İzoniazid ve rifampisini beraber içeren lipozom formülasyonlarının hazırlanması ve özelliklerinin türev spektroskopisi yöntemi ile incelenmsei

Abstract

Lipozomlar toksik yan etkiler oluşturmaksızın dolaşımda yüksek ilaç konsantrasyonu sağlayarak uzun süre kalırlar ve lipozomlanmış ilacın serbest formundan daha düşük dozda kullanılabilmesine imkan verirler. İzoniazid ve rifampisin tüberküloz tedavisi için en etkili ilaçlardır. Bu ilaçların tek başlarına yüksek dozda ve uzun süreli kullanılmasının rezistans gelişimine sebep olması nedeniyle, bunların kombine halde kullanılması tüberküloz tedavisi için idealdir. Bu çalışmada, lipozomların avantajları düşünülerek, izoniazid ve rifampisin aynı lipozom formülasyonunda hazırlanmıştır; izoniazid sulu fazda tutulmuş ve rifampisin lipid membrana bağlanmıştır. Lipozomlar doymuş ve doymamış fosfolipidlerle hazırlanmıştır. Lipozomların fraksiyonlardaki % büyüklük dağılımı, % ilaç tutulması ve zamana karşı lipozomlardan salınan % ilaç miktarı incelenmiştir. Lipozomal ilaç içeriğinin miktar tayininin zor olması sebebiyle, kolay ve hızlı bir metod olan türev spektroskopisi izoniazid ve rifampisin içeren lipozomlara, yapının tahribine gerek kalmadan ilaçların direk tayini için uygulanmıştır. Bu çalışma, izoniazid ve rifampisinin aynı lipozomal formulasyonunda hazırlanabileceğini ve türev spektroskopisi ile tayin edilebileceğini gösterir. Aynı zamanda, iki farklı ilaç yüklü lipozomal formülasyonlardan türev spektroskopisi ile ilaç tayinini sağlayan ilk çalışmadır. PREPARATION OF ISONIAZID AND RIFAMPICIN CO-ENCAPSULATED LIPOSOMAL FORMULATIONS AND CHARACTERIZATION OF THE PROPERTIES BY DERIVATIVE UV SPECTROSCOPY METHOD

Liposomes maintain a high drug concentration in the circulation for a long period of time without causing actual toxic side effects and liposome encapsulated drug can be used in low doses than its free form. Isoniazid and rifampicin are the most effective drugs for the treatment of tuberculosis. Since a resistance occurs while using the drugs alone in high doses and long terms, combination of these drugs are ideal for the treatment of tuberculosis. In this study, taking into consideration the advantages of liposomes, isoniazid and rifampisin were incorporated in the same liposome formulation; isoniazid was incorporated in the aqueous phase and rifampicin was bond to lipid membrane. Liposomes were prepared with saturated and unsaturated phosphoplipids. The size distribution % of liposomes in the fractions, the encapsulation %, and the release % of drugs from the liposomes versus time were investigated. Since it is hard to determine the loading capacity of liposomal drug content, derivative UV spectroscopy, which is a simple and rapid method, was applied to isoniazid and rifampicin entrapped liposomes for direct estimation of drugs without disturbing the liposomal form. This study provides that it is possible to encapsulate isoniazid and rifampicin into the same liposomal formulation and evaluate them by derivative UV spectroscopy. This is also the first study on determination of drugs from two different drug loaded liposomal formulations by derivative UV spectroscopy.
