Publication: Achillea sieheana stapf bitkisinden biyoaktivite yönlendirmeli etken madde izolasyonu
Abstract
Amaç: Bu çalışmanın amacı, endemik Achilllea sieheana Stapf bitkisinin toprak üstü kısımlarından elde edilen hekzan, diklorometan, etil asetat ve metanol ekstrelerinin antimikrobiyal, antienflamatuar ve sitotoksik aktivitelerini incelemek ve etkili bileşikleri izole ederek, yapılarını aydınlatmaktadır. Gereç ve Yöntem: Ekstrelerin antimikrobiyal aktivite (MİK) sonucuna göre etkili hekzan ekstresinden, antienflamatuar (5-LOX) ve sitotoksik (MTT) aktivite sonuçlarına göre ise diklorometan ekstresinden etkili bileşikleri saflaştırmak için kolon kromatografisi yöntemleri kullanılmıştır. Saflaştırılan bileşiklerin yapıları spektroskopik yöntemlerle aydınlatılmıştır. İzole edilen bileşiklerin biyolojik aktiviteleri incelenmiştir. Etil asetat ekstresindeki bileşikler ise LC-MS/ MS yöntemi ile tayin edilmiştir. Bulgular: A. sieheana bitkisinin hekzan ekstresinden steroit türevi β-sitosterol; flavonoit türevleri salvigenin, artemetin, öpatilin 7-metil eter, kastisin; seskiterpen lakton türevi lökodin bileşikleri izole edilmiştir. Diklorometan ekstresinden flavonoit türevleri sirsimaritin, santin, viskosin; seskiterpen laton türevi lökodin ve tanaphillin izole edilmiştir. Etil asetat ekstresinin LC-MS/ MS yöntemiyle kimyasal içeriği feruloilkinik asit, luteolin, isoramnetin, isoviteksin, krisoeriol olarak belirlenmiştir. Sonuçlar: Diklorometan ekstresinin HT-29, PC-3 ve A-549 hücre hatlarına karşı 4 μg/ mL’den düşük İK50 değeri göstermesi potansiyel antikanser ilaç olabileceğini düşündürmüş ve bu çalışma ile ilk kez endemik A. sieheana bitkisinin biyolojik aktiviteleri incelenerek, aktivite yönlendirmeli izolasyon işlemleri gerçekleştirilmiştir.
Objective: The aim of this study is to investigate the antimicrobial, anti-inflammatory and cytotoxic activities of hexane, dichloromethane, ethyl acetate, and methanol extracts obtained from the aerial parts of the endemic Achillea sieheana Stapf, and to isolate the active compounds and elucidate their structures. Material and Methods: Column chromatography methods were used to purify the active compounds from the hexane extract according to the antimicrobial activity (MIC) results of the extracts, and from the dichloromethane extract according to the anti-inflammatory (5-LOX) and cytotoxic (MTT) activity results. The structures of the purified compounds were elucidated by spectroscopic methods. The biological activities of the isolated compounds were investigated. The compounds in the ethyl acetate extract were determined by LC-MS/ MS method. Results: Steroidal derivative, β-sitosterol; flavonoid derivatives, salvigenin, artemetine, eupatilin 7-methyl ether, casticin and sesquiterpene lactone derivative, leucodin compounds from the hexane extract of A. sieheana. Sirsimaritin, santin, viscosine, leucodin, tanaphillin compounds were isolated from the dichloromethane extract. The chemical content of ethyl acetate extract was also determined as feruloyl quinic acid, luteolin, isorhamnetin, isovitexin, chrysoeriol by LC-MS/ MS method. Conclusion: The fact that dichloromethane extract showed an IC50 value of less than 4 μg/ mL against HT-29, PC-3, and A-549 cell lines suggested that it may be a potential anticancer drug and with this study, the biological activities of the endemic A. sieheana plant were examined for the first time, and activity-guided isolation was carried out.
