Nisergolin’in safsızlıkları olan 10-metoksi-6-metilergolin-8beta-metonol-5bromo-3-piridinkarboksilat (1-DN) ve 10-metoksi-1,6-dimetilergolin-8beta-metanol-5-kloro-3-piridinkarboksilat (5-CN) için HPLC ve diod dizi dedektör kullanılarak miktar tayini yöntemi geliştirilmesi

Abstract

Nisergolin'in safsızlıkları olan 10-metoksi-6-metilergolin-8b-metanol-5-bromo-3-piridinkarboksilat (1-DN) ve 10-metoksi-1,6-dimetilergolin-8b-metanol-5-kloro-3-piridinkarboksilat (5-CN) için yüksek basınçlı sıvı kromatografisi ve diod dizi dedektör kullanılarak miktar tayini yöntemi geliştirildi. Kromatografik koşullar: Kolon: Phenomenex, Luna, 5mm, C18 (2), 250 x 4,6 mm. Hareketli faz : 4 mM 1-oktansulfonikasit sodyum tuzu ve 6 mM tetrabutilamonyum hidrojen sülfat içeren 0,1 M amonyum asetat (pH : 5,9) / asetonitril; 1-DN için (62 : 38); 5-CN için (64 : 36). Akış hızı: 1,0 ml / dak. Geri kazanım 1-DN için % 100,65 ve 5-CN için % 100,32 dir. 30 mg NIC / tablette bulunan 1-DN miktarı 209,65 mg / tablettir (% 0,70). 30 mg NIC / tablette bulunan 5-CN miktarı 27,62 mg / tablettir (% 0,09). A quantification method was developed for the impurities of nicergoline, 10-methoxy-6-methylergoline-8b-methanol-5-bromo-3-pyridinecarboxylate (1-DN) and 10-methoxy-1,6-dimethylergoline-8b-methanol-5-chloro-3-pyridinecarboxylate (5-CN) by high performance liquid chromatography and diode array detector. Mobile phase: 0,1 M ammonium acetate solution containing 4 mM 1-octanesulfonicacid sodium salt and 6 mM tetrabutylammonium hydrogen sülfate (pH : 5,9) / acetonitrile (62 : 38) for 1-DN and (64 : 36) for 5-CN. Flow rate: 1,0 ml / min. The recovery was 100,65 % for 1-DN and 100,32 % for 5-CN. The amount of 1-DN in 30 mg NIC / tablet was 209,65 mg / tablet (0,70 %) and the amount of 5-CN in 30 mg NIC / tablet was 27,62 mg / tablet (0,09 %).
