Abstract
Ferulago confusa Velen. Bitkisinin Üzerinde Farmakognozik Araştırmalar Bu çalışmada Ferulago confusa Velen. bitkisinin meyvaları botanik,kimyasal ve biyolojik aktivite bakımlarından incelendi. 21 Temmuz 1989 tarihinde Fatih Ormanından toplanan bitkinin teşhisi Prof.Dr.Ertan Tuzlacı tarafından yapıldı.Bitkinin kurutulmuş örneği M.Ü.Eczacılık Fakültesi Herbaryumunda saklanmaktadır, (MARE 2200) . Bitkinin meyvaları kurutularak toz edildikten sonra önce petrol eteri ile Soxhlet aletinde ekstre edildi ve karışım yoğunlaştırıldı.Kalan materyal etanol ile yine Soxhlet aletinde ekstre edildi ve karışım yoğunlaştırıldı. (PE ekstresi:14,2320 g. , EtOH ekstresi:19,5330 g. ) Ekstreler ince tabaka kromatografisi ile tetkik edildi. Çalışmanın etanol ekstresi üzerinde yürütülmesine karar verildi. Silika jel ile hazırlanmış sütuna yoğunlaştırılmış etanol ekstresi yerleştirildikten sonra petrol eteri ile sütunun elüe edilmesine başlandı ve artan oranlarda kloroform,etilasetat ve etanol ile devam edildi. Alınan fraksiyonlar yoğunlaştırıldıktan sonra ince tabaka kromatografisi ile kontrol edilerek benzer olanlar birleştirildi. Elde edilen fraksiyonların kirlilikten kurtarılmaları için preparatif ince tabaka kromatografisi uygulandı. Kumarin tipi olduğu düşünülen bileşiklerin yapısı ultraviole (UV),infrared (IR),nükleer manyetik rezonans (1H-NMR) yöntemleri ve şahit maddelerle yapılan kromatografik karşılaştırmalar ile saptandı. F.confusa bitkisinin meyvalarından elde edilen kumarin tipi bileşikler olarak isoimperatorin,marmesin senecioat ve osthenol saptandı. Ayrıca bitkiden elde edilen maddelere ve bitkinin PE ve EtOH ekstrelerine Brine Shrimp (Artemia Salina) yöntemi uygulandığında bunların aktif oldukları görüldü. Yapılan antibakteriyel ve antifungal etki çalışmalarında F. confusa meyvalarının PE ve EtOH ekstrelerinin bazı bakteri ve mayalara etkin olduğu saptandı. Sonuç olarak , kumarin tipi bileşikler ve biyolojik aktivite bakımından incelenen F. confusa bitkisinin meyvalarında 3 tane kumarin tipi bileşik ve 1 tane triterpen yapıda olduğu düşünülen bileşik elde edildi.Ayrıca bitkinin meyvalarının antibakteriyel ve antifungal etkiye sahip oldukları tespit edildi.
Pharmacognosic Researches on Ferulago confusa Velen. In this research,the fruits of Ferulago confusa Velen. have been investigated chemically and from the biological activity point of view. The plant was collected from Fatih Forest , İstanbul on July 21st,1989. The identification of the plant is made by Prof.Dr.Ertan Tuzlacı and the sample is being kept in the M.U.Faculty of Pharmacy Herbarium (MARE 2200). The fruits of the plant were powdered after they were dried ,and were first extracted with petroleum ether in the Soxhlet apparatus. The mixture was evaporated and then the plant which was extracted with petroleum ether ,was extracted again with ethanol in the Soxhlet apparatus.The second mixture was also evaporated. (PE extract :14,2320 g. , EtOH extract:19,5330 g.) The extracts which were obtained as results of these extractions were investigated by using thin layer chromatography.As a result , it was decided to work on the ethanolic extract because of the coumarin type compounds found in it. For further research,the dried ethanol extract was applied to the column prepared with silica gel and then this column was started to be eluted first with petroleum ether and then, chloroform, ethyl acetate and ethanol respectively, in increasing volumes. The concentrated fractions obtained from the column chromatography ,were applied to thin layer chromatography and similar fractions were combined. Preparative thin layer chromatography was used in order to purify the fractions. The structure of the obtained coumarin type compounds are found by ultraviolet (UV),infrared (IR),nuclear magnetic resonance (1H-NMR) spectroscopies and also by chromatographic comparisons with the standart substances. As coumarin type compounds found in the fruits of Ferulago confusa ,there are isoimperatorin,marmecin senecioat,and osthenol. Also when the Brine Shrimp (Artemia Salina) method is applied to the coumarin compounds and to petroleum ether and ethanolic extracts,it was seen that all of them were quite active . In further studies on these extracts about their antibacterial and antifungal activities , the ethanolic and the petroleum ether extracts of F.confusa were found to have both antifungal and antibacterial activities against some bacteria and yeasts. As a result ,from the fruits of Ferulago confusa which have been investigated both for their coumarin type compounds and for its biological activity, 3 coumarin type compounds and 1 compound which is considered to be a triterpene in structure, are obtained and the fruits of the plant are found to have antibacterial and antifungal effects.
