Goa cDNA’ sının E.coli’ de pTrcHis 2A vektör sistemine klonlanması

Abstract

Bu çalışmada heterotrimerik G-proteinleri ailesinin bir üyesi olan Goa cDNA'sının ticari olarak üretilmiş bir ekspresyon ve saflaştırma sistemi olan Xpress System Protein Purification (Invitrogen) sistemine klonlanması gerçekleştirilmiştir. Goa cDNA'sı, John Hopkins Üniversitesinden Dr.Randall Reed tarafından pGEM-2 vektörü içerisinde gönderilmiştir. Buradan çıkarılan Goa dizisi çeşitli işlemlerden geçirilerek, daha önce hazırlamış bulunduğumuz pTrcHis2A vektörü ile birleştirilmiş ve ekspresyon hücrelerine aktarılmıştır. Bu sistem proteinin C-ucuna eklediği 6 histidin rezidüsü yardımıyla saflaştırmayı tek aşamada ve kolaylıkla gerçekleştirmeye olanak sağlamaktadır.Klonlama yöntem olarak başarıyla gerçekleştirilmekle birlikte PZR aşamasında gerçekleşen tek baz kayması nedeniyle ekspresyon test edilememiştir. Cloning to pTrcHis2A Vector System of Goa cDNA in E.coli

The aim of this work was to clone Goa cDNA into ''Xpress System Protein Purification system, a commercial kit produced by Invitrogen for easy expression and purification. Goa cDNA was very kindly provided by Dr.Randall Reed from John Hopkins University in a pGEM-2 vector. The Goa was extracted from this vector and inserted into the pTrcHis 2A vector, which had been prepared beforehand. This vector which adds histidine residues to the carboxyl terminus of proteins provides an easy, one-step purification procedure. We have successfully cloned the Goa but expression could not be achieved due to a base shift possibly occurred during the PCR.
