A fluorodensitometric method was developed for the determination of baclofen. The method is based on the reaction of baclofen with 4-chloro-7-nitrobenzofurazan and measurement of the fluoresence intensity at 540 nm after chromatographic separation on a silica-gel plate. A linear relationship was found between relative fluorescence intensity and concentration over the range 4-100 ng of baclofen per spot. The detection and determination limits are 0.4 and 4.0 ng of baclofen per spot, respectively. The method was applied to human plasma. The recovery of baclofen from plasma was 96.67%.
