Abstract

Lamium Purpureum Bitkisinin Prostat Kanseri Hücre Hattında Antikanser Etkisinin İncelenmesi Öğrenci Adı: Ayca GÜNER Danışman Adı: Prof. Dr. Pınar MEGA TİBER Ana Bilim Dalı: Biyofizik Amaç: Bu çalışmada, Lamium purpureum bitkisinden elde edilen ekstrelerin prostat kanser hücre hatlarında sitotoksik ve apoptotik etkisinin saptanması amaçlandı. Gereç ve Yöntem: LNCaP hücresinde Lamium purpureum bitkisinden elde edilen SSH, SSSE, SSK, SSEA ve SSME ekstrelerinin canlılık ve sitotoksisite üzerine etkisi WST-8 yöntemi ile belirlendi. Lamium purpureum ekstrelerinin IC50 değeri hesaplandı. JC-1 mitokondriyal membran potansiyeli testi ile apoptoza bağlı olarak meydana gelen potansiyeldeki değişimler incelendi. Ekstrelerin hücrelerde indüklediği Kaspaz 3/ 7 enzimatik aktivitesi belirlendi. Elde edilen sonuçlar GraphPad Prism 5 istatistik programında analiz edildi. Bulgular: L. purpureum bitkisinden elde edilen ekstratların 24 ve 48 saatlik inkübasyon sonrası IC50 değerleri sırasıyla SSH için 20.33 µM ve 19.93 µM; SSSE için 86.66 µM ve 48.81 µM; SSK için 78.07 µM ve 35.05 µM; SSEA için 70.41 µM ve 56.92 µM; SSME için 27.18 µM ve 15.35 µM olarak hesaplandı. Mitokondriyal membran potansiyelinde, SSH ve SSK için deney grubunda kontrol grubuna göre anlamlı bir floresans oranı yükselişi gözlenmedi. Kaspaz 3/ 7 aktivitesinde SSSE ve SSME için artmış görünse de istatistiksel olarak anlamlı bir değişiklik görülmemiştir. Sonuç: Araştırmamızda, L. purpureum bitkisinde elde edilen ekstrelerinin LNCap prostat kanser hücre hattında apoptotik indüksiyonu tetiklemediği ama sitotoksik etki gösterdiği görülmüştür. Etki mekanizmalarının daha detaylı anlaşılması için ileri çalışmalara gereksinim vardır.

Investigation of anticancer effect of lamium purpureum plant on prostate cancer cell line Student Name: Ayca GUNER Name of Supervisor: Prof. Pınar MEGA TIBER Department: Biophysics Objective: In this study, it was aimed to determine the cytotoxicity and anticancer effect of Lamium purpureum plant extracts in prostate cancer cell lines. Material and methods: The effects of SSH, SSSE, SSK, SSEA and SSME extracts obtained from Lamium purpureum plant on LnCap cell viability and cytotoxicity were determined by WST-8 method. The IC50 value of lamium purpureum extracts was calculated. The changes in the potential due to apoptosis were examined with the JC-1 mitochondrial membrane potential test. The apoptosis rate of the extracts in cells was determined by Caspase 3/ 7 enzymatic reaction. The obtained results were analyzed in GraphPad Prism 5 statistical program. Result: The IC50 values of the extracts obtained from the L. purpureum plant after 24 and 48 hours of incubation were 20.33 µM and 19.93 µM for SSH; 86.66 µM and 48.81 µM for SSSE; 78.07 µM and 35.05 µM for SSK; 70.41 µM and 56.92 µM for SSEA; 27.18 µM and 15.35 µM for SSME, respectively. No significant fluorescence rate increase was observed in the experimental group for SSH and SSK in mitochondrial membrane potential compared to the control group. Although caspase 3/ 7 activity appeared to be increased for SSSE and SSME, no statistically significant change was observed. Conclusion: In our study, it was observed that the extracts obtained from the L. purpureum plant did not trigger apoptotic induction in the LNCap prostate cancer cell line, but showed a cytotoxic effect. Further studies are needed to understand the mechanisms of action in more detail.
