Publication Open Access Vitamin D receptor and estrogen receptor gene polymorphisms in men with type 2 diabetes: Effects on Bone Metabolism(2022-12-01) ÜSTAY, ÖZLEM; YAVUZ, DİLEK; YÜKSEL, MERAL; YAVUZ D., YÜKSEL M., Sancak S., Yazici D., ÜSTAY Ö., Deyneli O., Akalin S.Purpose There is an increased fracture risk in type 2 diabetes mellitus [DM] patients independent of bone mineral density [BMD], both in men and women. Estrogen receptor [ER]-alpha and vitamin D receptor [VDR] gene polymorphisms may predispose patients to increased osteoporosis and fracture risk. This study aims to analyze the relationship of the ER-alpha gene and VDR gene polymorphisms with indicators of bone turnover and BMD in male type 2 diabetic patients. Methods Type 2 diabetic men diagnosed with diabetes for at least one year and healthy controls were included in this cross-sectional study. BMD was measured by dual X ray absorptiometry. Gene polymorphisms were evaluated with polymerase chain reaction-restriction length polymorphism. Serum iPTH, calcium, beta-CrossLaps (cTx), osteocalcin, and free testosterone levels were also evaluated. Results Participants were 141 type 2 diabetic men [55 +/- 8 years] and 100 healthy controls [53 +/- 7 years]. BMD measurements were not statistically different between the groups. While iPTH [p < 0.05] and serum calcium levels [p = 0.03] were higher in men with type 2 DM; beta-CrossLaps [p = 0.0001], osteocalcin [p = 0.005], and free testosterone [p = 0.04] were lower than controls. The differences in terms of the frequencies of VDR Apa, Taq, Bsm, Fok and ER-alpha polymorphisms were not statistically significant between the groups. No relationship was observed between polymorphisms and BMD in both groups. Yüksek anksiyete seviyeleri bellek fonksiyonlarını olumsuz etkilemektedir.2Amaç: İstemli tekerlek çevirme egzersizi, direnç egzersizi ve kombine egzersizuygulamalarının anksiyete düzeyleri ile bellek üzerindeki etkilerinin araştırılması ve alttayatan mekanizmaların ortaya konulması amaçlanmıştır.Yöntem: Çalışmada Wistar Albino erkek sıçanlarda (n=50) taklit cerrahi ve Parkinsongrupları oluşturulduktan sonra sedanter ve 3 farklı egzersiz protokolünün uygulandığı istemli(İE), rezistans (RE) ve kombine (KE: İE+RE) grupları oluşturuldu. Egzersizler 6 haftaboyunca (3 gün/hafta) uygulandı. PH modeli oluşturmak için sıçanlarda sağ mediyal önbeyine 6-OHDA (0.5 μl/dk) enjeksiyonu yapıldı. Taklit cerrahili gruplara 6-OHDA’nınçözücüsü verildi. Apomorfin uygulamaları sonrası rotasyon hareketi ile Parkinson modelideğerlendirildi. Obje tanıma testi ve delikli levha testleri sıçanlarda bellek fonksiyonlarını veanksiyete düzeylerini değerlendirmek için yaptırıldı. Beyin dokusunda antioksidan glutatyon(GSH) ve lipid peroksidasyonu belirteci malondialdehit (MDA) ve nötrofil infiltrasyonugöstergeci miyeloperoksidaz (MPO) aktivitesi, oksidan radikallerin belirteci luminol velusigenin ölçüldü. Verilerin analizinde tek yönlü ANOVA ardından Tukey-Kramer testi ilestudent’s t testi kullanıldı. Bulgular: Her 3 egzersiz ile Parkinsona bağlı gerileyen bellek fonksiyonu düzelmiştir(p<0.05-0.01). Sedantere kıyasla PH oluşturulmuş KE grubunda rotasyon hareketi azalırken,İE grubunda arttı (p<0.05-0.01). PH oluşturulmasıyla luminol ve lusigenin düzeyleri artarken,İE ile luminol azalmıştır (p<0.05-0.001). MPO aktivitesinin PH oluşturulmasıyla sedantergrubunda yükseldiği, ancak İE ve RE gruplarında baskılandığı gözlenmiştir (p<0.01-0.001).Her 3 egzersiz ile GSH düzeylerinin arttığı (p<0.05-0.01), KE ile MDA düzeylerinin düştüğübulundu (p<0.05). E. , YÜKSEL M., Cumbul A., ERCAN F., CABADAK H., AYDIN OMAY B., Alican İ.BACKGROUND: Previous studies found metformin as an effective agent to suppress oxidative stress, inflammation, and apoptosis in various inflammatory diseases. The present study investigated the effect of metformin against 2 experimental gastric injury models in rats, using macroscopical, histopathological, biochemical, and immunostaining studies. METHODS: After 24 hours of fasting, male Sprague-Dawley rats (280-400 g) (n = 8 per group) received indomethacin (80 mg/kg; indo ulcer group) or absolute ethanol (5 mL/kg; ethanol ulcer group) or vehicle orally by gavage. Metformin (500 mg/kg) was given orally for 3 days prior to indomethacin or ethanol challenge. Ranitidine (50 mg/kg) was given orally for 3 days before indomethacin or ethanol administration as a positive control. On day 3, the animals were euthanized 6 hours after indo or 1 hour after ethanol challenge. Gastric samples were used for macroscopic scoring, histopathological examinations, and biochemical assays. Trunk blood was collected for the assessment of interleukin-1β level. RESULTS: In both ethanol ulcer and indo ulcer groups, metformin decreased the extent of gastric lesions macroscopically and microscopically, improved the high chemiluminescence levels, and the percentage of terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL)-positive apoptotic cells compared with untreated ulcer groups. Gastric blood flow analysis revealed significant increases in both metformin-treated ulcer groups compared to untreated ulcer groups. This study, it was aimed to examine the possible anti-inflammatory, antioxidant, and neuroprotective effects of apigenin in the setting of the mild traumatic brain injury (TBI) model. Methods Wistar albino male rats were randomly assigned to groups: control (n = 9), TBI (n = 9), TBI + vehicle (n = 8), and TBI + apigenin (20 and 40 mg/kg, immediately after trauma; n = 6 and n = 7). TBI was performed by dropping a 300 g weight from a height of 1 m onto the skull under anesthesia. Neurological examination and tail suspension tests were applied before and 24 h after trauma, as well as Y-maze and object recognition tests, after that rats were decapitated. In brain tissue, luminol- and lucigenin-enhanced chemiluminescence levels and cytokine ELISA levels were measured. Histological damage was scored. Data were analyzed with one-way ANOVA. Results After TBI, luminol (p < .001) and lucigenin (p < .001) levels increased, and luminol and lucigenin levels decreased with apigenin treatments (p < .01-.001). The tail suspension test score increased with trauma (p < .01). According to the pre-traumatic values, the number of entrances to the arms (p < .01) in the Y-maze decreased after trauma (p < .01). In the object recognition test, discrimination (p < .05) and recognition indexes (p < .05) decreased with trauma. There was no significant difference among trauma apigenin groups in behavioral tests. Interleukin (IL)-10 levels, one of the anti-inflammatory cytokines, decreased with trauma (p < .05), and increased with 20 and 40 mg apigenin treatment (p < .001 and p < .01, respectively). The histological damage score in the cortex was decreased in the apigenin 20 mg treatment group significantly (p < .05), but the decrease observed in the apigenin 40 mg group was not significant. Conclusion The results of this study revealed that apigenin 20 and 40 mg treatment may have neuroprotective effects in mild TBI via decreasing the level of luminol and lucigenin and increasing the IL-10 levels. Materials and Methods: Wistar-albino rats were given (n=32) melatonin (4 mg/kg/day), antibiotic mixture (AB; 1g/L ampicillin + 1g/L neomycin + 1g/L metronidazole), melatonin+AB in drinking water for 12 days or tap water for 15 days (control group; n=8). Subsequently, ulcer was induced. All treatments were continued for three days. Gastric tissues were obtained for biochemical and histopathological examinations, and fecal samples from the rectum were stored for bacteriological measurements. Results: MPO and MDA levels were increased in untreated ulcer groups compared to the control group. In addition, the levels of luminol-lucigenin chemiluminescence (CL) and 8-OHdG and TNF-alpha and IL-8 protein expressions were also increased, while TNF-alpha, IL-8, MDA, 8-OHdG, luminol and lucigenin CL levels were significantly decreased in the melatonin-treated ulcer groups. However, melatonin+AB pretreatment increased antioxidant GSH levels and anti-inflammatory IL-10 levels, and suppressed caspase-3 activity and reduced MPO back to control level. Conclusion: We anticipate that melatonin treatment, which is an effective antioxidant and radical scavenger, can accelerate ulcer healing along with antibiotics and increase the variety of bacteria impaired by antibiotics in the colon.Publication Open Access Cerrahi menopoz oluşturulmuş sıçanların karaciğer ve böbrek dokularında oksidan/antioksidan dengenin korunmasında egzersizin ve östrojenin yararlı etkileri(2022-09-01) YÜKSEL, MERAL; ERCAN, FERİHA; YILDIRIM, ALPER; YEGEN, BERRAK; Tamer S. A. , Levent N., Yüksel M., Ercan F., Yıldırım A., Yegen B.Amaç: Bu çalışmanın amacı cerrahi olarak menopoz oluşturulan sıçanların böbrek ve karaciğerlerinde gözlenen histopatolojik ve fonksiyonel değişiklikleri ve östrojen veya egzersizin ya da östrojen-egzersiz kombinasyonunun oksidan hasar üzerine etkilerini araştırmaktır.Materyal ve Metot: Anestezi altında Sprague Dawley dişi sıçanlara (n=32) bilateral overiektomi uygulandı ve tüm sıçanlar rastgele olarak iki gruba ayrıldı. Sıçanların yarısına normal içme suyu, diğer yarısının içme sularına östrojen (1mg/kg/gün) eklendi. İki hafta sonra gruplar kendi içlerinde sedanter ve egzersiz (5 gün/hafta, 30 daki-ka, 8 hafta) gruplarına ayrıldı. Deney protokolünün sonun-da serum, karaciğer ve böbrek örnekleri biyokimyasal ve histopatolojik incelemeler için alındı. Femurda da histopa-tolojik değerlendirme yapıldı.Bulgular: Cerrahi olarak menopoz oluşturulan sıçan-larda östrojenin böbrek dokusunda nötrofil infiltrasyonunu ve reaktif oksijen türlerinin üretimini baskılayarak koruyu-cu etki gösterdiği, kemik kütlesinde hafif düzeyde artışa neden olduğu, ancak karaciğerin antioksidan glutatyon düzeyinde azalmaya yol açtığı belirlenmiştir. Buna karşın, östrojen uygulaması menopozda yapılan egzersiz nedeniy-le karaciğerde oluşan oksidan stresi engellemiştir. Egzer-sizle veya egzersize östrojen tedavisinin eklenmesiyle böbrek fonksiyonları önemli ölçüde etkilenmezken, kemik yapısında tek başına östrojene kıyasla daha olumlu deği-şiklikler gözlenmiştir.Sonuç: Östrojen replasmanı kemik dokusundaki olum-lu etkilerinin yanı sıra karaciğer ve böbrekte oksidan stresi azaltmakta ve özellikle karaciğerde egzersize bağlı gelişen oksidan stresi baskılayarak koruyucu etki göstermektedir.Publication Open Access Presepsin Levels in Experimental Sepsis and Massive Bowel Resection Models in Rats(2020) YÜKSEL, MERAL; Unay-Demirel, Ozlem; Ignak, Seyda; Orug, Taner; Yuksel, MeralPublication Metadata only The protective role of UMCA on bleomycin-induced lung fibrosis in rats(2022-11-05) AYDEMİR, SEZGİN; ÖZKAN YENAL, NAZİYE; GÜÇLÜ, HÜLYA; YÜKSEL, MERAL; ÖZBEYLİ, DİLEK; AYDEMİR S., ÖZKAN YENAL N., GÜÇLÜ H., YÜKSEL M., ÖZBEYLİ D., ERDOĞAN N.Bleomycin (BLM), an antibiotic drug, is used for the treatment of cancers like Hodgkin’s lymphoma, Kaposi’s sarcoma and cervical cancer. The most common adverse effects occurred during BLM treatment is lung toxicity, which manifest as fibrosis. UMCA® (Pelargonium sidoides root extract) is a commercial product to treat acute and chronic upper respiratory tract infections. UMCA® has antiviral, antibacterial, immunomodulatory and cytoprotective properties, which is related to its polyphenol, coumarin, anthocyanidin and flavonoid contents. In the present study, we aimed to investigate the possible protective effects of UMCA® on BLM induced lung fibrosis in rats. Wistar albino rats was included in this study and randomly divided into 4 groups (Control, BLM, BLM+UMCA and UMCA). The control group received physiological saline. UMCA® was orally applied at a dose of 200 mg/kg/day and single intratracheal administration of BLM was conducted at a dose of 10 mg/kg. All animals were decapitated after 10 days and lung tissues was removed for the biochemical and histopathological examinations. Malondialdehyde (MDA) and glutathione (GSH) levels and Myeloperoxidase (MPO) activity were determined. Chemiluminescence (CL) method using luminol and lucigenin probes, additional nitric oxide and peroxynitrite measurements were applied. Histopathological observations were analyzed with H&E and Gomori's one-step trichrome staining. Collagen contents of lung tissue were also determined with a spectrophotometric method in paraffin-embedded tissues. BLM elevated MDA levels and MPO activity and depleted GSH levels in the lung tissues (p<0.001). CL measurement levels were also increased in BLM group respect to the control. Contrary to this, UMCA® treatment reversed these effects, significantly (p<0.001). BLM caused alveolar structural disturbance and inflammatory cell infiltrations and collagen content was also significantly increased in BLM group compared to the control (65.7±10.6, p<0.001). UMCA® administration reduced degenerations and decreased collagen content in the lung tissue. In conclusion, UMCA® has an antioxidant and protective effects on BLM induced lung fibrosis in rats.
