Polymorphonuclear leukocytes (PMNs) were isolated with ficoll-hypaque gradient centrifugation method. MPO activity was assayed with modified o-dianisidine, GSH by Ellman's and MDA levels by Beuge's method. PMN functions and MDA levels of patients significantly decreased when compared with healthy volunteers. Ciprofloxacin significantly increased PMN functions, MPO activity and MDA levels of both groups. There are many reports about the traditional use and highly bioactive compounds of these plants. Therefore, it is fundamental to study in detail the chemical composition and biological activities of Achillea species. Thus, the objective of this study was to determine for the first time the effects of methods/solvents on biological activities (antioxidant, antimicrobial, and anti-urease) of Achillea nobilis L.subsp. neilreichii (Kerner) Formanek 15 extracts obtained from different extraction methods/solvents and then to analysis the phenolic contents of the active extracts. The phenolic contents in ethyl acetate and ethanol extracts, which showed strongest phenolic contents and antioxidant activity were analysed separately by high-performance liquid chromatography with diode array detection and were confirmed by liquid chromatography with quadrupole time-of-flight mass spectrometry using targeted mass spectra techniques with accurate mass. The active phenolic compounds were identified as dicaffeoylquinic acid, luteolin-7-O-glucoside, orientin, vitexin, luteolin, quercetin-methyl-ether, isorhamnetin-glucoside in ethyl acetate extract; chlorogenic acid luteolin-7-O-glucoside, quercetin-methyl-ether, luteolin, orientin, vitexin, dicaffeoylquinic acid, apigenin-6,8-di-C-glucoside, luteolin hexoside hexoside in ethanol extract. As a result of this study, dicaffeoylquinic acid, isorhamnetin-glucoside, chlorogenic acid, and apigenin-6,8-di-C-glucoside compounds were analysed for the first time in Achillea nobilis L. subsp. neilreichii (Kerner) Formanek. Therefore, it is thought that ethanol and ethyl acetate extracts can be used as a natural source in the future for food and pharmaceutical industry.Publication Metadata only Probiyotik ürünlerden izole edilen laktik asit bakterilerinin ultrasonikasyon sonrası antimikrobiyal etkinliğinin belirlenmesi(2023-08-15) RAYAMAN, PERVİN; RAYAMAN, ERKAN; BİNGÖL ÖZAKPINAR, ÖZLEM; Bolat G., RAYAMAN P., RAYAMAN E., BİNGÖL ÖZAKPINAR Ö.Giriş ve Amaç: Yeterli miktarlarda alındığında intestinal sistemi düzenleyerek konakçı sağlığı üzerine faydalı etkilere sahip canlı mikroorganizma suşları olarak tanımlanan probiyotiklerin bağırsak epiteliyal bariyer fonksiyonunu iyileştirdiği, immünomodülatör yanıtları düzenlediği, toksin reseptörlerini bozduğu, inhibitör maddeler ürettiği, çeşitli mekanizmalar kullanarak patojen mikroorganizmaları inhibe ettiği bilinmektedir. Dolayısıyla çalışmamızda, çeşitli probiyotik ürünlerden izole edilen laktik asit bakterilerinin ultrasonikasyon sonrası antimikrobiyal etkinliğinin belirlenmesi hedeflenmiştir. Gereç ve Yöntem: Çalışmamızda; Lactobacillus fermentum ATCC (Amerikan Tipi Kültür Koleksiyonu) 14931, ev yapımı kefirden izole edilen Lactobacillus kefiri ve eczane preparatından izole edilen Lactobacillus rhamnosus türü mikroorganizmaların in vitro koşullarda Escherichia coli ATCC 11229, Staphylococcus aureus ATCC 25922, Pseudomonas aeruginosa ATCC 27853, Salmonella typhimurium ATCC 14028, Klebsiella pneumoniae ATCC 4352 bakterileri ve Candida albicans ATCC 10231 mayası üzerine olan antimikrobiyal aktivitesi spot on lawn yöntemiyle belirlenmiştir. Çalışmamızda bulunan mikroorganizmalar Marmara Üniversitesi Farmasötik Mikrobiyoloji Anabilim Dalı’nın kültür stok koleksiyonundan temin edilmiştir. Çalışmamızdaki Laktobacillus türleri 15 ml’lik Falcon tüplerine aktarılmış ve MRSA (Mann Rogosa Sharpe Agar) içeren petrilere damlatılarak inkübe edilmiştir. İnkübasyon sonrası probiyotik mikroorganizmalara ultrason cihazında ms 73 prob, %50 genişlik ayarlanarak; 6 dakika boyunca ultrasonik homojenikatör uygulanmış ve MRSA besiyeri içeren petrilerin yüzeyine 5 μL damlatılarak inkübe edilmiştir. İnkübasyon sonrası probiyotik mikroorganizmaların bulunduğu petri plaklarının üzerine McFarland 0,5 standart bulanıklığına ayarlanmış bakteriler ve McFarland 1 standart bulanıklığına ayarlanmış maya süspansiyonu içeren yumuşak agar dökülmüş ve petriler bakteriler için 24 saat, maya için ise 48 saat süre ile 37°C’de inkübe edildikten sonra oluşan inhibisyon zonlarının çapı elektronik kumpas yardımı ile mm olarak ölçülüp kaydedilmiştir. Bulgular ve Sonuç: Çalışmada yer alan tüm probiyotik mikroorganizmalar ultrasonikasyon sonrası Escherichia coli ATCC 11229, Staphylococcus aureus ATCC 25922, Pseudomonas aeruginosa ATCC 27853, Klebsiella pneumoniae ATCC 4352 patojenlerine karşı başlangıç değerine göre daha fazla antimikrobiyal etki göstermiştir. Lactobacillus fermentum ve Lactobacillus kefiri türlerinin Candida albicans’a karşı başlangıçta etki edemezken ultrasonikasyon sonrası inhibisyon zonu oluşturduğu tespit edilmiştir. Sonuç olarak ultrasonikasyon işleminin probiyotik ürünlerden izole edilen laktik asit bakterilerinin antimikrobiyal etkinliğini arttırdığı gösterilmiştir. Microparticles were prepared by using levan polysaccharide produced by a halophilic bacterium Halomonas smyrnensis AAD6(T). To optimize efficiency of encapsulation process by precipitation method, three parameters were studied: drug and polymer concentrations and preparation rotating speed. The particles were characterized in vitro. The size of levan microparticles was changed between 0.404 mu m and 1.276 mu m. The surface charge was detected between +4.1 mV and +6.5 mV. The highest drug encapsulation capacity of the system was 74.7% and was depending on the polymer concentration. In dissolution studies, initial burst effect around 10-20% from all the formulations was observed and then the release was slowed down and continued at a constant level. In vitro antibiotic release from the microparticles was controlled with the drug carrier system and release fit to Higuchi kinetic model. All the released samples collected at different time intervals during dissolution studies have exhibited intrinsic bactericidal activity against Bacillus subtilis ATCC 6633. WST-1 cell proliferation and viability studies showed that VANCO-loaded levan microparticles at concentrations between 100 mu g/mL and 1000 mu g/mL were nontoxic to L929 cells. As conclusion, levan microparticulate system could be a potential carrier of antibiotic drugs such as VANCO.Publication Metadata only In vitro and In vivo biological activities and phenolic characterization of Thymus praecox subsp. skorpilii var. skorpilii(SPRINGER, 2019) RAYAMAN, ERKAN; Taskin, Turgut; Cam, Muhammet Emin; Taskin, Duygu; Rayaman, ErkanThymus species are used as food and spices in several Mediterranean and other countries as well as for medical purposes on account of their biological properties. The extraction procedures and solvents are an important step in processing bioactive constituents from the plant materials. Therefore, the aim of this study was to evaluate in vitro antioxidant, anticholinesterase, antimicrobial, anti-urease and in vivo anti-inflammatory activities of Thymus praecox subsp. skorpilii var. skorpilii using a variety of extracts. In addition, the phenolic characterization of the fraction methanol extract, which showed the highest phenolic contents and the strongest antioxidant and anticholinesterase activity were performed by HPLC-DAD and LC-MS/MS techniques. In the present study, the fraction methanol and direct methanol extracts exhibited strong DPPH, ABTS, and ferric reducing activity and strong antibacterial activity against Staphylococcus epidermidis strains. The fraction chloroform and fraction petroleum ether extracts showed the strongest anti-urease activity and also, the petroleum ether extract exhibited strong antibacterial activity against the Staphylococcus aureus strain. The fraction methanol and direct methanol extracts showed in vivo anti-inflammatory activity very similar to each other. As a result of this study, chlorogenic acid, luteolin-7-O-glucoside, 3-O-feruloylquinic acid, quercetin-3-O-hexoside, and apigenin-7-O-glucuronide compounds were analyzed for the first time in the methanol extract of the plant.
