Publication: Anne sütünde insülin benzeri büyüme faktörü I’in bağlayıcı proteinlerinin blotting metodu ile analizi
Abstract
ANNE SÜTÜNDE İNSÜLİN BENZERİ BÜYÜME FAKTÖRÜ I'İN BAĞLAYICI PROTEİNLERİNİN BLOTTİNG METODU İLE ANALİZİ Anne sütünde insülin benzeri büyüme faktörü bağlayıcı proteinlerinin belirlenmesi için öncelikle yağsız anne sütü SDS-PAGE elektroforezine tabi tutuldu ve bunu takiben Western Blotting yöntemi uygulandı. Blotting sonucu nitroselüloz membran 125I ile işaretli IGF-I ile bir gece inkübe edildi. Daha sonra Kodak BioMax MS-1 X-Ray filmi ile dört gün boyunca -70°C'de inkübasyona bırakıldı. Elde edilen filmin özel filtre kullanılarak fotoğrafı çekildi. Bu şekilde IGF-I ile bağlanan IGFBP-2 ve IGFBP-3'ün varlığı kanıtlanmış oldu.
ANALYSIS OF INSULIN LIKE GROWTH FACTOR-I BINDING PROTEINS IN HUMAN MILK USING WESTERN BLOTTING METHOD Defatted breast milk proteins were identified by SDS-Polyacryamide gel electrophosis and the protein bands were blotted by Western Blotting method. To idedntify the insulin like growth factor-I binding proteins, the blot was incubated overnight with 125I-labeled IGF-I. The blot was then exposed to Kodak BioMax MS-1 X-Ray film for four days at -70°C to screen the proteins. The film was developed and was shown that IGFBP-2 and IGFBP-3 were bounded by labeled IGF-I.
