K., Çilingir-Kaya Ö. T., Şener A.AbstractThe prothrombotic and inflammatory state plays a significant role in the occurrence of cardiovascular complications in type 2 diabetes mellitus. In this study, the antidiabetic, anti-inflammatory, and antiplatelet potentials of the extracts obtained fromRibes rubrumwere investigated. The antidiabetic, anti-inflammatory, and antioxidant activities of the ethanol and water extracts ofR. rubrumwere evaluated by in vitro methods. The total phenolic and flavonoid contents were also determined. The experimental diabetes model in rats was induced with streptozotocin (STZ). After hyperglycemia occurred, the ethanol extracts ofR. rubrum(RRE, at 100mg/kg and 500mg/kg doses) were administered to the treatment groups for 14days. Blood glucose, lipid profile, plasma, and pancreas tumor necrosis factor-α (TNF-α) levels were determined and compared at the end of the experiments. P-selectin levels and mitochondrial membrane polarization (MMP) of platelets were also measured. In vitro study, the RRE showed potent anti-inflammatory activity. Administration of RRE (at 100mg/kg doses) to diabetic rats lowered blood glucose level insignificantly. The results showed that there was an increment in levels of TNF-α in plasma and pancreas tissue of the diabetic group compared to the control group.R. rubrumextract regulated and normalized their levels in plasma and pancreatic tissue. RRE at both doses significantly decreased platelet P-selectin levels and prevented STZ-induced loss of MMP in platelets. The results of current research indicate that RRE extract has potent anti-platelet and anti-inflammatory effects and may be beneficial in preventing diabetic complications.Practical applicationsHyperglycemia causes dyslipidemia, advanced oxidative stress, platelet activation, and inflammation in diabetes mellitus. Plants with various medicinal properties are of worldwide interest for the treatment of diseases due to their biological activities. S. , ŞENKARDEŞ İ., BİTİŞ L.In this study, in vitro biological activities and total phenol/flavonoid contents of methanol extract (ISM) and its hexane (ISH), chloroform (ISC), ethyl acetate (ISEA) and aqueous methanol (ISAM) fractions obtained from aerial parts of Inula salicina were investigated. ISEA showed the highest antioxidant activity against DPPH and ABTS radicals with an IC50 value of 0.014 mg ml-1 for both assays. ISEA exhibited a good anti-inflammatory activity with an IC50 value of 0.060 mg ml-1 . ISEA was found to exhibit a moderate level of antidiabetic activity against α amylase enzyme with an IC50 value of 0.290 mg ml-1 . ISEA and ISM presented low and moderate inhibitory activity against acetylcholinesterase and butyrylcholinesterase enzymes with IC50 values of 0.577 and 0.279 mg ml-1 , respectively. ISC with MIC values of 78 and 156 µg ml-1 displayed a significant antimicrobial activity against Staphylococcus aureus and S. epidermidis, respectively. Almost all extracts had moderate effect against Candida species. The highest total phenolic and flavonoid contents were determined in ISEA with 574.8 mg GAE (gallic acid equivalent) g -1 extract and 30.48 mg QE (quercetin equivalent) g -1 extract, respectively. These results showed that ISEA had a good antioxidant and anti-inflammatory activity with moderate α-amylase and butyrylcholinesterase inhibitory activity. (MC) extract on high fat-diet (HFD) induced heart and aorta damage by evaluating oxidative stress and the endothelial nitric oxide system (eNOS). Materials and Methods: Wistar albino male rats were divided into 3 groups (n=7) as control, HFD and HFD+MC. Rats in HFD and HFD+MC groups were HFD fed for 16 weeks and in the last 4 weeks saline or MC (100 mg/kg) was administered orally (5 days/week). Triglyceride, cholesterol and HDL were estimated in blood serum. Tissue oxidative stress and inflammatory parameters were evaluated biochemically. Tissues morphologies, eNOS, inducible NOS (iNOS) and NADPH oxidase-2 (NOX-2)-immunopositive and apoptotic cells were evaluated histologically. Results: Altered serum lipid profiles, degenerated heart and aorta morphology, increased malondialdehyde, 8‐hydroxy‐2‐deoxyguanosine, tumor necrosis factor-alpha, monocyte chemoattractant protein-1 and myeloperoxidase levels and iNOS, NOX-2 immunopositive and apoptotic cells, decreased NO levels, eNOS-immunopositive cells in both tissues were observed in HFD group. All these parameters improved in HFD+MC group. Conclusion: This study revealed that HFD-induced obesity increased iNOS activation and oxidative stress in both tissues. MC regulated oxidant/antioxidant had balanced thus preventing heart and aorta damage via eNOS involvement.Publication Open Access Myrtus communis L. Myrtus communis L. (MC) extract has been reported to have antioxidant and anti-inflammatory effects. The aim of this study was to investigate the protective effects of MC extract on high-fat diet (HFD)-induced kidney and bladder damage. Materials and Methods: Wistar albino male rats were divided into three experimental groups: control, HFD and HFD+MC. Experimental groups were fed a standard diet (control group) or HFD (HFD and HFD+MC groups) for 16 weeks. MC extract (100 mg/kg) was administered to the HFD+MC group orally during the last 4 weeks (5 days/week) of the experiment. Highdensity lipoprotein, total cholesterol, triglyceride and leptin levels were measured in blood serum. Tissue malondialdehyde (MDA), glutathione (GSH), 8-hydroxy-2'-deoxyguanosine (8-OHdG) and myeloperoxidase (MPO) levels were evaluated biochemically. Kidney and bladder morphology, NADPH oxidase-2 (NOX-2) and nuclear factor-kappa B (NF-ҡB)-positive and apoptotic cells were evaluated histologically. Results: Lipid profiles altered and leptin levels increased in blood serum. MDA, 8-OHdG and MPO levels increased and GSH level decreased in kidney and bladder in the HFD group. Moreover, degenerated kidney and bladder morphology, increased NOX-2 and NF-ҡB-positive and apoptotic cells were observed in this group. All of these biochemical and histological parameters were ameliorated in the HFD+MC group. Conclusion: HFD-induced obesity causes kidney and bladder damage by oxidative and inflammatory processes. MC extract may reduce oxidative stress and inflammation and play a protective role in obesity-related kidney and bladder damage.Publication Metadata only Determination of anti-cancer properties of urtica dioica ethanol extract on hematopoietic cell lines(2022-09-01) MEGA TİBER, PINAR; ŞEN, ALİ; ORUN, OYA; Mega Tiber P., Karadeniz M., Sevinç S. K. , Şen A., Orun O.Aim: Urtica dioica is a natural phytotherapeutic agent with various effects such as antioxidant, anti-inflammatory, anti-fungal, hypotensive as well as anti-carcinogenic. In recent years, phytotherapeutic approaches to reduce side effects of available chemotherapeutic agents or to find new agents with higher toxicity in cancer cells relative to healthy ones, are area of intense research. Therefore, in this study, we investigated anti-carcinogenic effects of U. dioica ethanol extract on K562 human chronic myeloid erythrocyte cell lines and on lymphocytes isolated from healthy donors (PBMC).Material and Methods: U. dioica extract was isolated by using soxhlet extraction methodology. Anti-carcinogenic effects of the agent were analyzed with MTT assay (Cell Proliferation Kit I, Roche), Annexin V/PI (Molecular Probes A10788), mitochondrial membrane potential assay (JC-1, Abnova KA1324) and cell cycle analyses (PI staining in flow cytometer (FACS Calibur). PBMCs were prepared from healthy volunteers through Ficoll Hypaque density gradients.Results: Data indicated that treatment with U. dioica didn’t show up a dose-dependent cytotoxic effect on K562 cell line, but there was a 20% suppression at 25 μg/ml compared to PBMC (p<0.001). U. dioica extract yielded to an increase in apoptosis 13% and 41% for 25 μg/ml and 100 μg/ml, respectively, in comparison to untreated control cells (p=0.033). It has also induced cell cycle arrest at G0/G1 phase, in a dose-dependent manner. Accumulation at G0/G1 phase increased 9.6, 10 and 14% in 50, 75 and 100 μg/ml concentrations, respectively, compared to the control group (p<0.0001).Conclusion: Our findings explicitly show that the total ethanol extract of U. dioica is highly effective in K562 cells on the induction of apoptosis and cell cycle arrest at reasonably low concentrations and these effects significantly pronounced compared to healthy controls.Publication Open Access Myrtle improves renovascular hypertension-induced oxidative damage in heart, kidney, and aortic tissue(2022-07-01) ŞEN, ALİ; ŞENER, GÖKSEL; AKBAY, TUĞBA; Arslan S., Ozcan O., Gurel-Gokmen B., ÇEVİKELLİ YAKUT Z. This study aims to investigate the possible effects of myrtle leaf extract (Myrtus communis L.) on, heart, kidney and aorta tissues in the experimental renovascular hypertension (RVH). 32 male Wistar Albino rats were divided into four groups as control, hypertension, hypertension+ramipril, and hypertension+myrtle leaf extract treatment groups. RVH model was induced by Goldblatt\"s 2-kidney 1-clip method. 12 weeks later than the treatment blood pressures were recorded and oxidant and antioxidant parameters, tissue factor activity, and histological analysis were determined in the kidney, heart, and aortic tissues. The blood pressure levels of the hypertension group significantly increased compared to the control group. Administration of myrtle leaf extract and ramipril significantly decreased the increased blood pressure. In the hypertension group, oxidative damage increased in the kidney, heart, and aorta tissues. In the histological evaluation of tissues in RVH, heart muscle fibres degenerated. Bowman capsule and glomeruli dilated and tubules damaged in the kidney. Myrtle leaf extract administration regenerated the damages and degenerations. The administration of myrtle leaf extract restored the impaired oxidant-antioxidant balance in the heart, kidney and aorta tissues of hypertensive rats. Myrtle leaf extract can be considered as an alternative antihypertensive treatment target in the prevention of oxidative stress-induced damage in renovascular hypertension.Publication Open Access Protective effects of Rubus tereticaulis leaves ethanol extract on rats with ulcerative colitis and bio-guided isolation of its active compounds: A combined in silico, in vitro and in vivo study(2022-11-01) ŞEN, ALİ; ÖZBEYLİ, DİLEK; ERTAŞ, BÜŞRA; DOĞAN, AHMET; BİTİŞ, LEYLA; Şen A., Özbeyli D., Teralı K., Göger F., Yıldırım A., Ertaş B., Doğan A., Bitiş L., Şener G.The aim of this study was to evaluate the therapeutic effect of active ethanol extract obtained from the leaves of Rubus tereticaulis (RTME) against colitis, and to purify major compounds from this extract by bioassay-directed isolation. Rats with colitis induced via intra-rectal acetic acid administration (5%, v/v) received RTME or sulfasalazine for three consecutive days. On day four, all rats were decapitated, and the colonic tissue samples were collected for macroscopic score, colon weight, reduced glutathione (GSH), myeloperoxidase (MPO), and malondialdehyde (MDA) analyses. The active compounds and chemical composition of RTME were determined by bio-guided isolation and LC-MS/MS, respectively. Compared to the colitis group, the rats treated with RTME displayed significantly lowered macroscopic scores and colon wet weights (p < 0.001). These effects were confirmed biochemically by a decrease in colonic MPO activity (p < 0.001), MDA levels (p < 0.001), and an increase in GSH levels (p < 0.001). Kaempferol-3-O-β-d-glucuronide (RT1) and quercetin-3-O-β-d-glucuronide (RT2) were found to be the major compounds of RTME, as evidenced by in vitro anti-inflammatory and antioxidant activity-guided isolation. Their anti-inflammatory/antioxidant activities were also predicted by docking simulations. Additionally, quinic acid, 5-caffeoylquinic acid, quercetin pentoside, quercetin glucoside, quercetin-3-O-β-d-glucuronide, kaempferol-3-O-β-d-glucuronide, and kaempferol rutinoside were identified in RTME via using LC-MS/MS. RT2, along with other compounds, may be responsible for the observed protective action of RTME against colitis. This study represents the first report on the beneficial effects of RTME in an experimental model of colitis and highlights the potential future use of RTME as a natural alternative to alleviate colitis.Publication Open Access The effect of Cotinus coggygria L. ethanol extract in the treatment of burn wounds(2022-01-01) OKUYAN, BETÜL; ŞEN, ALİ; ŞENER, GÖKSEL; ERCAN, FERİHA; Erta B., OKUYAN B., ŞEN A., ERCAN F., Onel H., GÖGER F., Sener G.The overall aim of the present research is to evaluate for the first time the curative effect of Cotinus coggygria leaves on burn injury in an experimental burn model along with its anti-inflammatory and antioxidant activity potential. Also, phenolic compounds of C. coggygria were characterised by LC-MS/MS. Wistar albino rats weighing 200-250 g were exposed to 90 degrees C bath for 10 s to induce burn injury, involving 30% of the total body surface area. In the treatment groups, 5% C. coggygria ethanol extract was applied topically as a cream immediately after the burn. Blood and skin tissue samples were taken after decapitation at the 4th and 48th hours following the burn procedure. Interleukin 1-beta (IL-1 beta) and tumour necrosis factor (TNF-alpha) were determined in serum samples, and hydroxyproline, prostoglandin E2 (PGE2), and myeloperoxidase (MPO) activity and 8-hydroxy-2\"-deoxy-guanosine (8-OHdG) levels were determined in skin tissue samples. Increased levels of serum cytokines were decreased with C. coggygria treatment in both periods. MPO activity, prostaglandine (PGE2), and 8-OhdG levels increased, while hydroxyproline levels decreased due to burn damage. On the other hand, these parameters were returned to its normal levels with C. coggygria treatment. In addition, the tissue histology of animals treated with C. coggygria showed a complete epithelialization with increased collagenation. As a result, C. coggygria may be an alternative treatment approach for burns-induced skin damage and wounds.Publication Open Access Therapeutic effects of momordica charantia L. (MoC) ethanolic extract on ulcerative colitis (UC) and was explored in vitro and in vivo. Materials and Methods: The rats were divided into control (C), saline-treated colitis (AA), MoC extract-treated colitis (AA+MoC), and sulfasalazine (SS)-treated colitis (AA+SS) groups. Colitis was induced by acetic acid. MoC extract, SS or saline were given to the related groups for 3 days. Interleukine-1β, malondialdehyde, glutathione levels, myeloperoxidase activity, bax/bcl-2 ratio, caspase-9 and caspase-3 levels were measured in colon and macroscopic and histopathologic examinations were done. Total phenolic/flavonoid content and biological activity of MoC were evaluated by in vitro analysis. Results: Compared to the control group, with acetic acid application interleukin-1β levels, myeloperoxidase activity, malondialdehyde levels, bax/bcl-2 ratio, caspase-9 and caspase-3 levels were significantly upregulated, while glutathione levels were significantly decreased in the AA group. In contrast, MoC and SS treatments reduced interleukin-1β, malondialdehyde levels, myeloperoxidase activity, bax/bcl-2 ratio, and caspase-9 and caspase-3 levels. Glutathione levels increased upon MoC or SS treatment. Increased macroscopic and microscopic scoring with AA improved with MoC or SS treatment, but the MoC or SS treated groups had higher score values than the control. Also, in vitro results showed that MoC exhibited 2,2-diphenyl-1- picrylhydrazyl and 2,2\"-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid radical scavenging activity as well as significant antilipoxygenase activity. In addition, MoC extract showed a potent anti-inflammatory activity compared to standard indomethacin. Conclusion: Our biochemical, in vitro and histopathologic analysis indicate that MoC is likely to prove beneficial in UC therapy.Publication Open Access Cotinus coggygria scop. The objective of the present study is to elucidate for the frst time the efects of Cotinus coggygria (CC) ethanol extract (CCE) on colonic structure and infammation of acetic acid-induced ulcerative colitis in rats. Colonic damage was assessed using disease activity index score, enzyme-linked immunosorbent assay, and hematoxylin–eosin staining. Also, in vitro antioxidant activity of CCE was investigated by ABTS methods. Total phytochemical content of CCE was measured spectroscopically. Acetic acid caused colonic damage according to disease activity index and macroscopic scoring. CCE signifcantly reversed these damages. While the levels of proinfammatory cytokines TNFalpha, IL-1beta, IL-6, and TGF-1beta increased in tissue with UC, IL-10 level decreased. CCE increased infammatory cytokine levels to values close to the sham group. At the same time, while markers indicating disease severity such as VEGF, COX-2, PGE2, and 8-OHdG indicated the disease in the colitis group, these values returned to normal with CCE. Histological research results support biochemical analysis. CCE exhibited signifcant antioxidant against ABTS radical. Also, CCE was found to have a high content of total polyphenolic compounds. These fndings provide evidence that CCE might be beneft as a promising novel therapy in the treatment of UC in humans due to high polyphenol content and justify the use of CC in folkloric medicine for treatment of infamed diseases.
