This study aims to investigate the possible effects of myrtle leaf extract (Myrtus communis L.) on, heart, kidney and aorta tissues in the experimental renovascular hypertension (RVH). 32 male Wistar Albino rats were divided into four groups as control, hypertension, hypertension+ramipril, and hypertension+myrtle leaf extract treatment groups. RVH model was induced by Goldblatt\"s 2-kidney 1-clip method. 12 weeks later than the treatment blood pressures were recorded and oxidant and antioxidant parameters, tissue factor activity, and histological analysis were determined in the kidney, heart, and aortic tissues. The blood pressure levels of the hypertension group significantly increased compared to the control group. Administration of myrtle leaf extract and ramipril significantly decreased the increased blood pressure. In the hypertension group, oxidative damage increased in the kidney, heart, and aorta tissues. In the histological evaluation of tissues in RVH, heart muscle fibres degenerated. Bowman capsule and glomeruli dilated and tubules damaged in the kidney. Myrtle leaf extract administration regenerated the damages and degenerations. The administration of myrtle leaf extract restored the impaired oxidant-antioxidant balance in the heart, kidney and aorta tissues of hypertensive rats. Myrtle leaf extract can be considered as an alternative antihypertensive treatment target in the prevention of oxidative stress-induced damage in renovascular hypertension.Publication Open Access The effect of Cotinus coggygria L. ethanol extract in the treatment of burn wounds(2022-01-01) OKUYAN, BETÜL; ŞEN, ALİ; ŞENER, GÖKSEL; ERCAN, FERİHA; Erta B., OKUYAN B., ŞEN A., ERCAN F., Onel H., GÖGER F., Sener G.The overall aim of the present research is to evaluate for the first time the curative effect of Cotinus coggygria leaves on burn injury in an experimental burn model along with its anti-inflammatory and antioxidant activity potential. Also, phenolic compounds of C. coggygria were characterised by LC-MS/MS. Wistar albino rats weighing 200-250 g were exposed to 90 degrees C bath for 10 s to induce burn injury, involving 30% of the total body surface area. In the treatment groups, 5% C. coggygria ethanol extract was applied topically as a cream immediately after the burn. Blood and skin tissue samples were taken after decapitation at the 4th and 48th hours following the burn procedure. Interleukin 1-beta (IL-1 beta) and tumour necrosis factor (TNF-alpha) were determined in serum samples, and hydroxyproline, prostoglandin E2 (PGE2), and myeloperoxidase (MPO) activity and 8-hydroxy-2\"-deoxy-guanosine (8-OHdG) levels were determined in skin tissue samples. Increased levels of serum cytokines were decreased with C. coggygria treatment in both periods. MPO activity, prostaglandine (PGE2), and 8-OhdG levels increased, while hydroxyproline levels decreased due to burn damage. On the other hand, these parameters were returned to its normal levels with C. coggygria treatment. In addition, the tissue histology of animals treated with C. coggygria showed a complete epithelialization with increased collagenation. As a result, C. coggygria may be an alternative treatment approach for burns-induced skin damage and wounds.Publication Open Access The effects of Urtica dioica L. ethanolic extract against urinary calculi in rats(MARMARA UNIV, 2020-03-12) DOĞAN, AHMET; Keles, Rumeysa; Sen, Ali; Ertas, Busra; Kayali, Damla; Eker, Pinar; Sener, Tarik Emre; Dogan, Ahmet; Cetinel, Sule; Sener, GokselNephrolithiasis is common urological problem and stone formation has multiple underlying pathogenetic factors. We investigated the possible preventive and therapeutic effect of Urtica dioica ethanol extract (UD) on ethylene glycol-induced nephrolithiasis model in rats. Sprague-Daw ley rats were divided into lour groups (n = 10). The control group was given normal drinking water for 8 weeks and was administered vehicle by gastric gavage. Stone formation was induced by adding 0.75% ethylene glycol (EG) to their drinking water. UD (700 mg/kg) was given orally lor 8 weeks to the preventive group and I or last 4 weeks to the treatment respectively. At the end of the experiment, urine, blood samples and kidney tissues were obtained. In 24-hour urine samples, calcium and citrate levels were decreased and oxalate levels were increased in EG whereas LID treatment groups reversed these parameters back to control levels. In addition, serum levels of creatinine and urea were increased in EG while LID significantly reduced these parameters. Malondialdehyde, 8-hydroxydeoxyguanosine and tumor necrosis alpha levels, and caspase- 3 and N-acetyl-beta-glucosaminidase activities were elevated in EG group and showed a decrease in LID treated groups. Glutathione level was decreased in EG group, whereas it was increased in UD preventive group. Histological examination showed an improvement in UD treated groups. Our results suggest that UD is effective both in prevention and treatment for kidney stones. The mechanism underlying this effect may be the antioxidant effect of UD and the effect on the concentration of stone-forming components in the urine.Publication Open Access Bitter melon (Momordica charantia) fruit extract ameliorates methotrexate-induced reproductive toxicity in male rats(MARMARA UNIV, FAC MEDICINE, 2021-10-29) ŞEN, ALİ; Kanpalta, Fatma; Ozbeyli, Dilek; Sen, Ali; Cevik, Ozge; Sener, Goksel; Ercan, FerihaObjective: Methotrexate (MTX) is a drug commonly used for the treatment of malign neoplastic and inflammatory diseases. Anti-oxidant and anti-inflammatory effects of bitter melon (BM) were reported. The aim of this study was to examine the effects of BM fruit extract on MTX-induced testicular and epididymal damage. Materials and Methods: Sprague Dawley male rats were divided into three groups (n=8) as control, MTX and MTX+BM. A single dose of MTX (20 mg/kg) was injected intraperitoneally to the MTX and MTX+BM groups. BM fruit extract (600 mg/kg) was applied to the MTX+BM group orally for 5 days. Testes were examined for general histopathology, proliferating and apoptotic cells. The epididymis samples were used for the evaluation of sperm morphology. Oxidative and inflammatory markers were analysed biochemically. Results: Increased abnormal spermatozoa, degenerated seminiferous tubules with increased apoptotic cells and decreased proliferative cells were observed in the MTX group. TNF-alpha, IL-1 beta, 8-hydroxy-2-deoxyguanosine and caspase-3 levels increased, superoxide dismutase and catalase levels decreased in both testis and epididymis samples. All these histological and biochemical parameters were ameliorated in the MTX+BM group. Conclusion: Methotrexate causes testis damage by decreasing spermatogenic cells and increasing apoptosis through oxidative stress and inflammation. BM extract improves testis and epididymis damage with its possible anti-oxidant and anti-inflammatory effects.Publication Open Access Cotinus coggygria scop. The objective of the present study is to elucidate for the frst time the efects of Cotinus coggygria (CC) ethanol extract (CCE) on colonic structure and infammation of acetic acid-induced ulcerative colitis in rats. Colonic damage was assessed using disease activity index score, enzyme-linked immunosorbent assay, and hematoxylin–eosin staining. Also, in vitro antioxidant activity of CCE was investigated by ABTS methods. Total phytochemical content of CCE was measured spectroscopically. Acetic acid caused colonic damage according to disease activity index and macroscopic scoring. CCE signifcantly reversed these damages. While the levels of proinfammatory cytokines TNFalpha, IL-1beta, IL-6, and TGF-1beta increased in tissue with UC, IL-10 level decreased. CCE increased infammatory cytokine levels to values close to the sham group. At the same time, while markers indicating disease severity such as VEGF, COX-2, PGE2, and 8-OHdG indicated the disease in the colitis group, these values returned to normal with CCE. Histological research results support biochemical analysis. CCE exhibited signifcant antioxidant against ABTS radical. Also, CCE was found to have a high content of total polyphenolic compounds. These fndings provide evidence that CCE might be beneft as a promising novel therapy in the treatment of UC in humans due to high polyphenol content and justify the use of CC in folkloric medicine for treatment of infamed diseases.
