Abstract
FLUORİMETRİK PESTİSİD TAYİN YÖNTEMİ GELİŞTİRİLMESİ Pestisit kalıntısının izlenmesi, yiyecekler içindeki pestisitlere insanların maruziyetinin doğru olarak değerlendirilmesi açısından oldukça önemlidir. Bu nedenle gıda maddelerinde kalıntı düzeylerinin belirlenmesi ve kullanılan analitik yöntemlerin düşük düzeyde kalıntı ölçümü yapabilmesi önem kazanmaktadır. Kumaklor [3-( α-asetonil-p-kloro benzil)-4-hidroksikumarin] kumarin türevli bir pestisittir ve sıçan populasyonunu kontrol etmek için bir rodentisit olarak yaygın olarak kullanılır. Bu çalışmada, setiltrimetilamonyum bromurün (CTAB) miselli çözeltisi içinde kumaklorun tayini için fluorimetrik bir metod geliştirilmiştir. Tayin pH 9’da ve 0,002M CTAB kullanılarak yapılmıştır. Miselli çözeltilerdeki fluoresans emisyon şiddetleri 389 nm’de (uyarma 310 nm’de) ölçülmüştür. Kumaklorun lineer kalibrasyon aralığı 5-170 μg/ L’dir. Metodun tesbit ve tayin sınırları sırasıyla 14,18 ve 16,60 μg/ L’dir.
DEVELOPMENT OF METHOD FOR PESTICIDE DETERMINATION BY FLUORIMETRY Monitoring of pesticide residues in food is important from the viewpoint of the evaluation of the pesticide exposure to human being. Therefore, the determination of the residues levels in food and the analytical methods used for the trace level mesurement of the pesticide have acquired more importance. Coumachlor [3-( α-acetonyl-p-chloro benzyl)-4-hydroxycoumarin] is a coumarin-derivative pesticide and is widely used as rodenticide to take control the rat population. In this work, a fluorimetric method was developed for the determination of coumachlor in aqueous micellar solutions of cetyltrimethylammonium bromide (CTAB). The determination was performed at pH 9 and in the presence of 0.002M CTAB. In micellar solutions the fluorescence emission intensity was measured at 389 nm (excitation at 310 nm). The linear calibration range for coumachlor was 5–170 μg/ L. Limit of detection and quantification for the method were 14.18 and 16.60 μg/ L, respectively.
