Abstract
Türkiye’de Yetiştirilen Maviyemiş Türlerinde Ellajik Asit ve Resveratrol Miktarlarının HPLC Yöntemi İle Tayini Bu çalışmada, Ellajik Asit ve Resveratrol için yüksek basınçlı sıvı kromatografisi ve diod dizi dedektör ile miktar tayini yöntemi geliştirildi. Türkiye’de Karadeniz Bölgesi’nde kültürü yapılan dört farklı maviyemiş örneğinde (Bluecrop, Brigitta, Darrow ve Bluejay) bulunan Ellajik asit ve Resveratrol ilk kez analiz edildi. Kromatografik koşullar; hareketli faz: 5 mM Potasyum dihidrojenfosfat / Asetonitril’ den oluşmuştur. Dereceli Elüsyon yöntemi kullanılmıştır. Kolon : Luna ODS-2 RP- C18 (5μ m, 4.6 ×250 mm i.d.), Dedektör : DAD Dedektör, Dalgaboyu Ellajik asit için λ : 260 nm, Resveratrol için λ : 310 nm (Band genişliği: 4 nm), Akış Hızı : 1 mL/ dak., Enjeksiyon Hacmi: 20µL, Basınç: 162 bar Ellajik asit için konsantrasyon aralığı: 0.4963 – 11.5815 x 10-6 mol.L-1, y = 40.733x + 1.9204 (r2 = 0,9963); dedeksiyon limiti 0.1271 x 10-6 M, kantitatif tayin limiti 0.3814 x 10-6 M Resveratrol için konsantrasyon aralığı: 0.6571 – 10.9530 x 10-6 mol.L-1, y = 35.105x – 9.2686 (r2 = 0,9977); dedeksiyon limiti dedeksiyon limiti 0.1898 x 10-6 M, kantitatif tayin limiti 0.5694 x 10-6 M Geri kazanım Ellajik asit için % 91.85; Resveratrol için % 84.97 olarak bulunmuştur. Determination of the Amount of Ellagic Acid and Resveratrol in Blueberry Samples Grown in the Turkey by HPLC Method In this study, a quantification method was developed for Ellagic Acid and Resveratrol by high performance liquid choromatography and Diode array detector. Four different blueberry varities, grown in the Black Sea Region of Turkey, (Bluecrop, Brigitta, Darrow and Bluejay) were analyzed first time for Ellagic Acid and Resveratrol quantification. Chromatographic conditions; mobile phase: 5 mM Potassium dihydrogen phosphate / ACN and gradient elution was used.
