Publication: Karbamazepin Fe (III) etkileşiminin spektrofotometrik yöntemle incelenmesi
Abstract
Karbamazepin yapısı bakımından imipramin’e benzeyen antiepileptik olarak kullanılan iminostilben türevi bir ilaçtır. İlacın en sık görülen yan etkileri sindirim sistemi bozuklukları, nörolojik bozukluklar, antikolinerjik etkiler, alerjik cilt bozuklukları, kemik iliği depresyonu ve anemidir. Bu çalışmada karbamazepin Fe(III) etkileşimini in vitro ortamda incelenerek karbamazepinin, demir ile kompleks oluşturan maddelere benzer biçimde demir eksikliğine bağlı anemiye neden olup olmadığı yönünde kimyasal veriler elde edilmeye çalışılmıştır. Bu amaçla karbamazepin - Fe(III) ve önemli metabolitlerinden biri olan 10,11-dihidroksikarbamazepin - Fe(III) etkileşimi spektrofotometrik yöntemle uygun çözücü, reaksiyon süresi, reaksiyon sıcaklığı, pH ve belirteç miktarı gibi reaksiyon koşulları açısından incelenmiştir. Karbamazepin ve 10,11-dihidroksikarbamazepin ile Fe(III) arasındaki kimyasal etkileşimin spektrofotometrik yöntemin yanında farklı bir yöntemle de desteklenmesi amacıyla potansiyometrik yöntemle de çalışılmıştır. Yapılan çalışmalar sonucunda karbamazepin - Fe(III) etkileşiminin en iyi %10 dioksan-su çözücü ortamında, pH 2-3 aralığında, oda sıcaklığında, 5 dakika reaksiyon süresinde ve eşit konsantrasyonda gerçekleştiği bulunmuştur. 10,11-dihidroksikarbamazepin - Fe(III) etkileşiminin ise en iyi sulu çözeltide, oda sıcaklığında, 5 dakika reaksiyon süresinde, pH 2-3 aralığında ve iki katı konsantrasyonda 10,11-dihidroksikarbamazepin ile gerçekleştiği bulunmuştur. Spetrofotometrik yönteme destek amacıyla yapılan potansiyometrik çalışmalar sonucunda karbamazepin - Fe(III) ve 10,11-dihidroksikarbamazepin - Fe(III) etkileşimi gösteren titrasyon eğrilerinde ayrılmalar olmasına rağmen eğriler arasındaki farklar kullanılan potansiyometrik yöntemle hesaba olanak vermemiştir. Ancak eğrilerin az da olsa ayrılması zayıf bir etkileşime işaret etmekte bu da spektrofotometrik olarak bulunan sonuçları desteklemektedir.
Carbamazepine structure is similar to imipramine. It is an antiepileptic drug. It’s advers effects are GI system defect, neurologic defect, allergic skin reactions and bone marrow defect. In this study carbamazepine-Fe(III) interaction was investigated in in-vitro media in order to examine anemia owing to iron deficiency. The datas obtained via investigation of the compound that forms complexes with iron. Carbamazepine-Fe(III) interaction was investigated spectrophotometrically by means of approperiate solvent, reaction time, temprature and quantity of reactive. To suppor the spectrophotometric investigation study of the chemical compatibility of carbamazepine and 10,11 dihydroxycarbamazepine with Fe(III), the compatibility was investigated also using potentiometric method. At the end of the studies performed, the interaction reaction of carbamazepine with Fe(III) was found to occur optimally in 10% dioxan-water solvent medium, at the interval of pH=2-3, at room temprature, in 5 min reaction time and the same concentration. The interaction reaction of 10,11 dihydroxycarbamazepine which is one of the most important metabolites of carbamazepine with Fe(III) was found to ocur optimally in aqueous solution at room temprature, in 5 min reaction time, at the internal of pH=2-3 and in twice concentration of 10,11 dihydroxycarbamazepine. As a result of the potantiometric studies performed to support the spectrophotometric method; although there were deviations between the titration curves, which show interactions of carbamazepine- Fe(III) and 10,11 dihydroxycarbamazepine-Fe(III), the deviations between curves didn’t enable a calculation using potantiometric method. But even there were small deviations between the curves, these point out weak interactions and this supports the spectrophotometric results.
