Publication: Atopik Dermatit’li hastalarda dental folikül kök hücrelerin naive T lenfositlerin Th1 Ve Th2 yönünde farklılaşmasi üzerine etkisi
Abstract
Amaç: Bu çalışmada, Dental Folikül Mezenkimal Kök Hücrelerin (DF-MKH) Th2 baskın Atopik Dermatit hastalarının periferal kanından izole edilen lenfositleri üzerindeki immünomodülatör etkisinin in vitro ortamda araştırılması amaçlanmıştır.Yöntem: AD tanısı almış hastalar (n=9), Psoriasis hastaları (n=6) ve sağlıklı bireyler (n=6) çalışmaya dâhil edilmiştir. Bireylerin 20 ml venöz kanından Periferal Kan Mononükleer Hücre (PKMH) izolasyonu yapıldı. PKMH’ler uyaransız ve uyaranlı ortamda DF-MKH varlığında ve yokluğunda 72 saat kültür edildi. Süre sonunda CD4+/ CD8+T lenfosit proliferasyonu, hücre apoptoz belirteçleri, T regülatör hücre oranları, Naive/ bellek T hücre oranları ve kültür süpernatantından sitokin düzeyleri flow sitometri cihazında, GATA3, Tbet1 ve FoxP3 protein ekspresyonları Western Blot yöntemiyle analiz edildi.Bulgular: AD ve psoriasis hastalarında CD4+ ve CD8+T lenfositlerin DF-MKH varlığında, Fas(CD95), FasL (CD178) ve TNFRII yüzdesi anlamlı derecede baskılandı (p ‹0,05). Her iki hasta grubunda bellek T hücre oranı DF-MKH varlığında anlamlı olarak azalırken, Naive T hücreleri anlamlı olarak arttı (p ‹0,05). DF-MKH varlığında CD4+CD25+Foxp3+ oranı AD grubunda arttı (p ‹0,05). IL-17A, IL-6 ve TNF-α düzeyleri her iki hasta grubunda anlamlı olarak baskılandı (p ‹0,05). IFN-γ ve IL-10 düzeyleri AD hastalarında DF-MKH varlığında anlamlı olarak arttı (p ‹0,05). Sonuç: DF-MKH’lerin Th2 baskın AD hastalıklarında immün yanıtı düzelttiği saptandı. Sonuçlarımız DF-MKH’lerin AD’de immünomodülatör etkisi olduğunu ve tedavi yaklaşımlarında kullanılmasının uygun olabileceğini düşündürmektedir.
Objective: The purpose of our study is to investigate the immunomodulatory effects of Dental Follicle Mesenchymal Stem Cells (DF-MSCs) on lymphocytes isolated from peripheral blood of AD patients in vitro.Material and Methods: Patients with the AD (n=9) and psoriasis (n=6) and healthy subjects (n=6) were included in the study. Peripheral Blood Mononuclear Cells (PBMCs) were isolated from 20 ml of venous blood of all participants. Cells were cultured for 72 hours in the absence and presence of DF-MSCs. At the end of this period, Lymphocyte proliferation, CD4+CD25+FoxP3+ T regulatory cell frequency, apoptosis by AnnexinV/ 7AAD, naive and memory CD4+ and CD8+T lymphocyte ratio, supernatant cytokine levels, GATA-3, Tbet and FoxP3 expressions of cultures cells were analyzed.Results: The percentage of Fas (CD95), FasL (CD178) and TNFRII was significantly suppressed in the presence of DF-MKH in CD4+ and CD8+ T lymphocytes in AD and psoriasis patients (p <0,05). While the memory T cell ratio in both groups of patients was significantly decreased in the presence of DF-MKH and the Naive T cells increased significantly (p <0.05). CD4+CD25+Foxp3+ ratio was increased in the presence of DF-MKH in all patients’ group. IL-17A, IL-6 and TNF-α levels were significantly suppressed in both groups (p <0.05). IFN-γ and IL-10 levels were significantly increased in the presence of DF-MKH in AD patients (p <0.05). Conclusion: DF-MSCs regulated Th2 responses in AD patients. Our results support that DF-MSCs could have a beneficial therapeutic implication for AD.
