A., Özbek S., Cinar M., Alpsoy E., et al.Behçet’s disease is a complex inflammatory vasculitis with a broad spectrum of clinical manifestations. The purpose of this study was to investigate the genetics underlying specific clinical features of Behçet’s disease in a group of patients with > 20 years of follow up. A total of 436 patients with Behçet’s disease from Turkey were studied. Genotyping was performed using the Infinium ImmunoArray-24 BeadChip. After imputation and quality control measures, logistic regressions adjusting for sex and the first five principal components were performed for each clinical trait using a case-case genetic analysis approach. A weighted genetic risk score was calculated for each clinical feature. Genetic association analyses of previously identified susceptibility loci in Behçet’s disease revealed a genetic association between ocular lesions and HLA-B/MICA (rs116799036: OR=1.85, 95% CI=1.35-2.52, p-value=1.1x10-4 ). The genetic risk score was significantly higher in Behçet’s disease patients with ocular lesions compared with those without ocular involvement, and is explained by the genetic variation in the HLA region. New genetic loci predisposing to specific clinical features in Behçet’s disease were suggested when genome-wide variants were evaluated. The most significant associations were observed in ocular involvement with SLCO4A (rs6062789: OR=0.41 (95% CI=0.30-0.58), p-value=1.92x10-7 ), and neurological involvement with DDX60L (rs62334264: OR= 4.12 (95% CI 2.34 to 7.24), p-value = 8.85x10-7 ). Our results emphasize the role of genetic factors in predisposing to specific clinical manifestations in Behçet’s disease, and might shed additional light into disease heterogeneity, pathogenesis, and variability of Behçet’s disease presentation across populations.Publication Metadata only Comparison of some Macro and Micro Mutrients in Tomato Fruits Exported Abroad from Turkey According to the Growth Conditions(2022-05-18) ÖZYİĞİT, İBRAHİM İLKER; Özyiğit İ. İ., Can H., Uyanik O., Yalcin I., Demir G.Tomato is one of the most widely cultivated (conventionally, greenhouse or soilless agriculture) and important agricultural products among the members of the Solanaceae family as well as all vegetables. It also plays a significant role in healthy dietary nutrition with its ingredients such as lycopene which gives its natural color and is an abundant free radical scavenger, anthocyanin and vitamin contents. Soil properties and fertilization is a significant parameter for yield, mineral content and fruit quality of tomato. Turkey is one of the largest tomato producers in the world after China, India and the USA. Aim of this study aim is to determine the some macro and micro elements of edible tomato fruits from various origins of a significant producer country (Turkey) and also to compare the differences with tomatoes grown with different methods both in Turkey and other tomato producing countries. One of the most important steps in inductively coupled plasma mass spectroscopy (ICP-MS) technology is sample preparation. For the parts of each plant sample, the EPA 3051A analytical method was applied using nitric acid in Teflon cells. The prepared samples were thawed in a CEM Mars 5 microwave. The concentrations (in mg kg-1 ) of, Ca, Cu, Fe, K, Mg, Mn, Na and Zn in the dried samples have been determined by employing ICP- MS (Agilent 7700 Series). The order of locations with respect to the element concentrations from the highest to the lowest is Mediterranean and Southeastern Anatolia Regions>Marmara Region>Aegean Region>West part of Central Anatolia Region. Based on available elemental concentrations from the study conducted in major Turkish tomato production centers, we can extrapolate ideas about the necessary fertilization programs from mineral nutrients contents of tomato fruits. Statistically significant differences were obtained in mineral nutrients from greenhouse and field samples except for Mg, Mn, and Zn due to the fact that intensive regimes of production and fertilization were practiced in greenhouses. In terms of mineral nutrient content, tomato samples collected from different parts of Turkey showed close similarity with previous studies except for K content. K content was detected to be low in comparison with previous results. The K fertilization may have been ignored in the major tomato production sites of Turkey in contrast to other parts of the world. Z., MİDİ İ.Objective: This study aimed to compare the frequency of admission to hospital in patients with acute ischemic stroke before the COVID 19 pandemic, during the pre-vaccination period, and after the start of vaccination for COVID 19, and to evaluate the time window period between symptom onset to door time, door to CT scan time, door to needle time, and door to puncture time. In addition, it aimed to investigate the effects of the COVID-19 pandemic on the admission, evaluation, and initiation of acute treatment of patients with acute ischemic stroke. Methods: Patients presenting with acute ischemic stroke between March 2019-December 2019 (pre-pandemic), March 2020-December 2020 (pre-vaccination pandemic period), and March 2021-December 2021 (post-vaccination pandemic period) were included in the study. NIHSS was calculated by accordance with the neurological examination findings of the patients, cranial CT for the exclusion of bleeding and CT angiography images for the large vessel occlusions were performed, and the vital signs of the patients were recorded. IV tPA treatment was applied within the first 4.5 hours, and mechanical thrombectomy (MT) was performed in patients with large vessel occlusion. Results: Three hundred nineteen patients were included in the study. The times from symptom onset to emergency admission and from symptom onset to CT scan were found to be similar in all periods. The time from symptom onset to examine by a neurologist was found to be significantly longer in the vaccination period compared to the pandemic period. It was observed that the time from the door to needle time and the time from examine by a neurology doctor to needle time was statistically significantly shorter during the pandemic period (p<0.05). Conclusion: In our study in a tertiary hospital in Turkey, it was determined that the number of patients who was admitted with acute stroke clinic during the pandemic period was similar to other years and there was no delay in the initiation of treatment during the pandemic period. Door to needle times, as well as the time taken by the neurologist to examine and initiate IV-tPA treatment, were found to be shorter in the pre-vaccination pandemic period than in the pre-pandemic and post-vaccination periods.Publication Open Access Determination of the Significance of the Most Effective Nutrients on Lipase Production from Cryptococcus albidus D24(2022-06-01) PİNAR, ORKUN; KAZAN, DİLEK; Uras, Abdussamed; Uras A., Pinar O., Kazan D.In this study, the optimization of the medium components used for the production of lipase enzyme from Cryptococcus albidus D24 was performed using the Plackett-Burman statistical design method (PBD), and the most important nutrients affecting the production of lipase enzyme from D24 strain were determined as the first step. According to PBD, the highest lipase activity (19.34 U/ml/min) was obtained with medium including Tween 80 (X2) 2.5% (v/v), and (g/L) Peptone (X4) 8.0, Yeast Extract (X6) 7.5, Beef Extract (X7) 7.5, Malt Extract (X8) 7.5, NH4Cl (X9) 6.0, NaNO3 (X10) 1.5, (NH4)NO3 (X12) 6.0, (NH4)HCO3(X13) 6.0, MgSO4.7H2O (X15) 1.0, and KH2PO4 (X16) 2.0 at the end of 144 h cultivation. Regarding the concentration effect (CE) values obtained from PBD, NH4Cl (CE=7.1587), olive oil (CE=3.5544), (NH4)HCO3 (CE=3.0747), and tryptone (CE=2.1427) were evaluated as the more effective nutrients among the sixteen compounds studied. After that, the optimum concentrations of these effective compounds were experimented with Response Surface Methodology (RSM). Experimental results showed that the medium containing olive oil (X3) 1.5% (v/v), and (g/L) tryptone (X5) 3.0, NH4Cl (X9) 7.5, and (NH4)HCO3 (X13) yielded maximum lipase activity (12.03 U/ml/min) compared to other studied compounds. Although the maximum lipase activity obtained with RSM methodology was lower than that obtained by PBD, the cost of the nutrients used to produce one-unit enzyme is 0.104 Euro in the PBD, while only 0.0277 Euro is spent in RSM. In other words, the production of lipase using compounds coded X3, X5, X9, and X13 provides a cost-effective process.Publication Open Access Exploring the anticancer effects of brominated plastoquinone analogs with promising cytotoxic activity in MCF-7 breast cancer cells via cell cycle arrest and oxidative stress induction(2022-06-01) YILMAZ GÖLER, AYŞE MİNE; YILMAZ, BETÜL; Jannuzzı A. F.Plastoquinone analogs are privileged structures among the known antiproliferative natural product-based compound families. Exploiting one of these analogs as a lead structure, we report the investigation of the brominated PQ analogs (BrPQ) in collaboration with the National Cancer Institute of Bethesda within the Developmental Therapeutics Program (DTP). These analogs exhibited growth inhibition in the micromolar range across leukemia, non-small cell lung cancer (EKVX, HOP-92, and NCI-H522), colon cancer (HCT-116, HOP-92), melanoma (LOX IMVI), and ovarian cancer (OVCAR-4) cell lines. One brominated PQ analog (BrPQ5) was selected for a full panel five-dose in vitro assay by the NCI’s Development Therapeutic Program (DTP) division to determine GI50, TGI, and LC50parameters. The brominated PQ analog (BrPQ5) displayed remarkable activity against most tested cell lines, with GI50values ranging from 1.55 to 4.41 µM. The designed molecules (BrPQ analogs) obeyed drug-likeness rules, displayed a favorable predictive Absorption, Distribution, Metabolism, and Excretion (ADME) profile, and an in silico simulation predicted a possibleBrPQ5interaction with proteasome catalytic subunits. Furthermore, the in vitro cytotoxic activity ofBrPQ5was assessed, and IC50values for U-251 glioma, MCF-7 and MDA-MB-231 breast cancers, DU145 prostate cancer, HCT-116 colon cancer, and VHF93 fibroblast cell lines were evaluated using an MTT assay. MCF-7 was the most affected cell line, and the effects ofBrPQ5on cell proliferation, cell cycle, oxidative stress, apoptosis/necrosis induction, and proteasome activity were further investigated in MCF-7 cells. The in vitro assay results showed thatBrPQ5caused cytotoxicity in MCF-7 breast cancer cells via cell cycle arrest and oxidative stress induction. However,BrPQ5did not inhibit the catalytic activity of the proteasome. E., KOCA A.Two different zinc phthalocyanines bearing different numbers of 2-naphthoic acid anchoring groups at the peripheral positions were synthesized and characterized with UV–Vis, proton nuclear magnetic resonance (1H NMR), fouirer transform infrared (FT-IR), and matrix-assisted laser desorption/ionization mass (MALDI-TOF MS) spectroscopy. Then their electrochemical, and spectroelectrochemical performances were investigated to predict their suitability of them as photosensitizers in dye-sensitized solar cells (DSSC). In the voltammetric analysis results, [2,9,16-Tri-(4-carboxyethylphenoxy)-23-(4-[6-carboxy-2-naphtoxy]) substituted zinc(II) phthalocyanine (ZnPc(3)) and [2,9,16,23-tetra-(4-(6-carboxy-2-naphthoxy) substituted zinc(II) phthalocyanine (ZnPc(4)) illustrate similar electron transfer processes. The substituent environments of the complexes slightly influenced the position and reversibility of the redox couples. Redox processes of ZnPc(3) bearing unsymmetrical carboxyethylphenoxy and 2-naphthoic acid anchoring groups slightly shift towards the positive potentials concerning ZnPc(4) bearing symmetrical 2-naphthoic acid substituents. Peak positions of both complexes reflecting the highest occupied molecular orbital (HOMO) and the lowest unoccupied molecular orbital (LUMO) positions indicate the suitability of the complexes for the efficient charge carrier transferring from these photosensitizers to the semiconductor and redox mediator in the DSSC. Pc-based spectroelectrochemical responses of the complexes supported the HOMO and LUMO positions for both neutral and electrogenerated ZnPc species. DSSC responses indicated that ZnPc(3), which has the asymmetric carboxyethylphenoxy and 2-naphthoic acid substituents, gave higher DSSC efficiency with short-circuit photocurrent density (JSC) (9.54 mA cm−2), open circuit potential (VOC) (697 mV), fill factor (FF) (51%), incident monochromatic photon-to-current conversion efficiency (IPCE) (51%), and power conversion efficiency (ƞ) (3.4%) parameters concerning ZnPc(4) bearing symmetric 2-naphthoic acid anchoring groups.Publication Metadata only Kripto paraların karapara aklama yöntemi olarak kullanılması(2021-04-01) ÇAKIR, KERİM; Balcı M., Çakır K.Ekonomi ve finans dünyasındaki gelişmelere paralel olarak, suç işleyerek gelir elde eden kimseler karaparayı kripto para birimleri aracılığıyla aklamaya başlamıştır. Bu çalışmanın konusunu da karapara aklama yöntemlerinden biri haline gelen kripto paralarla karapara arasındaki ilişki oluşturmaktadır. Suç işlenerek elde edilen malvarlığı değerlerine bugün itibariyle sanal malvarlığı olarak kripto paralar da dahildir. Kripto para birimlerinin anonim ve küresel olması, merkezi bir otoriteye tabi olmaması karapara aklamayı kolaylaştırıcı özellikler olarak karşımıza çıkmaktadır. These materials are manufactured using hot forging or extrusion techniques, according to the application for different purposes. In this work, the metallographic and mechanical characteristics of three environmentally friendly brass alloys - CuZn40Pb2 (CW617N), CuZn38As (CW511L) and CuZn21Si3P (CW724R) - extruded and multi-directionally hot forged have been examined. By focusing on the flat bottom drilling operation, the effects of the process and type of alloying have been investigated. The examination has been accomplished on experimental data about tensile strength, impact fracture energy, cutting forces, entrance and exit burr diameters, as well as hole quality. Y.The rise of machine learning (ML) has recently buttressed the efforts for big data-driven precision oncology. This study used ensemble ML for precision oncology in breast cancer, which is one of the most common malignancies worldwide with marked heterogeneity of the underlying molecular mechanisms. We analyzed clinical and RNA-seq data from The Cancer Genome Atlas (TCGA) (844 patients with breast cancer and 113 healthy individuals) and the Molecular Taxonomy of Breast Cancer International Consortium (METABRIC) (1784 patients with breast cancer and 202 healthy individuals). We evaluated six algorithms in the context of ensemble modeling and identified a candidate mRNA diagnostic panel that can differentiate patients from healthy controls, and stratify breast cancer into molecular subtypes. The ensemble model included 50 mRNAs and displayed 82.55% accuracy, 79.22% specificity, and 84.55% sensitivity in stratifying patients into molecular subtypes in TCGA cohort. Its performance was markedly higher, however, in distinguishing the basal, LumB, and Her2+ breast cancer subtypes from healthy individuals. In overall survival analysis, the mRNA panel showed a hazard ratio of 2.25 (p = 5 x 10(-7)) for breast cancer and was significantly associated with molecular pathways related to carcinogenesis. In conclusion, an ensemble ML approach, including 50 mRNAs, was able to stratify patients with different breast cancer subtypes and differentiate them from healthy individuals. Future prospective studies in large samples with deep phenotyping can help advance the ensemble ML approaches in breast cancer. Zamanla farklılık gösteren toplumsal anlamların yaratılmasında, “güncel” oluşumları betimler. Dolayısıyla bireysel ve toplumsal da olsa dünyaya doğrudan yansıyan anlam değerlerinin inşasında, iletişimin bir mihenk taşı olduğu söylenilebilir. İnsanın, anlam değerlerini inşa ederken anlatım yollarından ve biçimlerinden biri olan sanat, iletişim ihtiyacı ile doğan ve gelişen bir yaratım süreci şeklinde nitelendirilebilir. Aynı zamanda dışavurumsal bir ifade olarak sanatın, estetik kimliğinden farklı olarak zamanla evrensel bir boyut kazanması, iletişim aracılığıyla mümkün olabilmiştir. M., YALÇIN S. S., Koller D., Boran P.BackgroundThe direct and indirect effects of the COVID-19 pandemic on children and youth people are well documented. Little is known about children\"s experiences of the pandemic in their own words. We aimed to explore Turkish children\"s experiences and views about the pandemic, across private and public educational systems. MethodsSix focus group discussions were conducted online with 30 children aged between 7 to 18 years in 2021. Children were enrolled through snowball sampling technique according to developmental age groups. A thematic analysis was conducted. ResultsWe identified five themes: Compliance with public health measures about preventing the spread of COVID-19, changes in daily routines, emotional responses to the pandemic, distance education, and adaptive responses. Overall, children were well informed and receptive to the public health preventive measures with the exception of older children\"s tendency to disregard physical distancing with friends. They reported frustration with those who did not comply with measures and believed that the authorities should strictly enforce public health requirements. Emotional responses comprised fear of family members and themselves being infected, anxiety produced by the uncertainty of the virus, and the loss of social support. There was a perceived disparity between students from public and private schools regarding academic self-competence. Positive aspects of the pandemic included positive interactions with family members and the acquisition of new hobbies. Although most children acknowledged the support of their parents to deal with challenges, children of health workers were particularly isolated in terms of emotional support. ConclusionOur findings offer additional insights and validate previous research on the negative and positive effects of COVID-19 from the child\"s perspective across private and public educational systems. Despite a broad range of antimicrobial drugs today, there are certain obstacles associated with the use of antimicrobial agents such as drug resistance and toxicity. Thus, medicinal chemists concentrate on designing novel antimicrobial drugs. In the search for new antimicrobial agents; 1,3,4-oxadiazole compounds have come forward due to their hydrolytic stability, good chemical and thermal stability. In the scope of this work, 2-(6-chloropyridin-3-yl)-5-(substitutedphenyl)-1,3,4-oxadiazole (4a-4i) were synthesizedusing 6-chloro-N\"-(substitutedbenzoyl)nicotinohydrazide (3a-3i). These compounds were screened for their antimicrobial activities against as gram-positive bacteria S. aureus, E. faecalis, as gram-negative bacteria E. coli, P. aeruginosa, as yeast C. parapsilosis, C. albicans, C. glabrata. C.Background: Many studies have discussed the effects of serum vitamin D deficiency in severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) patients. This study aimed to investigate the relationship between SARS-CoV-2 infection severity and free vitamin D (FVD) and bioavailable vitamin D (BAVD) levels in children. Methods: A prospective case–control study design was used. Participants were divided into three groups based on the World Health Organization COVID-19 Clinical Progression Scale. Serum 25-hydroxyvitamin D (ng/mL), albumin (g/L), and vitamin D binding protein (ng/mL) levels were evaluated to investigate the relationship between disease severity and FVD and BAVD levels. Results: In total, 82 participants were included in the study. Of those, 24.4% were uninfected (n=20), 50% had a mild case of SARS-CoV-2 (n=41), and 25.6% had a moderate case (n=21). There was a statistically significant difference in FVD and BAVD levels between the groups (p=0.026). Median FVD (p=0.007, Cohen's d=0.84) and BAVD (p=0.007, Cohen's d=0.86) levels were significantly higher in the mild group compared to the moderate group. FVD and BAVD metabolites were moderately positively correlated with lymphocyte counts (FVD: r=0.437, p<0.001; BAVD: r=0.439, p<0.001). Conclusions: This is the first study to demonstrate a relationship between SARSCoV-2 symptom severity and FVD and BAVD levels. The relationship between FVD and BAVD levels and lymphocyte counts could play an important role in symptom severity and should be evaluated in further studies.
