Publication Metadata only Taze sıkılmış ve ti̇cari̇ olarak satılan paketlenmi̇ş portakal suyunda metanol mi̇ktarının farklı saklama koşullarına göre İncelenmesi̇(2022-03-31) YARAT, AYŞEN; Yarat A., Çalşimşek Ö.Metanol alkollerin en basit üyesidir. Ağızdan, soluma yoluyla veya deri yoluyla çok küçük miktarlarda alınsa dahi ciddi zehirlenmelere neden olabilir. Bu çalışmada, taze sıkılmış ve ticari portakal suyunda metanol konsantrasyonları gaz kromatografi yöntemi ile belirlenmiş ve farklı saklama koşullarına bağlı olarak alkol oluşumu izlenmiştir. 0, 1, 7, 14, 30 ve 90. günlerde oda sıcaklığında, buzdolabında ve derin dondurucuda saklanan taze sıkılmış portakal suyundaki metanol değerleri 4-12 mg/dL civarındaydı. Ancak ticari portakal suyunda metanol her koşulda tespit edilememiştir. Bunun nedeni, ticari portakal suyunda bulunan alkol fermantasyonunu önleyen katkı maddeleri olabilir. Bu çalışmada taze portakal suyunda tespit edilen metanol değerleri, kandaki minimum öldürücü metanol konsantrasyonunun (40 mg/dL) altındaydı. Toksik limit göz önüne alındığında, arka arkaya çok miktarda taze portakal suyu tüketmek uygun olmayabilir. Depolama koşullarına göre ticari portakal suyunda metanol oluşmaması elbette iyidir. (M. oleifera) extracts exhibit multiple nutraceutical or pharmacological functions including anti-inflammatory, anti-oxidant, anti-cancer, hepatoprotective, neuroprotective, hypoglycemic, and blood lipid-reducing functions. Valproic acid (VPA) is a broad spectrum antiepileptic drug that is effective in migraine, bipolar disorder, anxiety and other psychiatric disorders. In addition to many known clinical side effects of VPA, it causes sialadenosis and dry mouth, and it is also claimed that VPA treatment prevents the growth of salivary gland tumors. In this study, the possible protective effect of M. oleifera alcoholic extract on the submandibular gland which is one of the major salivary glands, was investigated in VPA-treated rats. Submandibular gland lipid peroxidation values were significantly increased in the VPA group compared to the control group. Lipid peroxidation values were significantly decreased with the administration of M. oleifera extract. Kanser tedavisi için saf mAb’lara ihtiyaç vardır. Amaç: Bu çalışmanın amacı, mAb’ların saflaştırmasını Protein A kromatografisi, katyon değişim/anyon değişim kromatografisi veya çoklu mod kromatografi ile gerçekleştirmektir. Yöntem: Protein A rezini ile olan saflaştırma, mAb’ın Fc bölgesi ile kolon matriksine immobilize edilmiş Protein A ligandları arasındaki spesifik bağlanma ile gerçekleştirildi. Çoklu mod kromatografi rezinleri kullanılarak yapılan saflaştırmada ise iyonik ve hidrofobik etkileşimleri aynı anda gerçekleştirebilecek gruplara sahip olması nedeniyle saflaştırma rezini ile mAb arasındaki çoklu bağlanma etkileşimleri ile gerçekleştirildi. Bulgular: Protein A saflaştırma basamağı ile elde edilen mAb ürününün farklı bir saflaştırma basamağı ile de elde edilebileceği gösterildi. Her iki saflaştırma basamağı sonrası elde edilen ürünlerin HPLC yöntemleri ile kalite profillerinin birbirine benzer olduğu saptandı. Sonuç: İki farklı saflaştırma yöntemi ile benzer mAb ürününün elde edilebileceği gösterildi. Protein A rezini ile gerçekleştirilen saflaştırma basamağının %Verim değeri diğer saflaştırma basamağına göre bir miktar daha fazlaydı. Bunun sebebi, Protein A rezininin IgG’nin Fc bölgesine spesifik bağlanma özelliği göstermesinden dolayı idi. Drugs for the treatment of various diseases can adversely affect the structure and functions of the skin. Valproic acid/valproate (VPA) or 2-n-propylpentanoic acid is one of these drugs. Besides being used in the treatment of epilepsy and seizures, VPA is also effective in migraine, bipolar disorder, anxiety and other psychiatric disorders. Among the dermatological side effects of VPA, there are different effects such as hirsutism, hair loss, change in hair color, change in hair growth, and rarely skin rashes. Extracts from Moringa oleifera (M. oleifera) exhibit multiple nutraceutical or pharmacological functions, including anti-inflammatory, anti-oxidant, anticancer, hepatoprotective and neuroprotective functions. The oil obtained from the seed of this plant has cosmetic value and is known to be used as a moisturizer and skin care product in body and hair care. In this study, the possible protective effect of M.oleifera alcoholic extract on the skin in VPA-treated rats was investigated. Skin lipid peroxidation values increased significantly in the VPA group compared to the control group. Lipid peroxidation values were significantly reduced with the application of M. oleifera extract. M. oleifera extract may contribute to the prevention of dermatological side effects of antiepileptic drugs and the development of new therapeutics for skin health. Y. , YANARDAĞ R., YARAT A.© 2022The aim of current work was able to show the oxidant effect of cancer cells found in any part of the body on the liver and to investigate the possible protective effect of deuterium-depleted water (DDW) on this oxidant effect by determining of some liver parameters. Ehrlich ascites tumor bearing BALB/c mice were used for this purpose. BALB/c mice were selected randomly and divided into four groups (n = 5 in each group) as control group, tumor group, control+DDW group, tumor+DDW group, fifteen days after tumor cell injection, liver tissue samples were taken for all groups. In the tumor group, liver lipid peroxidation, sialic acid and protein carbonyl levels, xanthine oxidase, myeloperoxidase, catalase, gamma-glutamyl transferase, sorbitol dehydrogenase, glutathione peroxidase and glutathione reductase activities, were significantly higher than those in the control group while glutathione levels and paraoxonase1, sodium potassium ATPase, glutathione-S-transferase, alanine transaminase and aspartate transaminase activities decreased significantly. Compared with the tumor group, the changes in all parameters except sialic acid, catalase, alanine transaminase and aspartate transaminase were reversed in the DDW given tumor groups, while sialic acid and catalase values continued to increase, and alanine transaminase and aspartate transaminase values continued to decrease. This study aimed to investigate the changes in the brain after EX and/or CR in metabolic syndrome (MeS) induced by a high fructose diet in rats. Sprague-Dawley male rats were divided into five groups. Drinking water including 10% fructose solution was given to rats for 12 weeks to develop a MeS rat model. Animals with MeS were submitted to EX and/or CR for 6 weeks. Blood glucose, and brain tissue damage and antioxidant parameters were measured. Brain lipid peroxidation, sialic acid, mucin, fucose levels increased in the MeS group compared to the control (C) group. These parameters reduced significantly in the metabolic syndrome with caloric restriction (MeS+CR) group, and more significantly in the metabolic syndrome with exercise and caloric restriction group (MeS+EXCR), compared to the MeS group. Glutathione levels, superoxide dismutase and catalase activities decreased in the MeS group compared to the C group, increased both in the MeS+CR group, and MeS+EXCR group compared to the MeS group. F., Sacan O., Yanardag R., Yarat A.This study aimed to explore the potential protective impacts ofMoringa oleiferaextract on major alteration in salivary glands of rats exposed to sodium valproate (VA). Groups were defined as control, control+moringa extract, sodium valproate, and sodium valproate+moringa extract. Antioxidant and oxidant status, activities of digestive and metabolic enzymes were examined. VA treatment led to various biochemical changes in the salivary glands, including decreased levels of antioxidants like glutathione, glutathione-S-transferase, and superoxide dismutase (except for sublingual superoxide dismutase). Conversely, a decrease in alpha-amylase, alkaline and acid phosphatase, lactate dehydrogenase, protease, and maltase activities were observed. The study also demonstrated that VA induces oxidative stress, increases lipid peroxidation, sialic acid, and nitric oxide levels in the salivary glands. Total oxidant capacity was raised in all glands except in the sublingual gland. The electrophoretic patterns of proteins were similar.Moringa oleiferaextract exhibited protective properties, reversing these VA-induced biochemical changes due to its antioxidant and therapeutic attributes.
