Publication: Trigeminal nevralji hastalığında serotonin 2A reseptör geni 5-HTR2A rs6311 polimorfizminin araştırılması
Abstract
Amaç : Trigeminal nevralji (TN) trigeminal sinirin bir veya birden fazla dalının dağılım alanında ani, genellikle tekrarlayan ağrı olması durumudur. Serotonin (5-HT) merkezi sinir sisteminde nörotransmitterlerin etkilerini düzenleyen bir hormondur. Çalışmanın amacı, TN’nin genetik yönden incelenerek hastalardaki serotonin reseptör geni (5-HTR2A) ve rs6311 polimorfizminin duyarlılıkla ağrı şiddeti arasındaki ilişkisini araştırmaktır. Gereç ve Yöntem : Çalışmamıza 10 trigeminal nevralji hastası ve 10 sağlıklı birey katılmıştır. Hastalardan alınan venöz kanlardan izole edilen DNA’lar kullanılmıştır. 5-HTR2A rs6311 polimorfizminin genotiplenmesi, StepOnePlus cihazında Real-Time PCR ve üreticilerin protokollerine uygun olarak Taqman SNP Genotipleme Testleri genotipleme kitleri kullanılarak gerçekleştirilmiştir. Bulgular : 5HTR2A rs6311 analizinde 10 trigeminal nevralji hastasının 3’ünde CC, 5'inde CT ve 2'sinde TT genotipi olduğu belirlendi. Kontrol grubunda 4 kişide CC, 4 kişide CT ve 2 kişide TT genotipi saptandı. Sonuç : Bu çalışmada elde edilen verilen ki-kare analizleri, hasta ve kontrol grubu arasındaki TN genotip ve alelik frekans farklılıklarının istatistiksel açıdan anlamlı olmadığını göstermiştir. Objective : Trigeminal neuralgia is often recurrent pain in the area of distribution of more branches of the trigeminal nerve. Serotonin is a hormone that regulates the neurotransmitters of the central nervous system. The aim of the study is to examine the relationship between pain content and responses of the expanded 5-HTR2A by examining the genetic aspects of trigeminal neuralgia. Material and Methods : Our study inluded 10 patents with trigeminal neuralgia patients and 10 healthy subjects as controls. DNA was extracted from the patients’ venous blood. The 5-HTR2A rs6311 polymorphism in the DNA was determined by using real-time PCR on a StepOnePlus instrument and the Taqman SNP Genotyping Assays genotyping kit according to manufacturers' guidelines. Results : In the 5-HTR2A rs6311 analysis, it was determined that 3 of 10 trigeminal neuralgia patients had the CC, 5 CT and 2 TT genotypes. In the control group, CC genotype was detected in 4 individuals, CT genotype in 4 individuals, and TT genotype in 2 individuals. Conclusion : Chi-square analysis of the data obtained in this study revealed that the differences in TN genotype and allele frequencies between the patient and control groups were not statistically significant.
