Publication: Deneysel kolestaz kaynaklı hepatorenal hasar ve kardiyomiyopatinin tedavisinde küsküt (Cuscuta sp.) bitkisinin etkisinin incelenmesi
Abstract
Bu çalışmada, sıçanlarda safra kanalı bağlama (SKB) yöntemi ile geliştirilen kolestazın neden olduğu hepatorenal hasar ve kardiyomiyopatide Küsküt otu ekstresinin potansiyel koruyucu veya önleyici etkisinin incelenmesi amaçlanmıştır. Gereç ve Yöntem: Spraque Dawley sıçanlar Kontrol, Küsküt (Küs), SKB ve SKB+Küs olarak 4 gruba ayrılmıştır. Safra kanalı bağlama işlemi safra kanalının çift ligasyonu ile yapılmıştır. 28 gün boyunca iki gruba (Kontrol ve SKB ) serum fizyolojik (SF) ve diğer iki gruba da (Küs ve SKB+Küs) 250 mg/ kg Küsküt metanol ekstresi oral gavaj ile verilmiştir. 28. günün sonunda tüm hayvanlar dekapite edilmiş olup serum, kalp ve böbrek dokusu örnekleri alınmıştır. Serumda ALT (Alanin transaminaz), AST (Aspartat aminotransferaz), DB (Direkt Bilirubin) ve TB (Total Bilirubin) seviyeleri incelenirken kalp ve böbrek dokularında ise TGF-β (Dönüştürücü büyüme faktörü- beta), 8-OHdG (8-hidroksi-2’-deoksiguanozin), Na⁺/ K⁺-ATPaz (Sodyum-Potasyum Adenozin Trifosfataz) ve Hidroksiprolin seviyeleri ölçülüp histolojik analizler yapılmıştır. Bulgular: SKB grubu hayvanlarda serum ALT, AST, Direkt Bilirubin ve Total Bilirubin seviyeleri ile kalp ve böbrek dokularında 8-OHdG, TGF-β, ve hidroksiprolin düzeyleri yüksek Na⁺/ K⁺-ATPaz düzeyleri düşük bulunmuştur. Küsküt metanol ekstresi ile tedavi edilen grupta biyokimyasal parametreler anlamlı derecede (p<0,05-0,001) tersine çevrilmiştir. Biyokimyasal sonuçlar histolojik sonuçlarla desteklenmiştir. Sonuç: : Çalışmamız sonuçları, kolestaz kaynaklı gelişen hepatorenal hasar ve kardiyomiyopatide Küsküt otunun yararlı etkileri olabileceğini göstermektedir.
Objective: : In this study, it was aimed to investigate the potential protective or preventive effect of the extract of Cuscuta sp. in hepatorenal injury and cardiomyopathy due to cholestasis induced by bile duct ligation (BDL) in rats. Materials and Methods: Spraque Dawley rats were divided into 4 groups as Control, Cuscuta sp. (CUS), BDL and BDL+ CUS. Bile duct ligation was performed by double ligation of the bile duct. For 28 days, two groups (Control and BDL) were given physiological saline and the other two groups (CUS and BDL+CUS) 250 mg/ kg Cuscuta methanol extract by oral gavage. At the end of the 28th day, serum and tissue samples were collected from all animals after decapitation. While ALT (alanine transaminase), AST (aspartate aminotransferase), DB and TB levels were examined in blood samples, 8-OHdG, Na⁺/ K⁺-ATPase, TGF-β and Hydroxyproline were determined in heart and kidney tissues followed by histological analyses. Results: In animals with bile duct ligated, serum ALT, AST, DB and TB levels and TGF-β, 8-OHdG and Hydroxyproline levels in heart and kidney tissues were high and Na⁺/ K⁺-ATPase levels were low. Biochemical parameters were significantly reversed (p<0.05-0.001) in the group treated with Cuscuta methanol extract. Histological results also supported the biochemical results. Conclusion: The results of our study suggest that Cuscuta sp. may be beneficial in hepatorenal injury and cardiomyopathy caused by cholestasis.
