B., Tacal Aslan B., Yılmaz Ö. Ö., Polat T., Ulucan K.Amaç:Çürük oluşumu pek çok faktörün etkilediği bir süreçtir. Yapılan çalışmalar genetik faktörlerin de çürük oluşumunda rol oynadığını göstermiştir. Çalışmamızda, östrojen hormon mekanizması ve mensturasyon döngüsünün gen ekspresyon sürecini değiştirmesi sebebiyle, erkek bireylerde diş mineralizasyon sürecinde rol oynayanMMP3veVDRgenlerinde oluşan polimorfizmlerin çürük oluşumuna olan etkisinin incelenmesi amaçlanmıştır.Gereç ve Yöntem:Fakülte hastanesine başvuran 20-44 yaş aralığındaki erkek bireylerde rutin ağız içi muayenesi sonrasında; çürük, eksik ve restorasyonlu dişler (DMFT) indeksine göre tanı konulmuştur. Deney grubunu, ‘yüksek çürük risk’ (DMFT ≥ 14, n=28); kontrol grubunun ise ‘çürük lezyonu bulunmayan’ (DMFT=0, n=28) kişiler oluşturmuştur. Detaylı anamnez alınan katılımcılardan plak indeksi, sondalamada kanama ölçümü yapılmıştır.Katılımcılardan alınan kan örneklerinden DNA izolasyonları sonrasında,MMP3(rs679620)veVDR(rs731236)genotiplemesi Real-time PZR tekniği kullanılarak belirlenmiştir.Elde edilen sonuçlar istatistiksel yöntemler kullanılarakanlamlılık düzeyi p<0,05’te değerlendirilmiştir.Bulgular:Çalışmamıza katılan bireylerde sondalamada kanama derecesi %10’un altında ve DMFT=0 olan düşük risk grubundaki (kontrol) ve de sondalamada kanama derecesi %10’un altında olan yüksek risk grubundaki bireylerin (deney)MMP3veVDRpolimorfizmleri açısından kıyaslaması yapılmış ve gruplara göreMMP3dağılımları arasında istatistiksel olarak anlamlı farlılık bulunmuştur (p<0,001). Gruplara göre VDR dağılımları arasında istatistiksel olarak anlamlı farklılık bulunmamıştır (p=0,659)Sonuç:Çalışmamızın sınırları dahilinde MMP3(rs679620) gen polimorfizminin çürük oluşum üzerinde etkili;VDR(rs731236) polimorfizminin ise etkisiz olduğu sonucuna ulaşılmıştır. M., DOĞU KAYA B., MANAV ÖZEN A., Öztürk S., ALKAN E., YILMAZ ATALI P., KORKUT B., TAĞTEKİN D.Introduction: The purpose was to investigate the surface roughness and the color changes of nanohybrid and microhybrid resin composites subjected to calcium active whitening toothpastes with different RDA values through a novel Press-on Force-Guided (PFG) Brushing Simulator. Material and Methods: Twenty-eight disc-shape samples (10x2mm) of a nanohybrid (Herculite, Kerr) and a microhybrid (Z250, 3M) composites were prepared and randomly divided into 4 subgroups (n=7). The samples were polymerized with a LED-curing unit with an irradiance of 1000 mW/cm2 (Valo Cordless, Ultradent). Finishing and polishing procedures were performed with 2 fine grits of Sof-Lex Discs (3M). Samples were stored in distilled water at 37°C for 24 hours. Composite samples were brushed with a medium-bristle toothbrush (ROCS Pro 5940, ROCS) using the PFG brushing simulator (Marmara University, Turkey) for 60.000 strokes (correspondence to 6 years clinically) in a slurry containing toothpaste with 120 RDA (Ca Active Whitening, ROCS) and 70 RDA (Electro Whitening, ROCS) under 300-320g load (200 strokes/min). In particular, surface roughness (Ra) and color change (ΔE) were investigated utilizing profilometer (MarSurf PS 10, Mahr) and spectrophotometer (Easy-Shade-5, VITA). Analysis of variance was used for repeated measurements; Bonferroni test was used for pairwise comparisons. The level of significance was set at p<0.05. Results: After brushing Ra values did not show a significant difference (p>0.05) except Herculite group brushed with Electro whitening (p=0.05) which showed the most roughness change (36.99%). There is a significant difference between Herculite (1.163±0.701) and Z250 (0.400±0.154) in Ra values after electro-whitening brushing (p=0.002). No significant color change was observed in any group, regardless of composite or tooth-paste type (p>0.05). ΔE values of Herculite (1.007±0.463; 1.100±0.452) and Z250 (1.558±0.618; 1.196±0.835) brushed with Ca Active Whitening and Electro Whitening toothpastes were less than 1.8 respectively. Conclusion: Tooth brushing with Ca active whitening toothpastes (different RDA values) affected differently on the surface roughness of Herculite and Z250 due to their filler and resin composition, but there is no effect on color. Materials and Methods: 80 enamel samples were kept in the demineralization solution (pH 4.5, 37°C) for 96 hours. Demineralized enamel samples will be randomly divided into 8 groups (n=10) including a remineralizing agent(Toothmousse/GC, Remin Pro/VOCO, Medical Mineral Gel/ROCS), resin infiltration (ICON/DMG), and a combined treatment of both. All remineralization agents were applied in remineralization cycle 2 times a day for 1 week.Measurements made from the enamel samples at the baseline, after demineralization and remineralization procedures evaluated with the OCT (Topcon3D OCT-1 Maestro) and ultrasonic system (Novascope 4500:NDT) lesion depths were measured. The sample surfaces were examined under SEM/EDS (Zeiss EVO-MA10) at x1000, 2500, 5000 and 10000. Data were analyzed in R program with WRS2. Two-way Robust Anova, Benferroni Test, and ICC (Intra-Class Correlation Coefficient) was used to examine the agreement between measurements. Significance level was set at p<0.05. Results: There was no statistical difference between the groups in OCT (p=0.884) and Ultrasound (p=0.051) median values after the protocols. In both measurement methods, the effect of remineralization treatments was statistically significant for lession depth (p<0.01). OCT median values were 104.01 in baseline, 144.78 in post-demineralization, and 90.5 in post-remineralization. Ultrasound median values were 1.34 in baseline, 1.1 in postdemineralization, and 2.36 inpost-remineralization. The highest ultrasound median value was obtained in the Remin Pro+ICON after remineralization (2.67), while the lowest value was obtained after demineralization (1.04). As a result of EDS analysis, the lowest Ca ratio (%atomic) was observed on the grup 5-ICON applied enamel surface (9.35). Çalışmamızın amacı;VDRveMMP3genlerinde meydana gelen gen polimorfizmlerin etkilerinin değerlendirilip çürük risk modellemesi üzerine olan etkilerinin incelenmesidir.Gereç ve Yöntem:Çalışma populasyonu, fakülte hastanesine başvuran 20-44 yaş aralığındaki erkek bireylerde rutin ağız içi muayenesi sonrasında çürük, eksik ve restorasyonlu dişler (DMFT) indeksine göre tanı konulan ‘yüksek çürük risk’(DMFT ≥ 14) ve düşük çürük risk’(DMFT£5) olarak 2 gruptan oluşturulmuştur (n=160). Detaylı anamnez alınan katılımcılardan plak indeksi, gingival indeks sondalamada kanama, sondalama derinliği, klinik ataşman seviyesi ve tükürük tamponlama kapasitesi, tükürük streptekok mutans (SM) ve laktobasil (LB) sayısı ölçülmüştür.Katılımcılardan alınan kan örneklerinden DNA izolasyonları sonrasında,MMP3(rs679620)veVDR(rs731236)genotiplemesi ise Real-time PZR tekniği kullanılarak belirlenmiştir.Elde edilen sonuçlar istatistiksel yöntemler kullanılarakanlamlılık düzeyi p<0,05’te değerlendirilmiştir.Bulgular:Gruplar arası plak indeksi, gingival indeks, sondalama derinliği, klinik ataşman seviyesi, sondalamada kanama, tükürük akış hızı, tükürük tamponlama kapasitesi ve tükürük SM ve LB sayısı faktörleri arasında anlamlı düzeyde fark bulunmuştur (p<0,001). Resin matrix, filler type, size and amount, and photoinitiator types influence the degree of conversion. LED-cured bulk fill composites achieved sufficient polymerization. Scanning electron microscope (SEM) analysis revealed different patterns of surface roughness, depending on the composite material. Bulk fill materials showed surface characteristics similar to those of nanohybrid composites. Based on the thermal analysis results, glass transition (T-g) and initial degradation (T-i) temperatures changed depending on the bulk fill resin composites.Publication Metadata only Class IV composite restorations of central Incisors: One-shade vs multishade, case series(2022-04-23) DOĞU, BENGÜ; TARÇIN, BİLGE; YILMAZ ATALI, PINAR; DOĞU B., YILMAZ ATALI P., TARÇIN B.Aim: Fractures or deformities are common in central incisors. The aim of this case series was to compare and evaluate incisal/class IV restorations using 3 different composites: OptiShade (Kerr), Amaris (Voco), and Ceram-X Duo (Dentpsly-Sirona). Case Description: 3 patients with the complaint of fractures on their anterior teeth were applied to the clinics of Restorative Dentistry Department (Case1 18-year-old female, Case 2 20-year-old female, and Case 3 17-yearold male). Following radiographic and clinical examinations, enamel fracture was detected in the incisal 1/3 of the central incisors and it was decided to perform direct composite resin restorations (CRR) as the treatment plan. 37% orthophosphoric acid (Kerr) was used in selective-etch mode. 3 different universal adhesive agents (OptiBond Universal, Kerr; Futurabond U, Voco; and Prime&Bond Universal, Dentsply-Sirona) were applied to the cavities according to manufacturer’s instructions. CRRs were completed with nano-hybrid (Amaris TL and D2, Voco), one-shade (OptiShade Light and Medium, Kerr), and ceramic reinforced (Ceram-X Duo D2 and E1, Denstply-Sirona) composites using incremental technique. Valo-Cordless (Ultradent) was used for polimerisation with 1000 mW/cm2. Finishing and polishing procedures were performed with polishing discs (Bisco) and Twist Dia (Kuraray) polishing wheels. Patients were recalled at 1 and 3-months for clinical evaluation and scores according to FDI criteria. Results: The esthetic, functional and biological evaluation of the restorations according to the FDI criteria in 1 and 3-months follow-up were scored as 1. Discussion: One shade universal composite provide advantage in color selection for the clinician, however composites with different contents and brands vary in opacity, color, and brightness. Considering the esthetic expectation of the patients and the longevity of class IV CRR, the material should be selected considering the shape, color and fracture line of the tooth structure and the optic properties of the material.Publication Metadata only Effect of sodium ascorbate and delayed treatment on the shear bond strength of composite resin to enamel following bleaching(WOLTERS KLUWER MEDKNOW PUBLICATIONS, 2016) TÜRKMEN, CAFER; Turkmen, C.; Guleryuz, N.; Atali, P. Y.Background: The effect of bleaching on enamel surfaces, as well as exploring methods of preventing the weakening of bond strength of two different adhesives to bleached enamel surfaces. Objective: This study compared the shear bond strength (SBS) of two different adhesives on bleached enamel immediately after bleaching, with a 1-week delay, and following application of an antioxidizing agent after bleaching. Materilas and Method: The enamel surfaces of 140 incisors were divided into 14 groups. Two unbleached enamel groups (n = 20) were prepared as negative controls of the Adper Single Bond 2 total-etch and Clearfil Tri S Bond self-etch adhesives. The remaining surfaces were randomly divided into two bleaching groups treated with 35% Beyond Max (n = 60) and 38% Opalescence Xtra Boost (n = 60). Each bleaching group was then divided into two subgroups using the total and self-etch adhesive systems. Specimens were bonded immediately after bleaching, after treatment with 10% sodium ascorbate (SA) gel, or after 1-week of immersion in artificial saliva. All groups were restored with composite resin. After thermocycling, SBS was measured and data were analyzed. Results: In the control groups, there was no statistically-significant difference in SBS between self-and total-etch adhesives. Among all bleaching groups, no statistically-significant differences in SBS were noted, although the SA-treated delayed bonding (1-week) groups had the highest values. Two types' four composites were studied: Surefill SDR (SDR) and Xtra Base (XB) as bulk-fill flowable materials; QuixFill (QF) and XtraFill (XF) as packable bulk-fill materials. Samples were prepared for each analysis and test by applying the same procedure, but with different diameters and thicknesses appropriate to the analysis and test requirements. Thermal properties were determined by thermogravimetric analysis (TG/DTG) and differential scanning calorimetry (DSC) analysis; the Vickers microhardness (VHN) was measured after 1,7, 15 and 30 days of storage in water. The degree of conversion values for the materials (DC, %) were immediately measured using near-infrared spectroscopy (FT-IR). The surface morphology of the composites was investigated by scanning electron microscopes (SEM) and atomic-force microscopy (AFM) analyses. The sorption and solubility measurements were also performed after 1, 7, 15 and 30 days of storage in water. In addition to his, the data were statistically analyzed using one-way analysis of variance, and both the Newman Keuls and Tukey multiple comparison tests. The statistical significance level was established at p < 0.05. According to the ISO 4049 standards, all the tested materials showed acceptable water sorption and solubility, and a halogen light source was an option to polymerize bulk-fill, resin-based dental composites.
