Publication: Sentetik piretroidlerde enantiomerik analiz yöntemlerinin geliştirilmesi
Abstract
SENTETİK PİRETROİDLERDE ENANTİOMERİK ANALİZ YÖNTEMLERİNİN GELİŞTİRİLMESİ Pek çok pestisit ve çevre kirleticileri enantiomerleri içeren kiral bileşiklerdir. Kiral enantiomerlerin ve diasteryomerlerin laboraturlarda ayrım ve seçimi zor olmakla birlikte son yıllarda özellikle biokimya ve farmasötik endüstride organik bileşiklerin ve enantiomerlerin sentez ve ayrımının önemli olması nedeniyle oldukça ilgi görmüştür. Bununla birlikte enantiomerik pestisit analizi hala gelişmektedir. Permethrin sentetik piretroid sınıfındadır. Birkaç kiral merkeze sahip piretroidler bitkilerden elde edildiği gibi sentetik olanları da vardır. Sentetik piretroid insektisitlerin suda yüksek toksititelerinin olması sebebiyle çevresel etkileri önemlidir. Permethrin aktif maddesinde cis izomerler insektisidal olarak aktif olanlardır ve bu nedenle de en toksikleridir. Yüksek Performans Sıvı Kromatografisinde kiral ayırmalarda kiral seçici; ya kiral sabit faz ya da kiral hareketli faz katkısı olarak kullanılır. Bu çalışmada permethrinin ayrım ve izolasyonu için kiral katkı ilavesiyle kiral seçimli kromatografik bir yöntem geliştirmeye çalıştık. Permethrin Agilent HPLC 1100 Series cihazda analiz edildi. Kromatografik analiz Hipersil Siyano, Hipersil Silika ve Nukleosil C-18 kolonlarda siklodekstrin, siklodekstrin her bir kolona hareketli faz katkısı olarak ilave edilerek gerçekleştirildi. Enantiorezolüsyona farklı parametrelerin (hareketli faz kompozisyonu (organik çözücü yüzdesi, tampon türü ve pH), hareketli faz, akış hızı ve siklodekstrin ve siklodekstrin derişimleri) etkileri çalışıldı. Bu farklı parametrelerin ayırmaya etkileri değerlendirildi.
DEVELOPMENT OF THE ANALYSIS METHODS FOR ENANTIOMERIC SYNTHETIC PYRETHROIDS Many pesticides and enviromental contaminants are chiral compounds consisting of enantiomers. While the chiral enantiomers and diastereomers are difficult to seperate and distinguish in the laboratory, in recent years there has been considerable interest in the synthesis and separation of enantiomers of organic compounds, especially because of their importance in the biochemistry and pharmaceutical industry. By the way enantiomeric pesticide analysis is still improving. Permethrin belongs to a class of chemicals called pyrethroids. Pyretroids which has several chiral centers can be derived from plants and also there are sysntetic pyrethroids too. Synthetic pyrehtroid insecticides' enviromental significances are important because of their high aquatic toxicty. In permethrin active ingredient, the cis isomers are the most insecticidally active and therefore the most toxic. A chiral selector can be employed as either a chiral stationary phase or a chiral mobile phase additive in High Performance Liquid Chromatography (HPLC) chiral seperations. In this study, we tried to develop chiral-selective chromatographic methods to seperate and isolate enantiomers of permethrin using chiral mobil phase additive. Permethrin is resolved on an Agilent 1100 Series HPLC. The chromatografic analysis performed on a Hypersil Cyano, Hypersil Silica, and Nucleosil C-18 columns. cyclodextrin, cyclodextrin was added to the mobile phase on each column as the chiral mobile phase additive. The effect on enantioresolution of different parameters (Composition of the mobile phase (percentage of organic solvent, type of buffer and pH), mobile phase, flow rate, and concentration of cyclodextrin, and cyclodextrin) was studied. The effect of these different parameters on the seperation was evaluated.
