Abstract
AMAÇ: Çalışmamızda, sağlıklı ve deneysel olarak diyabet oluşturulmuş sıçanlarda, hyalüronik asitin (HA) gargara
formunun topikal olarak uygulamasının palatinal yara iyileşmesi üzerine etkilerinin klinik ve histomorfometrik
olarak incelenmesi amaçlandı.
GEREÇ VE YÖNTEM: Çalışmamızda 60 adet erkek Wistar albino sıçan kullanıldı. Sıçanlar sağlıklı ve deneysel
diyabet oluşturulan 2 ana gruba ayrıldı. Rastgele seçilen 30 sıçana intraperitoneal olarak 110 mg/kg nikotinamid
uygulanmasından 15 dakika sonra distile su içinde çözündürülen streptozotosin tek doz 65mg/kg intraperitonal
olarak enjekte edilerek diyabet oluşturuldu. Her iki gruptaki sıçanların palatinal bölgelerinin ortasında 5 mm
çapında kemiğe kadar mukozal defekt oluşturuldu. Takiben her iki gruptan 6’şar sıçan 0. günde sakrifiye edildi.
Ardından sağlıklı ve diyabetik gruplar kendi içlerinde HA ve salin uygulanan 2 alt gruba ayrıldı. Yara bölgesine
günde iki kez 1 ml HA veya salin uygulandı. Defektler oluşturulduktan 0, 7 ve 14 gün sonra, yara alanları
(YA) fotoğraflar üzerinden ölçüldü. Örneklere rutin histolojik doku takibi uygulandıktan sonra alınan kesitlere
hematoksilen-eozin boyaması yapıldı ve epitelyal kenarlar arasındaki ortalama mesafe (EKM) histomorfometrik
olarak belirlendi.
BULGULAR: Ortalama YA tüm gruplarda zamanla istatistiksel olarak azaldı (p<0.05). Sağlıklı ve diyabetik HA
grupları, sağlıklı ve diyabetik salin grupları ile karşılaştırıldığında YA’ da önemli bir azalma gözlendi (p<0.05). EKM,
diyabetik salin grubu hariç tüm gruplarda başlangıca kıyasla önemli ölçüde azaldı (p<0.05). Sağlıklı ve diyabetik
HA grupları, sağlıklı ve diyabetik salin gruplarına kıyasla EKM ‘de önemli bir azalma gösterdi (p<0.05).
SONUÇ: HA’nın topikal uygulamasının, diyabetik sıçanların palatinal bölgelerinde oluşturulan yaralarda epitelyal
kapanmayı hızlandırdığı görüldü.
AIM: The aim of this study was to to assess clinically and histomorphometrically the effect of topically applied hyaluronic acid (HA) on palatal wound healing in nondiabetic (ND) rats and in rats with diabetes induced. MATERIAL AND METHOD: 60 Wistar albino male rats were divided into ND and diabetic (D) groups. Diabetes was induced to 30 randomly selected rats by administration of freshly prepared solution of streptozotocin dissolved in distilled water at a dose of 65 mg/kg intraperitoneal, 15 min after intraperitoneal administration of 110 mg/kg body weight nicotinamide. Excisional wounds, 5 mm diameter, were made in the centre of the palate. After wound induction, 6 animals from each group were sacrificed. Then, the ND and D groups were further subdivided into saline and HA groups. 1 ml of HA or saline was delivered to the wound twice a day, The wound areas (WA) were measured photographically at 0, 7 and 14 days after wound creation. After routine histological tissue follow-up was applied to the samples, hematoxylin&eosin staining was performed on the sections taken and the mean distance between epithelial margins (EM) were determined histomorphometrically. RESULTS: The mean WA decreased statistically with time in all groups (p<0.05). The both HA groups showed a significant reduction in the WA than the both saline groups (p<0.05). The mean distance between EM decreased significantly compared to baseline in all groups (p<0.05) except the diabetic saline group. ND and D HA groups showed a significant reduction in EM compared to the ND and D saline groups (p<0.05). CONCLUSION: It can be concluded that topical application of HA accelerated epithelial closure in palatinal wound healing in rats with diabetes induced.
