    Alpha-actinin-3 R577X Polymorphism Profile of Turkish Professional Hip-Hop and Latin Dancers

    Actins are small globular filaments functioning in cell processes like muscle contraction, and stabilized to the sarcomeric Z-discs by actin binding proteins (actinins). One of the important gene coding for actin binding proteins in fast twitch fibers is alpha-actinin-3 (ACTN3). In this research, we have conducted a gene profile study investigating the genotype and allele distributions of ACTN3 R577X polymorphism in Turkish professional hip-hop and latin dancers and compared them to non-dancers as a control group. 30 professional dancers and non-dancers were recruited for the study. A genotyping procedure was carried out by a newly introduced four-primer PCR methodology. For statistical analysis, the Chi-square test was used to compare data between the groups (p<0,05 evaluated as significant). Numbers and the percentages of dancers were 2 (7%), 21 (70%) and 7(23%) for RR, RX and XX genotypes, respectively. The same numbers and the percentages were 15 (50%), 8 (15%) and 7 (23%) for RR, RX and XX genotypes, respectively, for the controls. Allele numbers and percentages of the R allele were 38 (63%) and 25 (42%) for the control and dancers, respectively. For X allele, the respective number for control and dancers were 22 (37%) and 35 (58%). No significant difference was detected between the groups in the terms of genotypes and alleles. Only RX genotype was significantly different between dancers and non-dancers (p<0,05). The results of the present and the first study, associating professional dancers and ACTN3 R577X polymorphism, is suggesting that RX genotype may have an genetic advantage for the physical predisposition for dancing, at least in the terms of ACTN3 R577X polymorphism. Prospective studies with extended numbers of dancers focusing on the influence of ACTN3 R577X polymorphism are required for confirmation of these findings.

    Distribution of alpha-actinin-3 rs1815739 and angiotensin-1 converting enzyme InDel polymorphisms in Turkish bodybuilders

    The determination of the genetic profiles of successful athletes and the effects of these genetic parameters on athletic performance is gaining increasing interest. The majority of studies assessing the genetics of athletes usually analyse the most well-known genetic variations in athletes associated with the different specialties. The aim of the present study was to analyse the ACE InDel and ACTN3 rs1815739 polymorphisms in Turkish bodybuilders. A total of 11 male bodybuilders were recruited and genotyped for these polymorphisms. The respective percentage of the ACE II, ID and DD genotypes were 18, 73 and 9. For the ACTN3 genotype, the respective frequencies were 55 and 45 for the RX and RR genotypes. No XX genotype was detected. The allelic counts were 12 (55%) for I and 10 (45%) for the D alleles of ACE; and 12 (55%) and 10 (45%) for R and X alleles, respectively, for the ACTN3 genotype. Additionally, 5 athletes had ID + RX genotypes in terms of ACE InDel and ACTN3 rs1815739 polymorphisms, respectively. These results indicate the importance of endurance related alleles of ACE and ACTN3 in bodybuilders. The results of the present are in agreement with previous studies, highlighting a potential association between specific polymorphisms and the endurance-related nature of bodybuilders. Further studies with larger cohorts are required to understand the association between these polymorphisms and specific parameters performance in bodybuilders.

    Preliminary findings of the distribution of alpha-2-adrenergic receptor (ADRA2A) rs1800544 polymorphisms in kickboxing players

    Objective: Adrenergic receptors play an important role in providing neurotransmitter release from the sympathetic nervous system and adrenergic neurons. The rs1800544 polymorphism in 3’-untranslated region of Alpha-2-Adrenergic Receptor (ADRA2A) is if caused by C>G tranversion at position –1291. In this study, we objective to analyze the rs1800544 distribution in kickboxing players. Methods: A total of 12 male players and 101 sedentary individuals (as a control group) participated in the study. Following DNA isolation, rs1800544 polymorphism of ADRA2A gene was determined by real-time polymerase chain reaction (Rt-PCR). Statistical analysis was carried out by chi-square test. Results: CC, CG and GG genotypes and percentages for rs1800544 polymorphism were analyzed as 5 (41,7%), 4 (33,3%) and 3 (25%), respectively. C allele was counted as 14 (58,3%) and G allele as 10 (41,7%). In the control group CC, CG and GG were found as 34 (33,6%), 53 (52,5%), 14(13,9%), respectively. C allele was counted as 121 (60%) and G allele as 81 (40%). When we compare athletes and controls in the terms of genotypes, we detected no statistically significant difference (p>0,3940). For the allele comparison, we detected no significant difference between groups (p>0,0871). Conclusion: Determination of gene combinations in athletes is of great importance for determination of the effect of genetic factors in athletic performance. In our cohort, CC genotype was more frequently found, and C allele was also higher in players. We think that the effect of ADR2A2 gene on athletic performance might be determined more clearly by the fact that this study is an example of similar studies involving higher numbers of subjects.
