    In vitro carbonic anhydrase inhibitory effects of the extracts of satureja cuneifolia

    Carbonic anhydrases catalyze the interconversion between carbon dioxide and bicarbonate. Carbonic anhydrase inhibition has therapeutic importance and there are many drugs which use this mechanism. Carbonic anhydrase inhibitors are used as diuretic, antiglaucoma, antitumor and antiepileptic agents. As an alternative for synthetic chemicals with strong side effects, natural products have gained popularity in the recent years. In this study, Satureja cuneifolia, a medicinal plant used as folk medicine, was investigated for its inhibition effects on carbonic anhydrase I and II enzymes. Four different extracts were obtained with maceration method and three different solvents were used. The results have shown that methanol extracts of Satureja cuneifolia has the strongest inhibition activity on the enzymes with the IC50 values of 31 µg/mL for hCA I and 12 µg/mL for hCA II. Further purification and analytical studies will be needed to obtain the active natural molecules and their potential for pharma and food industries.

    Antioxidant and antimicrobial activities of various extracts from Stachys cretica subsp. bulgarica Rech.f., Stachys byzantina K. Koch and Stachys thirkei K. Koch

    Background and Aims: Stachys L. species, which are members of the Lamiaceae family, have long been applied for their therapeutic benefits especially to cure cough, infection, genital tumors, ulcers, inflammatory illnesses, and wounds in Anatolian traditional medicine. In the current study, various extracts prepared from the aerial parts of Stachys cretica subsp. bulgarica Rech.f. (SC), Stachys byzantina K. Koch (SB), Stachys thirkei K. Koch and were tested for their in vitro antioxidant, antibacterial, and anticandidal properties. Methods: The aerial parts of three Stachys species were sequentially extracted using n-hexane, chloroform, and methanol. Aqueous extracts of each sample was also prepared by infusion process. The total phenolic content of each extract was determined and the contribution of the biological activities in the samples was evaluated. To assess the antioxidant capacity, samples were studied using CUPRAC activity, DPPH• free radical scavenging, and FRAP methods. The antimicrobial activity of the extracts was tested against 7 bacteria and 3 yeast. Results: The infusion and methanol extract exhibited the strongest antioxidant potential and also had the highest percentage of phenolics among the studied extracts. The n-hexane extracts of all studied species showed considerable antifungal activity with MIC values ranging from 312.5-78.12 mg/L. Conclusion: According to our results, three Stachys species were found to be beneficial for their antioxidant and antimicrobial properties.     In vitro antioxidant and cytotoxic activity of Muscari neglectum growing in Turkey

    Muscari species have been used as antirheumatic, stomachic, diuretic, expectorant and anti verruca in folk medicine. The species have also been used as food for humans and animals in Turkey. In this study, the antioxidant and cytotoxic activities of different extracts obtained from Muscari neglectum were studied. The antioxidant capacities of the extracts were evaluated with DPPH free radical scavenging activity, metal chelating capacity and ABTS cation radical scavenging capacity assays. The cytotoxic potentials of the extracts were determined by MTT and LDH assays on HeLa-cells (human cancer cell line) and NRK-52E-cells (rat kidney cell line). The aqueous extracts from M. neglectum bulbs (IC50 : 2.83 +/- 0.54 mg/mL) and herba (IC 50 : 8.52 +/- 1.3 mg/mL) exhibited stronger DPPH free radical scavenging activity than other extracts, respectively. The ethanol extract from M. neglectum bulbs showed the highest ABTS (27.88 %) cation radical scavenging activity. According to the metal chelating activity test results, the aqueous extracts from M. neglectum herba and bulbs showed the strongest metal chelating activity (28.99% and 28.07%, respectively).

    The effects of Origanum onites in streptozotocin-induced diabetes mellitus in rats

    © 2022 Marmara University Press.Diabetes mellitus is a chronic disease characterized by decreased insulin synthesis and/or increased blood glucose due to insulin resistance. In this study, streptozotocin (STZ)-induced diabetic rats were evaluated and it was aimed to investigate the protective effects of Origanum onites L. (OO) against possible changes in these parameters. Male Sprague-Dawley rats weighing 300-400g were divided into three groups control (C), diabetes mellitus (DM) and diabetes mellitus + OO (OO group). DM was induced by administration of STZ 60 mg/kg dose intraperitoneally (i.p.) after 48 hours, rats with blood glucose values higher than 200 mg/dL were considered DM. Origanum extract was administered i.p. to the OO group at 50 mg/kg per day for 6 weeks. Serum AST, ALT, creatinine, and inflammatory cytokine levels were measured. MDA and GSH levels were measured in liver tissues. AST, ALT, creatinine, and MDA levels were found to be increased in the DM group, while a significant decrease in these levels was observed in the treated group. While GSH values fell in the DM group, a significant increase occurred in the OO group (n=6, p<0.0001; two-way ANOVA). When the plasma levels of cytokines were examined, an increase was observed in the DM group and a significant decrease was observed in the OO group. When we evaluate our findings, we think that OO has a protective effect against complications that may occur in DM by preventing oxidant damage and inflammation. Further studies are needed on the protective effects of OO in DM.

    Camellia Sinensis Leaves Hydroalcoholic Extract Improves the Alzheimer's Disease-Like Alterations Induced by Type 2 Diabetes in Rats

    Objective: Novel investigations have confirmed that hyperglycemia is strictly associated with the development of cognitive impairment and dementia. Sodium-dependent glucose transporter (SGLT) inhibitors, which are oral antidiabetic drugs, are currently being investigated as the medication in Alzheimer's disease (AD). In our study, Camellia sinensis (green tea), which inhibits sodium-dependent glucose transporter-1 (SGLT-1), was used in the treatment of type 2 diabetes mellitus (T2DM)-induced AD-like alterations via its antidiabetic effects. Methods: High-fat diet/streptozotocin-treated rat model was chosen to provide T2DM-induced AD-like alterations. Antidiabetic effects were evaluated with the measurement of blood glucose level (BGL), oral glucose tolerance test (OGTT), and insulin tolerance test (ITT). On the other hand, novel object recognition test (NORT), open field test (OFT), passive avoidance test (PAT), and Morris's water maze (MWM) test were performed to investigate the anti-Alzheimer's effects of C. Sinensis. Results: C. sinensis tolerated BGL for a short time but metformin, the first medication prescribed for T2DM, tolerated BGL during the test for 120 min. C. sinensis increased the number of square crosses and the frequency of grooming activity in a similar manner to metformin in OFT. C. sinensis treatment improved exploratory behavior and memory retention components in NORT. The step-through latency decreased in HFD/STZ-treated rat model but it improved with metformin and C. sinensis treatment in PAT. According to the results obtained by the MWM test, C. sinensis treatment slightly improved learning. Conclusion: C. sinensis improved short-term memory and increased the locomotor activity in rats according to the results obtained by NORT, OFT, and PA.

    Investigation of the Effects of Biological Activity and Steeping Time of Four Different Black Teas Commercially Valuable in Turkey

    Objective: The aim of this study was to compare the phenolic, tannins contents and biological activities (antioxidant, anticholinesterase) of four different black teas commercially valuable in Turkey. Subsequently, to examine the effect of the steeping time on the chemical contents (phenolic and tannins contents) and biological activities of the samples. Methods: The antioxidant activities of samples were examined by 2,2-diphenyl-1-picryl-hydrazyl (DPPH), cupric reducing antioxidant capacity (CUPRAC) and ferric reducing/antioxidant power (FRAP) methods. The amount of total phenolic and tannins contents contained in the samples was determined using the Folin-Ciocalteu reagent (FCR) method. In addition, the anticholinesterase activities of different samples were evaluated by Ellman method. Results: According to the results obtained, filtered tea bag (FT) sample obtained after 5 min steeping exhibited stronger DPPH radical scavenging activity than other samples. In addition, it was found that powder ceylon tea (PCT) samples obtained after 60 min and 10 min steeping had the highest FRAP and CUPRAC values respectively. In this study, it was also determined that the PCT samples obtained after 5 min, 10 min, 15 min, and 30 min steeping contained higher phenolic and tannins contents than the other samples. In anticholinesterase experiment, powder tea (PT) sample obtained after 5 min steeping was found to have the strongest anticholinesterase activity. Conclusion: As a result, it was determined that the amount of phenolic contents generally increased with the infusion time. It was also found that the antioxidant, anticholinesterase activities and tannins content of the tea samples did not show significant change due to the time of the infusion.

    Phytochemical screening and biological evaluation of Salvia hydrangea Dc. ex Benth. growing in eastern Anatolia

    Salvia species have lately gained significant interest as a result of their suitable utilization in various industries. In the current study, S. hydrangea, one of the most consumed sages in the eastern region of Turkey, has been evaluated for phytochemical composition as well as in vitro pharmacological potential comparatively for the first time. The phytochemical composition of S.hydrangea was investigated by LC-MS/MS, GC-FID, and GC/MS. To reveal its biological activities, antioxidant, antimicrobial, and also acetylcholinesterase activities of different solvent extracts such as water, n-hexane, chloroform, and methanol were determined. According to GC-GC/MS analysis, the primary components of the oil were identified as camphor (46.0%), 1,8-cineole (7.5%), camphene (6.8%), limonene (6.5%), beta-pinene (6.11%) and alpha-pinene (5.6%). Additionally, in the infusion and methanol extract, rosmarinic acid and luteolin glycoside were detected as predominant phenolics by LCMS/MS. In DPPH center dot, CUPRAC, and FRAP test results of the samples indicated strong to moderate antioxidant ability in all samples studied, additionally, among them, the infusion exhibited significant acetylcholine inhibition properties comparable with galanthamine. With regard to antimicrobial activity, all of the tested microorganisms had MIC values ranging from 15 to 2000 mu g/mL. Based on these findings, S. hydrangea may have promising properties for a variety of industrial applications in the pharmaceutical, food, and cosmetic industries. (C) 2022 SAAB.     The in vitro and in vivo investigation of biological activities and phenolic analysis of Helichrysum plicatum subsp. plicatum

    In Turkey, Helichrysum genus is represented by 26 taxa belonging to 20 species in Turkish flora of which 14 ones arc endemic to Turkey. The aerial parts of Helichrysum plicatum subsp. plicatum are used kidney stones, kidney and stomach ailments. The extraction procedures and solvents are important step in processing of bioactive constituents from the plant materials. Therefore, the aim of this study was to evaluate in vitro antioxidant, antimicrobial, anti-urease, anticholinesterase and in vivo anti-inflammatory activities of Helichrysum plicatum subsp. plicatum different extracts. In addition, the phenolic characterization of the Soxhlet and maceration methanol extracts which showed significant antioxidant, anti-urease, antimicrobial, anti-inflammatmy and anticholinesterase activities were performed by HPLC-DAD and LC-MS/MS. In the present study, the Soxhlet methanol extract exhibited strong antioxidant, antimicrobial and anticholinestemse activities than other extracts. The maceration methanol extract showed the strongest anti-urease activity. The Soxhlet methanol and maceration methanol extracts showed in vivo anti-inflammatory activities very close to each other. As a result of this study, chlorogenic acid, dicaffeoylquinic acid, luteolin, luteolin-7-O-glucoside, naringenin-O-hexoside and isoquercitrin compounds were analysed in plant. Therefore, it is thought that methanol extracts can be used as a natural source in the future for food and pharmaceutical industries.

    Identification, antioxidant and cytotoxic potentials of casticin in Vitex agnus-castus fruit from different geographical regions of Turkey

    Purpose: To evaluate the antioxidant and cytotoxic effects, and casticin contents of Vitex agnus-castus (VAC) fruit extract from five geographical regions of Turkey. Methods: Casticin determination in VAC fruit extracts was performed using HPLC-DAD method. The method was validated for linearity, sensitivity, selectivity, accuracy and precision. The methanol extracts of the VAC fruits were analyzed for antioxidant effects using 2,2-diphenyl-1-picrylhydrazyl (DPPH center dot) radical scavenging capacity, metal-chelation capacity assay, radical cation scavenging capacity (ABTS(center dot+)), and total phenolic content. The cytotoxic potential of the extracts against NRK-52E and HeLa cells were determined using MTT and LDH assays. Results: The casticin content of the extracts ranged from 0.048 and 0.152 %. Total phenol content was in the range of 36.67 +/- 0.7 to 74.20 +/- 1.02 mg gallic acid equivalent (GAE)/100 mg of extracts. High-to-moderate antioxidant properties were observed in the extracts. Cytotoxicity data demonstrated that all extracts showed high cytotoxic effects on HeLa cell line when compared to the NRK-52E cell line (p < 0.05). Conclusion: These results suggest that VAC fruits (Monk's pepper fruits) from Aegean sea coasts have a potential for use in the preparation of phytopharmaceutical products.
