periodontitis. Methods: Sixty systemically healthy participants were included in this study; the groups are as follows: gingivitis (G), chronic periodontitis (CP), generalized aggressive periodontitis (GAP) and periodontally healthy (H). NSPT was applied to G group patients for 2 sessions, to CP and GAP group patients for 4 sessions. Plaque and gingival indices, probing depth (PD), bleeding on probing (BOP), and clinical attachment level (CAL) were documented at baseline and 3 months post – treatment and early morning salivary samples were collected. ELISA was used to detect melatonin levels in saliva. Pittsburgh Sleep Quality Index (PSQI) questionnaire was performed to evaluate of sleep quality of patients. Results: At baseline, significant difference in gingival index, PD, BOP and CAL values was detected among all groups (p<0.05). Following NSPT, clinical measurements improved in G, CP, and GAP groups significantly (p<0.05). While salivary melatonin concentration of all groups was similar at baseline (p>0.05), a significant elevation in the level of salivary melatonin was found only in the G group after NSPT (p<0.05). PSQI scores differed significantly among groups (p<0.05). Conclusion: The salivary melatonin levels in the presence of gingivitis and periodontitis varied at baseline and elevated following NSPT parallel to the improvement in clinical parameters.

    Effect of Deuterium Depletion on Erythrocytes in_x000D_ Experimental Diabetes

    In the development of diabetic complications, oxidative stresscaused by hyperglycemia plays an important role. Deuterium depletionhas protective effect against cellular damage and tumorsuppressor effect in cancer cell lines. The purpose of this studywas to examine and evaluate the effect of deuterium depletion onerythrocyte oxidant and antioxidant system parameters in experimentaldiabetes induced by streptozotocin (STZ) in rats. On the30th or 40th day after STZ injection, blood samples were taken andblood glucose levels were measured. Erythrocyte hemolysateswere also prepared. Lipid peroxidation and glutathione levels andglutathione-S-transferase, superoxide dismutase and catalase enzymeactivities of erythrocytes were determined. At the 30 or 40day periods, deuterium depleted water (DDW) consumption haddifferently affected oxidant-antioxidant parameters of erythrocytes.The clear effect of deuterium depletion in drinking waterfrom 150 ppm to 85 ppm was observed in glutathione levels andit increased erythrocyte glutathione levels in both the control anddiabetic groups. In conclusion, deuterium depletion may have protectiveeffects on diabetes induced oxidative stress. Further studiesare needed to be done to determine the mechanism of actionand the useful dosage and the application period of DDW.

    Chard extract increased gastric sialic acid and ameliorated oxidative stress in valproic acid-administered rats

    Valproic acid, one of the most used drugs for epilepsy patients, has some known side effects. Chard extract has many pharmacological activities. The study aims to evaluate whether valproic acid might interfere with oxidative metabolism in gastric tissue and whether chard ameliorates these effects. The Sprague Dawley rats were divided into four groups (n=8); control, chard-given control, valproic acid, and chard-given valproic acid. The aqueous extracts of chard leaves were given 1 h before the administration of valproic acid for 7 days. On the 8th day, the animals were sacrificed under anesthesia and gastric tissues were homogenized. When compared to the control group, valproic acid significantly increased malondialdehyde and catalase activity, while superoxide dis- mutase activity decreased. Chard administration increased glutathione and, sialic acid levels and decreased malondialdehyde levels and superoxide dismutase activity in the valproic acid group. Based on these findings, since chard increased gastric sialic acid levels, we may suggest that chard may protect gastric mucosa may be through its antioxidant effects.
