Çalışmamızın amacı;VDRveMMP3genlerinde meydana gelen gen polimorfizmlerin etkilerinin değerlendirilip çürük risk modellemesi üzerine olan etkilerinin incelenmesidir.Gereç ve Yöntem:Çalışma populasyonu, fakülte hastanesine başvuran 20-44 yaş aralığındaki erkek bireylerde rutin ağız içi muayenesi sonrasında çürük, eksik ve restorasyonlu dişler (DMFT) indeksine göre tanı konulan ‘yüksek çürük risk’(DMFT ≥ 14) ve düşük çürük risk’(DMFT£5) olarak 2 gruptan oluşturulmuştur (n=160). Detaylı anamnez alınan katılımcılardan plak indeksi, gingival indeks sondalamada kanama, sondalama derinliği, klinik ataşman seviyesi ve tükürük tamponlama kapasitesi, tükürük streptekok mutans (SM) ve laktobasil (LB) sayısı ölçülmüştür.Katılımcılardan alınan kan örneklerinden DNA izolasyonları sonrasında,MMP3(rs679620)veVDR(rs731236)genotiplemesi ise Real-time PZR tekniği kullanılarak belirlenmiştir.Elde edilen sonuçlar istatistiksel yöntemler kullanılarakanlamlılık düzeyi p<0,05’te değerlendirilmiştir.Bulgular:Gruplar arası plak indeksi, gingival indeks, sondalama derinliği, klinik ataşman seviyesi, sondalamada kanama, tükürük akış hızı, tükürük tamponlama kapasitesi ve tükürük SM ve LB sayısı faktörleri arasında anlamlı düzeyde fark bulunmuştur (p<0,001). Çevresel risk faktörlerinde sosyoekonomik durum, diş fırçalama sıklığı, koruyucu diş tedavisine yönelik uygulama durumları açısından gruplar arasında anlamlı düzeyde fark tespit edilmiştir (p<0,001).MMP3rs679620 polimorfizminin de çürük risk belirlemesinde etkili olduğu (p<0,001);VDRpolimorfizminin ise etkili olmadığı (p=0,862) tespit edilmiştir.DMFT’ ye etki eden değişkenlerin incelenmesinde Adımsal Regresyon Analizi yapılarak model 7 adımda incelenmiştir ve modeller istatistiksel olarak anlamlı bulunmuştur (p<0,001).Çevresel risk faktörü olarak plak indeksi, sondalamada kanama, şekerli ara öğün tüketim sıklığı, tükürük tamponlaması,MMP3rs679620 gen polimorfizminin içerenModel 5 DMFT’ nin %58’ini açıklamıştır.Sonuç:Çürük risk modelinde; plak indeksi, sondalamada kanama, şekerli ara öğün tüketim sıklığı, tükürük tamponlaması,MMP3rs679620 gen polimorfizminin çevresel risk faktörü olarak modelde yer alması, bireylerin risk gruplarının belirlenmesinde etkili olacaktır.Anahtar Kelimeler:Çürük risk değerlendirmesi, gen-çevre etkileşimi,MMP3, Polimorfizm, VDR.Publication Metadata only The investigation of genetic polymorphisms in the carbonic anhydrase VI gene exon 2 and salivary parameters in type 2 diabetic patients and healthy adults(SPRINGER, 2012) YARAT, AYŞEN; Ozturk, Leyla Koc; Ulucan, Korkut; Akyuz, Serap; Furuncuoglu, Halit; Bayer, Hikmet; Yarat, AysenThe aim of this study was to investigate carbonic anhydrase (CA) VI Exon 2 single nucleotide polymorphism (SNP) and its possible association with salivary parameters in type 2 diabetic patients compared to healthy adults. Caries status was measured by using the DMFT (number of decayed, missing, and filled teeth) index. Unstimulated whole saliva and blood samples were taken. SNPs of CA gene exon 2 were determined by PCR and DNA sequencing. Salivary CA activity and buffering capacity were determined by the method of Verpoorte and Ericson, respectively. Furthermore, salivary pH was measured with pH paper and salivary flow rate was calculated. Salivary buffering capacity and pH were significantly lower in diabetic patients than those of healthy subjects (P < 0.05). Salivary flow rate, CA activity and DMFT levels did not differ between groups (P > 0.05). Four SNPs were detected; their pubmed database number are rs2274327 (C/T), rs2274328 (A/C), rs2274329 (G/C) and rs2274330. While first three of those were responsible for amino acid changes, the last one was not. The frequencies of SNPs were not significant between groups (P > 0.05). Positive significant correlation was found between CA activity and the frequency of SNPs. There was no correlation between the SNPs frequencies and pH or buffering capacity. SNPs found in this study may be related to salivary CA activity in diabetics.Publication Metadata only A pilot study for determination of anxiety related SLC6A4 promoter S and L alleles in healthy Turkish athletes(C M B ASSOC, 2017) ULUCAN, KORKUT; Corak, A.; Kapici, S.; Sercan, C.; Akkoc, O.; Ulucan, K.We aimed to analyze the allelic distribution of solute carrier family-6 member-4 promoter region in Turkish athletes. Recent studies showed the association of lesser expressing S allele with anxiety. Genotype percentages for LL, LS and SS genotypes were found as 46, 35 and 19, respectively. 38% of the males had LL, %38 had LS and 24% had SS genotypes. Percentages of LL, LS and SS genotypes were 54, 31 and 15 in females, respectively. 15 (58%) male and 18 (69%) females had L, 11 (42%) male and 8 (31%) females had S alleles. Variations in the association of the SLC6A4 alleles with neuropsychiatric disorders according to different nationalities have been reported. This is the first report showing that LL genotype and L allele in Turkish athletes was more frequent than SS genotype and S allele.Publication Open Access Obesity is associated with IL-6 gene polymorphisms rs1800795 and rs1800796 but not SOCS3 rs4969170(2023-03-01) ULUCAN, KORKUT; Koc G., Doran T., Uygur M. M., Kirac D.Background: An imbalance of inflammatory factors can stimulate obesity by inducing chronic inflammation in adipose tissue. Interleukin-6 (IL-6) is a cytokine with both inflammatory and anti-inflammatory functions. Suppressor of cytokine signaling 3 (SOCS3) acts as an inhibitor for a number of cytokine signals. The IL-6 and SOCS3 genes are known to be involved in lipid and energy metabolism, although it is unclear how these genes relate to obesity. The aim of this study is to determine whether the obesity risk is associated with the IL-6 (rs1800795, rs1800796) and SOCS3 (rs4969170) gene polymorphisms. Methods and results: Based on their body mass index (BMI) scores, 185 people were determined, of whom 90 were from the control group and 95 were obese. Anthropometric measurements and biochemical parameters of the study subjects were documented during the examination. Genomic DNA isolation was performed from the blood samples of all participants. IL-6 (rs1800795, rs1800796) and SOCS3 (rs4969170) polymorphisms were detected by real-time quantitative polymerase chain reaction (qRT-PCR) from genomic DNA samples. The IL-6 rs1800795 and rs1800796 variants showed a significant difference between the control and obese groups (p = 0.027; p = 0.013). The SOCS3 rs4969170 variation did not substantially differ between the control and obese groups (p = 0.825). Conclusion: In our study, IL-6 rs1800795(G/C) and rs1800796(G/C) polymorphisms appeared to be a risk factor for obesity. The C allele was associated with the obesity phenotypes. However, the SOCS3 rs4969170 (A/G) polymorphism was not linked to an increased risk of obesity. IL-6 polymorphisms may be new targets for obesity treatment.Publication Open Access Sprint and Anaerobic Power with the Soccer-Specific ACTN3 Gene: A Distintive Example(2024-03-01) TACAL ASLAN, BESTE; POLAT, TOLGA; YILMAZ, ÖZLEM ÖZGE; ULUCAN, KORKUT; Söyler M., Zileli R., Özkamçı H., Diker G., Sever M., Bayrakdaroglu S., Ön S., Can İ., Ulucan K., Tacal Aslan B., et al.The aim of this study is twofold: (1) to identify differences in certain anaerobic parameters (10m sprint, 30m sprint, anaerobic power, and Illinois agility tests) between professional and amateur soccer players, and (2) to determine whether there is a difference in theACTN3gene polymorphism between professional and amateur soccer players. Ultimately, the goal is to reveal which parameters contribute to the differentiation in these two aspects. A total of 133 volunteer soccer players, including 71 professionals and 62 amateurs, participated in the research. DNA extraction from buccal epithelial cells was performed using a commercial kit to determine the genetic background of the athletes, and Real-Time PCR was conducted for genotyping. Statistical analysis of the findings obtained from the test results was performed using the SPSS 23 (SPSS Inc., Chicago, IL, USA) package program. The homogeneity of variance of the data was assessed using the Levene Test, and normal distribution analyses were conducted using the Shapiro-Wilk Test. Chi-square and Mann-Whitney U tests were employed for parameter analysis. The significance level was set at p<0.05. Evaluation of the data in our study revealed no statistically significant difference inACTN3rs1815739 gene polymorphism between the groups (p>0.05). However, there is a statistically significant difference in anaerobic parameters (10m sprint, 30m sprint, and anaerobic power) except for the Illinois test (p<0.05). Ş., ULUCAN K.Objective: Adrenergic receptors play an important role in providing neurotransmitter release from the sympathetic nervous system and adrenergic neurons. The rs1800544 polymorphism in 3’-untranslated region of Alpha-2-Adrenergic Receptor (ADRA2A) is if caused by C>G tranversion at position –1291. In this study, we objective to analyze the rs1800544 distribution in kickboxing players. Methods: A total of 12 male players and 101 sedentary individuals (as a control group) participated in the study. Following DNA isolation, rs1800544 polymorphism of ADRA2A gene was determined by real-time polymerase chain reaction (Rt-PCR). Statistical analysis was carried out by chi-square test. Results: CC, CG and GG genotypes and percentages for rs1800544 polymorphism were analyzed as 5 (41,7%), 4 (33,3%) and 3 (25%), respectively. C allele was counted as 14 (58,3%) and G allele as 10 (41,7%). In the control group CC, CG and GG were found as 34 (33,6%), 53 (52,5%), 14(13,9%), respectively. C allele was counted as 121 (60%) and G allele as 81 (40%). When we compare athletes and controls in the terms of genotypes, we detected no statistically significant difference (p>0,3940). For the allele comparison, we detected no significant difference between groups (p>0,0871). Conclusion: Determination of gene combinations in athletes is of great importance for determination of the effect of genetic factors in athletic performance. In our cohort, CC genotype was more frequently found, and C allele was also higher in players. We think that the effect of ADR2A2 gene on athletic performance might be determined more clearly by the fact that this study is an example of similar studies involving higher numbers of subjects.Publication Open Access Single nucleotide polymorphisms in IL-1A RS1800587, IL-1B RS1143634 and vitamin D receptor rs731236 in stage III grade B/C periodontitis(2022-06-01) ÖZTÜRK ÖZENER, HAFİZE; ASLAN, BEGÜM; AĞRALI, ÖMER BİRKAN; ULUCAN, KORKUT; YILDIRIM, HATİCE SELİN; KURU, LEYLA; Ozener H., ASLAN B., Eken B. C., ULUCAN K., KURU L.The purpose of the study is to determine the prevalence of interleukin (IL)-1A (rs1800587), IL-1B (rs1143634) and vitamin D receptor (VDR) (TaqI, rs731236) gene polymorphisms in the Turkish population and their association with Stage III Grade B/C periodontitis. Systemically and periodontally healthy individuals (N = 100) and Stage III Grade B/C periodontitis patients (N=100) based on clinical and radiographic examination were included in this research. Clinical attachment level, probing depth, bleeding on probing, plaque and gingival indices of the subjects were measured. Genotyping of IL-1A (rs1800587), IL-1B (rs1143634) and VDR (rs731236) polymorphisms was conducted by Real Time PCR. Allelic and genotypic distributions of IL-1A (rs1800587) gene polymorphism were not associated with periodontitis (p > 0.05). In IL-1B (rs1143634) gene polymorphism, the C allele was detected more frequently in healthy individuals compared with the periodontitis patients (p=0.045). CC genotype and C allele in VDR (rs731236) gene polymorphism was higher in periodontitis patients (p=0.031, p=0.034, respectively). In comparison with Grade B periodontitis patients and healthy subjects, CC genotype and C allele were observed more frequently in the Grade B periodontitis in terms of alleles (C/T) and genotypes for VDR (rs731236) polymorphism (p=0.024, p=0.008, respectively). This study presents that the VDR (rs731236) polymorphism are associated with enhanced susceptibility to Stage III periodontitis in the Turkish population. Furthermore, VDR (rs731236) polymorphism may be used as an identification criteria to discriminate Grade B and Grade C in Stage III periodontitis.Publication Open Access Evaluation of Association between PPARGC1A Gene Polymorphism and Competitive Performance of Elite Athletes(2022-10-01) ULUCAN, KORKUT; Bulğay C., Zorba E., Akman O., Bayraktar I., Kazan H. H. , Ergün M. A. , Ulucan K.PPARGC1A (rs8192678) gen polimorfizmi ve atletik performans arasıdaki ilişkiler konusunda sınırlı sayıda araştırma mevcuttur. Bu nedenle, bu çalışmada PPARGC1A genindeki rs8192678 polimorfizminin Türk elit atletizm sporcularının yarışma performansları ile olası ilişkisin araştırılması amaçlamaktadır. Çalışmaya 18-35 yaşları arasında toplam 60 elit sporcu (31 sprint/güç ve 29 dayanıklılık) ve 20 kontrol/sedanter gönüllü olarak katıldı. Atletlerin kişisel en iyi (PB)/yarışma performans düzeylerini belirlemek için Uluslararası Atletizm Federasyonları Birliği (IAAF) puan ölçeği kullanıldı. Tüm ekzom dizilimi (WES), katılımcıların kanından izole edilen genomik DNA tarafından gerçekleştirildi. rs8192678 polimorfizmi ile PB arasındaki ilişki, cinsiyet ve spor deneyimi ayarlaması ile tek yönlü kovaryans analizi (ANCOVA) ile incelendi. Bulgulara göre grup içi ve gruplar arasında, G/G, G/A ve A/A genotip dağılımları arasında anlamlı bir farklılık tespit edilmemiştir (p>0.05). Sonuç olarak, PPARGC1A rs8192678 polimorfizmi ile sporcuların performansı arasında anlamlı hiçbir ilişki tespit edilmemiştir. Ancak, yapılan araştırma hakkında daha net bilgi verebilmek için daha fazla katılımcı ile yapılması önerilir.
