Publication Open Access Ethanol extract of Cotinus coggygria leaves accelerates wound healing process in diabetic rats(TAYLOR & FRANCIS LTD, 2016-11) ŞEN, ALİ; Aksoy, Halil; Sen, Ali; Sancar, Mesut; Sekerler, Turgut; Akakin, Dilek; Bitis, Leyla; Uras, Fikriye; Kultur, Sukran; Izzettin, Fikret VehbiContext:Cotinus coggygria Scop. (Anacardiaceae) leaves that were used as wound healing in traditional Balkan and Anatolian folk medicine, could be potentially effective in treating diabetic wounds.Objective: This study investigates biochemical and histological effects of ethanol extract of C. coggygria (CCE) on excision wound model in diabetic rats.Materials and methods: This study was conducted on diabetic Wistar albino rats, which were injected by a single dose (50mg/kg i.p.) streptozotocin. Afterward an excision wound model was created in all animals; diabetic control rats were applied topically simple ointment and diabetic treatment rats were applied topically 5% (w/w) ointment with CC, once a day during the experimental period. Malondialdehyde, glutathione and hydroxyproline levels in wound tissues were investigated at the end of 3rd, 7th, and 14th days. Histopathological examination was also performed.Results: Hydroxyproline content was significantly increased in the CCE treated group versus control after the 3rd and 7th days (15.33 versus 11.83; 19.67 versus 15.67mg/g, p<0.05; respectively). A statistically significant elevation in glutathione at the end of 3rd, 7th, and 14th days (5.13 versus 1.58, p<0.05; 4.72 versus 1.88, p<0.05; 3.83 versus 1.88mol/g, p<0.05, respectively) and a statistically significant decrease in malondialdehyde level at the end of 7th day (4.49 versus 1.48nmol/g, p<0.05) were determined in the treated group versus control group. These results were also supported by histological analyses.Discussion and conclusion: These findings indicate that CCE accelerated the cutaneous wound healing process in diabetic wounds, in confirmation of its traditional use.Publication Metadata only Chemical composition of endemic Stachys subnuda Montbret & Aucher ex Benth. essential oil and its anti-inflammatory and antioxidant activities(TAYLOR & FRANCIS INC, 2019) DOĞAN, AHMET; Sen, Ali; Kurkcuoglu, Mine; Bitis, Leyla; Dogan, Ahmet; Baser, Kemal Husnu CanThe essential oil of aerial parts of Stachys subnuda Montbret & Aucher ex Benth (S. subnuda) was analyzed by gas chromatography (GC) and GC/mass spectrometry (MS). Anti-inflammatory activity was evaluated by lipoxygenase inhibition assay. The antioxidant activity was determined by 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azinobis-3-ethylbenzothiozoline-6-sulfonic acid (ABTS) assays. Sixty-two constituents, comprising 97.1% of the total essential oil composition, were identified. Phytol (25.2%), beta-caryophyllene (11.2%), germacrene D (6.9%), and hexadecanoic acid (6.9%) were the major compounds of aerial part essential oils of S. subnuda. The essential oil showed significant radical scavenging activity with 50% inhibition (IC50) values of 0.124 and 0.204 mg/mL for ABTS and DPPH radicals, respectively. The essential oil exhibited moderate anti-inflammatory activity with an IC50 value of 0.419 mg/mL. The results indicated that S. subnuda essential oil had good anti-inflammatory and antioxidant activity. Also, this is first study on chemical composition, anti-inflammatory and antioxidant activity of S. subnuda essential oil.Publication Metadata only Effect of Horse-chestnut seed extract on matrix metalloproteinase-1 and-9 during diabetic wound healing(WILEY, 2019) ŞEN, ALİ; Aksoy, Halil; Cevik, Ozge; Sen, Ali; Goger, Fatih; Sekerler, Turgut; Sener, AzizeThe effects of aqueous-ethanol extract of Horse chestnut (HCE) on MMP-1 and MMP-9 expressions during cutaneous wound healing in diabetic rats were investigated in this study. The expressions of MMP-1 and MMP-9, wound closure, myeloperoxidase (MPO) activity, hydroxyproline, and malondialdehyde (MDA) levels in wound tissue were measured. Quercetin glucuronide in HCE was identified as main compound using a LC-MS/MS. The hydroxyproline level was significantly increased in the treated group versus control after the 3rd and 7th days (p < 0.05). The MDA level and MPO activity were significantly lower in the treatment group (p < 0.05). MMP-1 gene expression level in treated rats was increased in the 7th day while it was reduced in 14th day. MMP-9 gene expression level in treated rats was decreased in 7th, and 14th days compared to control (p < 0.05). These results show that HCE accelerated the cutaneous wound-healing process in diabetic rats via MMP-1 and MMP-9 regulation.Publication Metadata only THERAPEUTIC POTENTIAL OF MYRTUS COMMUNIS SUBSP COMMUNIS EXTRACT AGAINST ACETIC ACID-INDUCED COLONIC INFLAMMATION IN RATS(WILEY, 2017) ŞEN, ALİ; Sen, Ali; Yuksel, Meral; Bulut, Gizem; Bitis, Leyla; Ercan, Feriha; Ozyilmaz-Yay, Nagehan; Akbulut, Ozben; Cobanoglu, Hamit; Ozkan, Sevil; Sener, GokselThe aim of this study was to evaluate the effect of ethanol extract from leaves of Myrtus communis subsp. communis (MC) on acetic acid (AA)-induced ulcerative colitis in rats. On the fourth day of colitis induction, all rats were decapitated. Colitis was assessed by macroscopic and microscopic scores and by measuring malondialdehyde (MDA) and glutathione (GSH) levels, myeloperoxidase (MPO) activity, luminol, lucigenin, nitric oxid and peroxynitrite chemiluminescence (CL). Colitis caused significant increases in the colonic MDA levels, MPO activity, CL values, macroscopic and microscopic damage scores along with significant decrease in tissue GSH level. However, treatment with MC extract reversed all these biochemical indices, as well as histopathological alterations induced by AA with the protective effects being similar to that of sulphasalazine treatment. The study showed that MC extract could alleviate colitis in rats and can be considered an alternative therapeutic approach for management of inflammatory bowel diseases (IBD).Publication Open Access Two Acylated Isoscutellarein Glucosides with Anti-Inflammatory and Antioxidant Activities Isolated from Endemic Stachys Subnuda Montbret & Aucher ex Benth(SLOVENSKO KEMIJSKO DRUSTVO, 2019-12-18) DOĞAN, AHMET; Sen, Ali; Goger, Fatih; Dogan, Ahmet; Bitis, LeylaIn this study, we report anti-inflammatory and antioxidant activities of two acylated isoscutellarein glucosides isolated from ethyl acetate extract of Stachys subnuda aerial part. 4'-O-methylisoscutellarein-7-O-2 ''-O-(6 ''' acetyl-beta-D-allopyranosyl)-beta-D-glucopyranoside (SS1) and isoscutellarein-7-O-2 ''-O-(6 '''-O-acetyl-beta-D allopyranosyl)-beta-D -glucopyranoside (SS2) were isolated as major compounds from ethyl acetate extract (SSEA). Also, 2 hydroxycinnamic acid derivatives, and 5 isoscutellarein glucoside derivatives in the SSEA were identified using LC-MS/MS. SS1 with IC50 values of 2.35 and 1.98 mu g/mL and SS2 with IC50 values 13.94 and 12.76 mu g/mL showed fairly strong antioxidant activity against DPPH (1,1-diphenyl-2-picrylhydrazyl) and ABTS (2,2'-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid) radicals, respectively. SS1 and SS2 inhibited 5-lipoxygenase (5-LOX) activity with IC(50 )values of 47.23 and 41.60 mu g/mL, respectively. The results demonstrated that SS1 and SS2 have significant anti-inflammatory and antioxidant potential. Acylated flavonoid glycosides have been first reported for Stachys subnuda. Also, the activities of SS1 and SS2 have been investigated for the first time in this study.Publication Open Access Antioxidant and anti-inflammatory activity of fruit, leaf and branch extracts of Paliurus spina-christi P. Mill.(MARMARA UNIV, FAC PHARMACY, 2018-04-06) ŞEN, ALİ; Sen, AliIn this study, it is aimed to investigate antioxidant and anti-inflammatory activities of ethanol extracts and their n-hexane, chloroform, ethylacetate, and aqueous ethanol fractions of the fruits, leaves, and branches of Paliurus spinachristi P. Mill. (Rhamnaceae). The antioxidant activity of the extracts was tested by DPPH and ABTS methods and the results were expressed as IC50 values. The total phenolic content was determined by Folin-Ciocalteu method and the results were expressed as mg of gallic acid equivalent per g dry extract. Anti-inflammatory activity of extracts were evaluated against 5-lipoxygenase enzyme. In particular, all extracts obtained from branches of P. spina-christi, except hexane extract, showed strong antioxidant activity against DPPH and ABTS radicals. Ethyl acetate and ethanol extracts of branches of P. spina-christi showed the highest antioxidant activity in DPPH and ABTS assays with IC50 values of 15.54 and 22.06 mu g/ml, respectively. Also, the highest total phenol content was found in the ethylacetate extract of branches of P. spina-christi (286.6 mg/g). Ethyl acetate and ethanol extracts of leaves of P. spina-christi with IC50 values of 66.28 and 76.75 j mu g/ml were found to have the best anti-inflammatory activity in the lipoxygenase (LOX) inhibition assay used to evaluate anti-inflammatory activities of extracts. These results demonstrate that ethanol and ethyl acetate extracts from branches and leaves of Paliurus spina-christi have significant antioxidant and anti-inflammatory activity. Also, these results confirm the folk use of the plant as an antiinflammatory medicine.Publication Open Access Biological activities of aerial parts extracts of endemic Tanacetum argenteum subsp. argenteum(MARMARA UNIV, FAC PHARMACY, 2017-04-01) ŞEN, ALİ; Sen, Ali; Ozakpinar, Ozlem Bingol; Tan, Seher Birteksoz; Kultur, Sukran; Uras, Fikriye; Bitis, LeylaThe objective of the present study was to investigate the antimicrobial and anti-proliferative activities of n-hexane, chloroform, methanol and aqueous methanol extracts obtained from Tanacetum argenteum subsp. argenteum aerial parts. Anti-proliferative activity was tested in vitro against four human cancer cell lines (A549: lung adenocarcinoma, Hela; cervix adenocarcinoma, HT-29: colon adenocarcinoma, MCF-7; breast adenocarcinoma) using MTT assay. Antimicrobial activity was assessed by micro-broth dilution technique against Staphylococcus aureus ATCC 29213, Staphylococcus epidermidis ATCC 12228, Escherichia coli ATCC 25922, Enterococcus faecalis ATCC 29212, Klebsiella pneumoniae ATCC 4352, Pseudomonas aeruginosa ATCC 27853, Proteus mirabilis ATCC 14153, and Candida albicans ATCC 10231. Hexane extract inhibited proliferation of HT-29 and MCF-7 by 75 and 74% while chloroform extract against the same cancer cell lines displayed inhibition of 89 and 73% at the concentration of 30 mu g/mL, respectively. Also, chloroform extract at the same concentration showed significant anti-proliferative activity against A-549 and HeLa with inhibition values of 75% and 62%. Chloroform extract exhibited moderate antibacterial activity against Staphylococcus aureus and S. epidermidis with the MIC values of 625 mu g/mL. Methanol and aqueous methanol extracts showed weak antimicrobial activity against Staphylococcus epidermidis and Candida albicans with MIC values of 1250 mu g/mL. Evidences show that the key symptoms of AD are mainly caused by cholinergic system dysfunction which has a role in cognitive disorders. Cholinergic pathways especially muscarinic receptors like M-1 subtype also have a major role in learning, memory, cognitive functions and emotional state. There is no available permanent treatment currently to cure AD or to change its progression. This study was designed to investigate the factors that play important role in pathogenesis of AD and to compare the effects of Galantamine treatment with effects of Myrtus communis treatment. The expression level of M-1, ACh, BDNF; AChE activity, GSH level, MDA and MPO activity and AChE gene expression were investigated in scopolamine-induced rat model. Results showed that, administration of MC significantly improves the SCOP-induced reduction of latency and object recognition time; increasing BDNF, M-1 and ACh receptor expression levels in the different brain regions. Additionally, MC showed an increased in AChE by enhancing GSH activity and reducing MDA level and MPO activity. In conclusion MC considered as a possible novel therapeutic approach that can be a valuable alternative way in the prevention and treatment of AD.Publication Open Access Antioxidant and anti-inflammatory activity of capitula, leaf and stem extracts of Tanacetum cilicicum (Boiss.) Grierson(2019-06-13) DOĞAN, AHMET; Aybeniz YILDIRIM;ALİ ŞEN;AHMET DOĞAN;LEYLA BİTİŞIn this study, various extracts obtained different parts of Tanacetumcilicicum were investigated for in vitro antioxidant and anti-inflammatoryactivity. Antioxidant activity was tested with three methods; namely 2.2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging capacity, 2.2′‐azinobis (3‐ethylbenzothiazolin‐6‐sulphonic acid) (ABTS) radical cation scavengingcapacity, and ferric-reducing antioxidant power (FRAP) assays. Total phenolicand flavonoid contents of extracts were determined by Folin-Ciocalteu andaluminum chloride methods, respectively. Also, anti-inflammatory activity ofthese extracts was evaluated by 5-lipoxygenase inhibition assay. Ethyl acetateextract of capitula of T. cilicicum (TCCEA) showed the highest antioxidantactivity with IC50 values of 22.44 and 30.86 μg/mL against DPPH and ABTSradicals, respectively. At the same time, the highest ferric reducing power wasfound in the TCCEA (42.2 mg TE/g extract). The highest total phenolic contentshave been detected in TCCEA and ethyl acetate extract of leaves of T. cilicicum(TCLEA) with value of 174.1 and 175.6 mg GAE/g extract, respectively.Similarly, the highest total flavonoid contents have been detected in TCCEA andTCLEA with values of 26.94 and 30.48 mg QE/g extract, respectively. TCCEAexhibited strong anti-inflammatory activity with IC50 value of 9.44 μg/mL whencompared to standard indomethacine (22.39 μg/mL). These results demonstratethat TCCEA has a significant antioxidant and anti-inflammatory activity.
