    In vitro investigation of antimicrobial, enzyme inhibitory and free radical scavenging activities of Inula salicina L.

    In this study, in vitro biological activities and total phenol/flavonoid contents of methanol extract (ISM) and its hexane (ISH), chloroform (ISC), ethyl acetate (ISEA) and aqueous methanol (ISAM) fractions obtained from aerial parts of Inula salicina were investigated. ISEA showed the highest antioxidant activity against DPPH and ABTS radicals with an IC50 value of 0.014 mg ml-1 for both assays. ISEA exhibited a good anti-inflammatory activity with an IC50 value of 0.060 mg ml-1 . ISEA was found to exhibit a moderate level of antidiabetic activity against α amylase enzyme with an IC50 value of 0.290 mg ml-1 . ISEA and ISM presented low and moderate inhibitory activity against acetylcholinesterase and butyrylcholinesterase enzymes with IC50 values of 0.577 and 0.279 mg ml-1 , respectively. ISC with MIC values of 78 and 156 µg ml-1 displayed a significant antimicrobial activity against Staphylococcus aureus and S. epidermidis, respectively. Almost all extracts had moderate effect against Candida species. The highest total phenolic and flavonoid contents were determined in ISEA with 574.8 mg GAE (gallic acid equivalent) g -1 extract and 30.48 mg QE (quercetin equivalent) g -1 extract, respectively. These results showed that ISEA had a good antioxidant and anti-inflammatory activity with moderate α-amylase and butyrylcholinesterase inhibitory activity. Also, ISC exhibited a significant antimicrobial activity against Staphylococcus species.

    Chemical composition, antiradical, and enzyme inhibitory potential of essential oil obtained from aerial part of Centaurea pterocaula Trautv

    The chemical composition, free radical scavenging, alpha-amylase inhibitory, and 5-lipoxygenase inhibitory activity of the essential oil obtained from the aerial parts of Centaurea pterocaula Trautv. were examined. Twenty-nine compounds corresponding to 93.2% of the oil were identified in the oil of C. pterocaula. Hexadecanoic acid (13.9%), caryophyllene oxide (11.5%), spathulenol (11.4%), (E)-beta-damascenone (5.0%), hexahydrofarnesyl acetone (5.0%), and tetradecanoic acid (4.9%) were the main compounds of the oil. The oil exhibited good anti-inflammatory activity, inhibiting 5-lipoxygenase with an IC50 value of 86.22 mu g/mL. The oil was found to be active against DPPH radical and alpha-amylase with IC50 values of 83.05 and 79.66 mu g/mL respectively, while it showed moderate activity against ABTS radicals with IC50 value of 360.9 mu g/mL. To the best of our knowledge, this is first report on the chemical composition of C. pterocaula essential oil from Turkey; as well as on the biological activity of its essential oil.

    Therapeutic effects of momordica charantia L. Ethanolic extract on acetic acid-induced ulcerative colitis in rats

(MoC) ethanolic extract on ulcerative colitis (UC) and was explored in vitro and in vivo. Materials and Methods: The rats were divided into control (C), saline-treated colitis (AA), MoC extract-treated colitis (AA+MoC), and sulfasalazine (SS)-treated colitis (AA+SS) groups. Colitis was induced by acetic acid. MoC extract, SS or saline were given to the related groups for 3 days. Interleukine-1β, malondialdehyde, glutathione levels, myeloperoxidase activity, bax/bcl-2 ratio, caspase-9 and caspase-3 levels were measured in colon and macroscopic and histopathologic examinations were done. Total phenolic/flavonoid content and biological activity of MoC were evaluated by in vitro analysis. Results: Compared to the control group, with acetic acid application interleukin-1β levels, myeloperoxidase activity, malondialdehyde levels, bax/bcl-2 ratio, caspase-9 and caspase-3 levels were significantly upregulated, while glutathione levels were significantly decreased in the AA group. In contrast, MoC and SS treatments reduced interleukin-1β, malondialdehyde levels, myeloperoxidase activity, bax/bcl-2 ratio, and caspase-9 and caspase-3 levels. Glutathione levels increased upon MoC or SS treatment. Increased macroscopic and microscopic scoring with AA improved with MoC or SS treatment, but the MoC or SS treated groups had higher score values than the control. Also, in vitro results showed that MoC exhibited 2,2-diphenyl-1- picrylhydrazyl and 2,2\"-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid radical scavenging activity as well as significant antilipoxygenase activity. In addition, MoC extract showed a potent anti-inflammatory activity compared to standard indomethacin. Conclusion: Our biochemical, in vitro and histopathologic analysis indicate that MoC is likely to prove beneficial in UC therapy.

    Platanus orientalis (Plane Tree) Extract Protects Against Hyperoxaluria Induced Kidney Damage

    The aim of this study is to determine whether Platanus orientalis (PO) which has anti-inflammatory, antioxidant and diuretic properties and used in the treatment of kidney stones as traditional folk medicine, will reduce or prevent the stone formation in the urinary system. To simulate the urolithiasis model 0.75% ethylene glycol (EG) and 1% ammonium chloride (AC) were applied to Sprague-Dawley rats. The rats were divided into four groups (n=8). The control group was given standard drinking water for 5 weeks. EG group received 0.75% EG in their drinking water containing 0.75% EG and 1% AC. PO extract (100 mg/kg) was given orally for 5 weeks to the preventive group and for last 2 weeks to the therapeutic group, respectively. At the end of experiment, 24-hour urine and kidney samples were obtained. In urine samples, calcium and citrate levels were decreased and oxalate level was increased in the EG group. In kidney samples myeloperoxidase, caspase-3, N-acetyl-β-glycosaminidase (NAG) activities, malondialdehyde (MDA), 8-hydroxy-2-deoxyguanosine (8-OHdG), tumor necrosis factor-α and interleukin-1β levels were increased while superoxide dismutase activity and glutathione levels were decreased in the EG group. These biochemical parameters returned to control levels in both PO treatment groups. Histological findings also correlate with these results. Our findings are suggested that PO treatments is effective in both preventive and therapeutic groups.
