Sambucus nigra L. türünün in vitro yara iyileştirici etkisinin değerlendirilmesi

Abstract

Amaç: Bu çalışmada S. nigra L. türünün dal, yaprak ve çiçeklerinden elde edilen ekstrelerin ve fraksiyonların in vitro yara iyileştirici etkileri ilk defa araştırılmıştır. Gereç ve Yöntem: S. nigra bitkisinin ekstrelerinin ve fraksiyonlarının in vitro sitotoksik aktiviteleri NIH3T3 hücrelerinde MTT yöntemi, aktif fraksiyonlarının in vitro yara iyileştirici aktiviteleri ise NIH3T3 hücrelerinde çizik testi kullanılarak değerlendirildi. Ekstre ve fraksiyonların toplam fenolik bileşik miktarı FCR, in vitro antioksidan aktiviteleri DPPH ve ABTS, in vitro anti-enflamatuvar aktiviteleri ise 5-lipoksijenaz inhibitör aktivite yöntemleriyle değerlendirildi. Bulgular: MTT bulguları S. nigra dal ekstresi ile dal etil asetat ve kloroform fraksiyonlarının yüksek proliferatif etkisi olduğunu, çizik testinde etil asetat fraksiyonunun kloroform fraksiyonundan yüksek yara alanı kapattığını göstermiştir. DPPH analizinde kontrole göre çiçek ekstresi ve dal etil asetat fraksiyonu orta düzey antioksidan aktivite; ABTS analizinde kontrole göre çiçek ekstresi orta, dal etil asetat fraksiyonu iyi antioksidan aktivite; 5-lipoksijenaz inhibisyon testinde kontrole göre çiçek ekstresi ve dal sulu etanol fraksiyonu orta derece anti-enflamatuar aktivite göstermiştir. Sonuçlar: Elde edilen verilerde S. nigra dalının yara iyileşmesine olumlu etkileri olduğunu ve bitkinin geleneksel halk tıbbında yara iyileştirici olarak kullanımını doğrulamaktadır. Ayrıca bu sonucun in vivo yara iyi edici aktivite çalışmalarıyla desteklenmesine ihtiyaç vardır.

Objective: In this study, in vitro wound healing effects of extracts and fractions obtained from branches, leaves and flowers of S. nigra L. were investigated for the first time. Materials and Methods: In vitro cytotoxic activities of extracts and fractions of S. nigra plant were evaluated using MTT method on NIH3T3 cells, and in vitro wound healing activities of active fractions were evaluated using scratch test on NIH3T3 cells. The total amount of phenolic compounds of the extracts and fractions were evaluated by FCR, in vitro antioxidant activities by DPPH and ABTS, and in vitro anti-inflammatory activities were evaluated by 5-lipoxygenase inhibitory activity methods Results: MTT findings showed that S. nigra branch extract and branch ethyl acetate and chloroform fractions had a high proliferative effect, and in the scratch test, the ethyl acetate fraction covered the wound area higher than the chloroform fraction. Flower extract and branch ethyl acetate fraction moderate antioxidant activity in DPPH analysis compared to control; In ABTS analysis, flower extract and branch ethyl acetate fraction good antioxidant activity; In the 5-lipoxygenase inhibition test, the flower extract and the branch aqueous ethanol fraction had moderate anti-inflammatory activity. Conclusion: The data obtained confirm that the branch of S. nigra has positive effects on wound healing and the use of the plant as a wound healer in traditional folk medicine. In addition, this result needs to be supported by in vivo wound healing activity studies.
