Publication: Ganoderma lucidum ve ticari ürünlerinin in vitro antioksidan aktivitelerinin ve bazı biyoaktif bileşenlerinin tayini ve karşılaştırılması
Abstract
Amaç: Bu çalışmada, son yıllarda sağlık alanında ve özellikle kanser tedavisindetedaviye destek amaçlı kullanılan Ganoderma lucidum tıbbi mantarının farklı formlarıve piyasada satılan kapsül formlarının in vitro antioksidan aktiviteleri ile bazı biyoaktifbileşenlerinin tayini ve karşılaştırılması amaçlanmıştır.Gereç ve yöntem: Ganoderma lucidum mantarı halk arasında çay olarak veya ekstresiiçilerek kullanılmaktadır. Piyasada kapsül formları da satılmaktadır. Üreticisindenkurutulmuş ve ekstrakt halinde temin edilen mantar örnekleri ve piyasadan alınankapsül formları, infüzyon yöntemi ile ekstrakte edildi. Mantarların öğütülmüş,dilimlenmiş, ticari ekstrakt halinde ve tablet formlarının antioksidan aktiviteleriCUPRAC, FRAP, ABTS ve DDPH yöntemi, toplam fenolik madde içeriklerini iseFolin-Ciocalteu ayıracı ile UV-VIS spektrofotometre kullanılarak tayin edildi. Troloksve gallik asit standartları kullanılarak elde edilen absorbans/ inhibisyon değerlerindenGanoderma lucidum’un farklı formlarının antioksidan kapasiteleri ve toplam fenolikbileşen miktarları TEAC ve GAE cinsinden hesaplandı.Bulgular: Öğütülmüş ve dilimlenmiş örneklerin antioksidan aktiviteleri (9,66-38,0mmol TEAC/ g örnek), ticari ekstraktların (0,3-10,91 mmol TEAC/ g örnek) arasında;kapsül formlarının ise antioksidan aktiviteleri (2,46-136,64 mmol TEAC/ g örnek)arasında; toplam fenolik bileşen miktarları sırası ile (29,14-116,21 mg GAE/ g örnek)arasında; ticari ekstre için fenolik bileşen miktarı ortalama (12,62 mg GAE/ g örnek)olarak bulunmuştur.Sonuçlar: Ganoderma Lucidum’un antioksidan özelliğe sahip olduğu ticari ekstraktınöğütülmüş ve dilimlenmiş formlara göre daha yüksek antioksidan ve fenolik bileşeniçerdiği görülmüştür. Kapsül formlarının da karşılaştırıldığında antioksidan aktiviteyeve fenolik içeriğe sahip olduğu, farklı 3 formun da birbiri ile kıyaslandığındafarklılıklar gösterdiği bulunmuştur. Kullanımda Ganoderma lucidum mantarının tercihedilmesini, günümüzde sentetik gıdalar yerine doğal gıda takviyesi ve bitkilerletedavide doktor kontrolünde kullanımının önerilebileceğini düşündürmüştür.
Aim: In this thesis, it was aimed that in recent years as a medicinal mushroom whichis used in the field of health and especially in the treatment of cancer to determine andcompare the in vitro antioxidant activities with some bioactive components of differentforms of Ganoderma lucidum and its capsule forms where sold in the market.Material and Method: Ganoderma lucidum mushroom is popularly used as a tea orby drinking its extract. Capsule forms are also available on the market. Mushroomsamples obtained from the manufacturer in dried form and capsule forms obtainedfrom the market were extracted by infusion method. The antioxidant activities of themushrooms in the ground, sliced, commercial extract and tablet forms weredetermined using the CUPRAC, FRAP, ABTS and DDPH method, and the totalphenolic content of the mushrooms using the Folin-Ciocalteu reagent and UV-VISspectrophotometer. Antioxidant capacities and total phenolic component amounts ofdifferent forms of Ganoderma lucidum were calculated from the absorbance / inhibition values obtained by using trolox and gallic acid standards in terms of TEACand GAE.Findings: Among the ground and sliced samples (9,66-38,0 mmol TEAC/ g sample),commercial extracts (0,3-10,91 mmol TEAC/ g sample) for antioxidant activitydetermination; among the capsule forms, antioxidant activities (2.46-136.64 mmolTEAC / g sample); total phenolic component amounts were found to be between(29.14-116.21 mg GAE / g sample) respectively, and phenolic component amount forcommercial extract (12.62 mg GAE / g sample).Results: Commercial extract of Ganoderma Lucidum has been found to contain higherantioxidant and phenolic components than milled and sliced forms. It should be suggested that Ganoderma Lucidum shouldbe preferred over synthetic products in the treatment with herbs and as a foodsupplement, but it can be recommended to be used under doctor control in medicaltreatment.
