Publication: Deneysel omurilik hasarına bağlı cinsel işlev bozukluğunda mezenkimal kök hücre tedavisinin incelenmesi
Abstract
Amaç: Bu çalışmamızda kahverengi yağ dokusu kaynaklı mezenkimal kök hücrelerin (kYD-MKH) omurilik hasarına (OH) bağlı erektil disfonksiyon (ED) tedavisi üzerindeki koruyucu etkileri araştırıldı.Gereç ve Yöntem: Çalışmamızda 250-300 g ağırlığında 24 adet erkek Wistar sıçan kullanıldı. Hayvanlar; kontrol, OH+T ve OH+kYD-MKH tedavili 3 gruba ayrıldı. OH lı gruplarda anestezi altında T5 ve T12 omurları ağırlık düşürme modeli uygulandı. 5 μL içinde 3x105 hücre kYD-MKH’leri 5 dakika süre ile OH’lı bölge içine lokal olarak uygulandı. Dört hafta sonunda tüm sıçanların nörolojik muayeneleri ve ardından intrakavernozal basınç (İKB) ölçümleri yapıldı. Dekapitasyon sonrasında omurilik ve korpus kavernozum doku örnekleri alındı. Dokuda nöronal nitrik oksit sentaz (n-NOS), proto-onkogen protein olan c-FOS ve sinir büyüme faktörü (NGF) tayinleri yapıldı.Bulgular: Omurilik hasarı İKB değerlerini kontrole göre artırdı. Tedavi olarak taşıyıcı uygulanan OH li grupta omurilik ve korpus kavernozum dokusunda NGF ve n-NOS seviyeleri düşük, c-FOS ise yüksek bulundu. Kök hücre tedavisi, bu değerleri geri çevirdi.Sonuç: kYD-MKH tedavisi, omurilik ve korpus kavernozum dokularında hasarı düzeltmiş ve böylece cinsel işlev bozukluğunu da normale getirmiştir.
Aim: The aim of this study is to investigate the protective effects of adipose derived mesenchymal stem cell (AD-MSC) on erectile dysfunction (ED) due to spinal cord injury (SCI). Material and Method: In our study 24 male Wistar rats weighing 250-300 g were used. Animals were divided 3 group as control, SCI+vehicle, and SCI+AD-MSC. In the OH groups, a weight drop model was applied to the T5 and T12 vertebra under anesthesia. 3 x 105 cells in 5 μL AD-MSC were applied for 5 minutes by local transplantation into the OH region. At the end of the four-week period, neurological examinations of all rats and then intracavernosal pressure (ICP) and mean artery pressure (MAP) measurements were performed. Spinal cord and corpus cavernosum tissue samples of the decapitated rats were taken. Neuronal nitric oxide synthase (n-NOS), proto-onkogen protein c-FOS and nerve growth factor (NGF) analyses were performed in the tissues.Results: Spinal cord injury increased ICP values compared to control. Nerve growth factor and n-NOS levels were low and c-FOS was high in the spinal cord and corpus cavernosum of the SCI group treated with carrier as treatment. Stem cell therapy has reversed these values to back.Conclusion: As a result of this study, AD-MSC therapy ameliorated and reversed spinal cord and corpus cevarnosum tissues and restored ED.
