    In vitro antioxidant and cytotoxic activity of Muscari neglectum growing in Turkey

    Muscari species have been used as antirheumatic, stomachic, diuretic, expectorant and anti verruca in folk medicine. The species have also been used as food for humans and animals in Turkey. In this study, the antioxidant and cytotoxic activities of different extracts obtained from Muscari neglectum were studied. The antioxidant capacities of the extracts were evaluated with DPPH free radical scavenging activity, metal chelating capacity and ABTS cation radical scavenging capacity assays. The cytotoxic potentials of the extracts were determined by MTT and LDH assays on HeLa-cells (human cancer cell line) and NRK-52E-cells (rat kidney cell line). The aqueous extracts from M. neglectum bulbs (IC50 : 2.83 +/- 0.54 mg/mL) and herba (IC 50 : 8.52 +/- 1.3 mg/mL) exhibited stronger DPPH free radical scavenging activity than other extracts, respectively. The ethanol extract from M. neglectum bulbs showed the highest ABTS (27.88 %) cation radical scavenging activity. According to the metal chelating activity test results, the aqueous extracts from M. neglectum herba and bulbs showed the strongest metal chelating activity (28.99% and 28.07%, respectively).

    Phenolic composition and biological properties of Achillea nobilis L. subsp neilreichii (Kerner) Formanek

    Achillea species are widely distributed medicinal plant throughout the world and have been used since ancient time. There are many reports about the traditional use and highly bioactive compounds of these plants. Therefore, it is fundamental to study in detail the chemical composition and biological activities of Achillea species. Thus, the objective of this study was to determine for the first time the effects of methods/solvents on biological activities (antioxidant, antimicrobial, and anti-urease) of Achillea nobilis L.subsp. neilreichii (Kerner) Formanek 15 extracts obtained from different extraction methods/solvents and then to analysis the phenolic contents of the active extracts. The phenolic contents in ethyl acetate and ethanol extracts, which showed strongest phenolic contents and antioxidant activity were analysed separately by high-performance liquid chromatography with diode array detection and were confirmed by liquid chromatography with quadrupole time-of-flight mass spectrometry using targeted mass spectra techniques with accurate mass. The active phenolic compounds were identified as dicaffeoylquinic acid, luteolin-7-O-glucoside, orientin, vitexin, luteolin, quercetin-methyl-ether, isorhamnetin-glucoside in ethyl acetate extract; chlorogenic acid luteolin-7-O-glucoside, quercetin-methyl-ether, luteolin, orientin, vitexin, dicaffeoylquinic acid, apigenin-6,8-di-C-glucoside, luteolin hexoside hexoside in ethanol extract. As a result of this study, dicaffeoylquinic acid, isorhamnetin-glucoside, chlorogenic acid, and apigenin-6,8-di-C-glucoside compounds were analysed for the first time in Achillea nobilis L. subsp. neilreichii (Kerner) Formanek. Therefore, it is thought that ethanol and ethyl acetate extracts can be used as a natural source in the future for food and pharmaceutical industry.

    Fabrication, characterization and fibroblast proliferative activity of electrospun Achillea lycaonica-loaded nanofibrous mats

    The use of natural compounds such as biocompatible and non-toxic plant extracts, without undesired side effects, in tissue engineering applications, is highly preferred compared to chemical drugs. In this study, the characterization and performance of electrospun Achillea lycaonica-loaded (0.125, 0.250 and 0.500, wt%) poly (lactic acid) (PLA) (8%, w/v) nanofibrous mats for skin tissue engineering were investigated. SEM, FTIR, DSC, and tensile strength test of the electrospun nanofibers have been investigated. Drug releasing test and cell culture study were also carried out. Achillea lycaonica-loaded nanofibrous mats in 0.250 (wt%) and 0.500 (wt%) demonstrated excellent cell compatibility and increased the viability of NIH/3T3 (mouse embryo fibroblast) cells within 72 h. According to the results, Achillea lycaonica-loaded PLA nanofibers have proper tensile strength and controlled release. The working temperature range enlarged for the composites having higher plant extract content. Consequently, Achillea lycaonica-loaded nanofibrous mats have a great potential in skin tissue engineering applications.

    Characterization of Phenolic Constituents and Pharmacological Activity of Achillea vermicularis

    The aim of the present study was to compare for the first time, the antiinflammatory, antioxidant and antiurease activities of 15 different extracts of Achillea vermicularis, which were obtained from three extraction methods using five different solvents. Phenolic contents in chloroform extract showed strong antiinflammatory and antioxidant activity and were analysed by high-performance liquid chromatography with diode-array detection and confirmed by liquid chromatography quadrupole time-of-flight mass spectrometry. The active phenolic compounds in chloroform extracts were identified as chlorogenic acid, caffeic acid, rutin, dicaffeoylquinic acid, naringenin, quercetagetin 3,6-dimethyl ether, and 8-hydroxysalvigenin. All these compounds except caffeic acid were detected for the first time in Achillea vermicularis. These results indicated that chloroform extract of the plant possessed a high potential to be used as a natural source in food and pharmaceutical industry.

    Investigation of the Effects of Biological Activity and Steeping Time of Four Different Black Teas Commercially Valuable in Turkey

    Objective: The aim of this study was to compare the phenolic, tannins contents and biological activities (antioxidant, anticholinesterase) of four different black teas commercially valuable in Turkey. Subsequently, to examine the effect of the steeping time on the chemical contents (phenolic and tannins contents) and biological activities of the samples. Methods: The antioxidant activities of samples were examined by 2,2-diphenyl-1-picryl-hydrazyl (DPPH), cupric reducing antioxidant capacity (CUPRAC) and ferric reducing/antioxidant power (FRAP) methods. The amount of total phenolic and tannins contents contained in the samples was determined using the Folin-Ciocalteu reagent (FCR) method. In addition, the anticholinesterase activities of different samples were evaluated by Ellman method. Results: According to the results obtained, filtered tea bag (FT) sample obtained after 5 min steeping exhibited stronger DPPH radical scavenging activity than other samples. In addition, it was found that powder ceylon tea (PCT) samples obtained after 60 min and 10 min steeping had the highest FRAP and CUPRAC values respectively. In this study, it was also determined that the PCT samples obtained after 5 min, 10 min, 15 min, and 30 min steeping contained higher phenolic and tannins contents than the other samples. In anticholinesterase experiment, powder tea (PT) sample obtained after 5 min steeping was found to have the strongest anticholinesterase activity. Conclusion: As a result, it was determined that the amount of phenolic contents generally increased with the infusion time. It was also found that the antioxidant, anticholinesterase activities and tannins content of the tea samples did not show significant change due to the time of the infusion.

    In vitro and In vivo biological activities and phenolic characterization of Thymus praecox subsp. skorpilii var. skorpilii

    Thymus species are used as food and spices in several Mediterranean and other countries as well as for medical purposes on account of their biological properties. The extraction procedures and solvents are an important step in processing bioactive constituents from the plant materials. Therefore, the aim of this study was to evaluate in vitro antioxidant, anticholinesterase, antimicrobial, anti-urease and in vivo anti-inflammatory activities of Thymus praecox subsp. skorpilii var. skorpilii using a variety of extracts. In addition, the phenolic characterization of the fraction methanol extract, which showed the highest phenolic contents and the strongest antioxidant and anticholinesterase activity were performed by HPLC-DAD and LC-MS/MS techniques. In the present study, the fraction methanol and direct methanol extracts exhibited strong DPPH, ABTS, and ferric reducing activity and strong antibacterial activity against Staphylococcus epidermidis strains. The fraction chloroform and fraction petroleum ether extracts showed the strongest anti-urease activity and also, the petroleum ether extract exhibited strong antibacterial activity against the Staphylococcus aureus strain. The fraction methanol and direct methanol extracts showed in vivo anti-inflammatory activity very similar to each other. As a result of this study, chlorogenic acid, luteolin-7-O-glucoside, 3-O-feruloylquinic acid, quercetin-3-O-hexoside, and apigenin-7-O-glucuronide compounds were analyzed for the first time in the methanol extract of the plant. This latter study revealed the effects of the extraction methods and phenolic compounds on its biological properties.

    Chemical composition and antioxidant, cytotoxic, and insecticidal potential of Valeriana alliariifolia in Turkey

    Valeriana is a common plant species used for various healing purposes in folk medicine since antiquity. This study investigates the phytochemical profile, antioxidant, cytotoxic, and insecticidal activity of Valeriana alliariifolia Adams, a species that has traditionally been used in Turkey. For the analyses we prepared four root extracts of V. alliariifolia Adams using hexane (HM1), chloroform (CM1), ethanol (EM1), and water (WM1) for maceration. Additionally, two extracts were also prepared from its roots by maceration separately with ethanol (EM2) and water (WM2). One sample was prepared as a water infusion (WI), according to the procedure used in Turkish traditional medicine. The 2,2-Diphenyl1-picrylhydrazyl (DPPH) scavenging and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS) radical cation scavenging activity tests showed that ethanol extracts had the strongest antioxidant activity: EM1 (IC50 - DPPH: 17.694 mu g/mL; ABTS: 23.8 mu g/mL) and EM2 (IC50 - DPPH: 20 mu g/mL; ABTS: 21.5 mu g/mL). The hexane extract, HM1, was the most cytotoxic (IC50<10 mu g/mL against HepG2 and HUVEC) and EM2 strongly cytotoxic (IC50<10 mu g/mL against HepG2 and IC50: 11.96 mu g/mL against HUVEC). The extracts with demonstrated cytotoxic activities were further examined to check their insecticidal activity against adult female mosquito Aedes aegypti and first instar Ae. aegypti larvae. HM1 was the most effective (90 +/- 10 %), which was consistent with its cytotoxic activity. Because of the high antioxidant, cytotoxic, and insecticidal activities, we ran phytochemical analyses of the HM1, EM1, and EM2 extracts with GC-MS (for HM1) and LC-MS/MS (for EM1 and EM2). We also analysed the composition of the essential oil obtained from V. alliariifolia roots by micro-distillation in order to compare its content with HM1, which contains volatile compounds. Phytochemical analyses revealed that the major compound in HM1 was isovaleric acid (16 %) and in the essential oil 1,8-cineole (2.9 %). EM1 and EM2 contained 5-O-caffeoylquinic acid (chlorogenic acid), verbascoside (acteoside), and 3,5-dicaffeoylquinic acid as major components. In the light of our findings and available literature, we can conclude that V. alliariifolia has a good bioactive potential that could be used for different purposes, including the development of new agents for the treatment of various diseases. The difference in the content between the essential oil and HM1 was remarkable. It suggests that the variability observed in the activity of the samples was a result of composition and that, therefore, the aim of treatment should dictate which type of preparation is to be selected. An added value of our study is that it determined verbascoside and methylquercetin rutinoside for the first time in the Valeriana extracts.
